전면 성장한 BAEC 에 2 mM 티미딘(Thymidin, Sigma-Aldrich, USA)을 24 시간 동안 처리하여 세포의 성장을 중지시킨 후 PBS 으로 2회 세척하였다. 그 후 스크래퍼 찰과기(scraper)를 이용하여 창상선을 만든 후 다시 PBS 로 2회 세척한 뒤 다양한 농도의 라이코펜을 처리하여 24시간까지 시간 별로 확인하였다. 세포이동 정도는 일정한 시면적에 이동한 세포 수를 계산하여 평가하였다.

MAPK(Mitogen activated protein kinases)는 다양한 세포생리활성에 관여하는 세포신호전달물질이다. MAPK 중에서 가장 중요한 패밀리 구성원으로는 ERK(extracellular signal regulated kinase), JNK(c-Jun-N-terminal kinase), p38 MAPK 등이 있다. 한편, 활성화된 eNOS에 의해 만들어지는 산화질소(nitric oxide)는 혈관확장에 관여하는 등 혈관내피세포에서 중요한 생리적 기능에 관여하는 혈관 조절인자로 잘 알려져 있다. 따라서 라이코펜이 관여하는 세포생리활성과 세포신호전달물질의 관계를 조사하기 위한 알기 위한 실험을 진행하였다.
Mitogen activated protein kinases (MAPKs) are cell signaling agents involved in a variety of cell physiological activities. Among the most important family members of MAPK are extracellular signal regulated kinase (ERK), c-Jun-N-terminal kinase (JNK), and p38 MAPK. Meanwhile, nitric oxide produced by activated eNOS is well known as a vascular regulator that is involved in important physiological functions in vascular endothelial cells such as vascular expansion. Therefore, experiments were conducted to investigate the relationship between lycopene-associated cell physiological activity and cell signaling molecules.

전면 성장한 BAEC에 10 μM의 라이코펜을 0, 5, 15, 30, 60 분 동안 처리한 후 분해 완충액(50 mM Tris, 150 mM NaCl, 0.1 mM PMSF, 1.0 mM Na 바나데이트, 1% [v/v] Triton X-100, 10.0% 글리세롤, pH 7.4)으로 분해시킨 뒤 원심분리를 통해 상층액을 획득하여 단백질 분석으로 정량하고 동일한 양의 단백질을 SDS-PAGE를 이용하여 분리시켰다. 분리시킨 단백질은 PVDF (polyvinylidene fluoride) 막으로 이동시켜 고정하고, 분석하고자 하는 표적 단백질에 해당하는 1차 항체와 반응 후, 2차 항체를 반응시켜 화학발광 검출법을 통해 X-ray 필름으로 현상하여 세포 신호 전달 물질의 변화를 관찰하였다.

또한, 단백질 인산화 효소 및 산화질소 생산효소 억제제를 처리하여 세포성장, 세포이동에 어떠한 영향을 미치는 지 알아보기 위하여 혈청기아 상태인 DMEM 에 BAEC 을 16시간 동안 배양한 후, p38 (SB-203580, 10 μM), JNK (SP-600125, 10 μM), ERK (PD-98059, 20 μM) 그리고 eNOS (L-NAME, 2.5 μM)의 인산화 억제제를 1 시간 동안 처리 한 후 라이코펜 10 μM을 처리하여 앞선 방법과 동일한 방법으로 확인하였다.
