Description

숙시닐화 아텔로 콜라겐 제조방법 {Method for preparing succinylated atelocollagens}Method for preparing succinylated atelocollagen {Method for preparing succinylated atelocollagens}

The present invention relates to a method for separating atelocollagen and high purity atelo collagen prepared by using the same. More specifically, atelocollagen is economically separating high purity atelocollagen from animal tissues by removing impurities efficiently and conveniently. It relates to a separation method and high purity atelo collagen prepared using the same.

In addition, the present invention relates to a method for producing modified atelocollagen, which is dissolved in a neutral solution and has excellent biocompatibility, and thus can be applied to various formulations, and a modified atelocollagen prepared using the same.

In addition, the present invention relates to a collagen-based matrix having a three-dimensional matrix structure and a method for producing the same by using the atelocollagen and the modified atelocollagen as described above to improve the mechanical strength by making a hard layer and a porous layer and crosslinking them. It is about.

To regenerate tissue capable of bioengineering tissues, it is necessary to provide an environment in which various cells can be differentiated and proliferated. A crucial role for maintaining tissue morphology and ensuring cell growth is extracellular matrix (Extra-). Cellular Matrix).

In order to artificially make tissues, a technology that basically provides an environment in which cells grow and proliferates cells is required. In general, in the field of artificial tissue engineering, various growth factors are used for cell proliferation while providing a growth environment of cells using extracellular matrix such as collagen or proteoglycan.

Representatives of natural polymers used as biomaterials for making artificial tissues include collagen, fibronectin, vitronectin, and laminin among extracellular substrates. For example, collagen or fibrinocytes contain a peptide (so-called "RGD peptide") sequence consisting of arginine-glycine-aspartic acid that induces cell adhesion. Artificially arranging the RGD peptide on the surface of the biomaterial provides an environment that induces adhesion between the surface of the biomaterial and the cell, thereby mimicking the function of natural tissue by combining the surface of the biomaterial with surrounding cells.

In particular, in the field of applying protein as a biomaterial, it can be said that collagen is one of the very important natural materials among the various natural polymers described above. This is because collagen, which is distributed in almost all tissues in vivo, is an essential protein for building a living body by forming and maintaining organs and tissues in combination with cells as a structure for supporting and proliferating cells.

Collagen is a major protein component of the extra-cellular matrix, and it is contained in soft tissues such as bones and teeth as well as soft tissues including connective tissues such as skin, tendons, and blood vessels. It takes about one third of the protein, has a certain order, and the cells gather to form the basic structure of tissues or organs. Without collagen, multicellular animals cannot exist. Therefore, when trying to regenerate a damaged part of a living body to its original tissue, if the extracellular matrix of the tissue is supplied to the damaged part, the tissue cell will have regeneration ability, thus providing collagen as a basic matrix of the artificial tissue substitute. It is very advantageous.

On the other hand, there are many tissues containing collagen such as skin, ligaments, bones, blood vessels, amniotic membranes, pericardium, heart valves, placenta, and corneas, but the types of collagen are different for each tissue. In particular, type 1 collagen is most widely used in tissue engineering because it is contained in almost all tissues such as skin, ligaments and bones.

In addition, there is a part called telopeptide that does not form a helix at both ends of the collagen type 1 molecule, which is the main cause of the immune response. Atelocollagen is used.

The type 1 collagen separation method that is generally used up to now is treated with trypsin to separate animal cells from cells, remove minerals and various glycoproteins from the extracellular matrix, and utilize the intrinsic polarity or acid solubility of collagen. To remove acid-insoluble collagen. In order to separate type 1 collagen in this manner, a precipitation method, a chromatography method using a urea buffer solution, and a centrifugation method are conventionally used. As described above, pepsin is treated to remove telopeptides that cause an immune reaction. have.

However, since the conventional type 1 collagen separation method has to go through a multi-step process, the separation process is inconvenient, time-consuming and expensive, as well as the urea used to separate type 1 collagen. In addition, there is a problem in that separation and inactivation of pepsin used for telopeptide removal are necessary. That is, the conventional type 1 collagen separation method makes the separation process more complicated and difficult, and makes it difficult to economically obtain high-purity type 1 collagen.

In addition, despite the excellent wound healing effect by using collagen, biomaterials using collagen still have limitations for use in human tissues due to their low tensile strength and biodegradable properties.

Due to the characteristics of collagen, research on artificial tissues using collagen has been actively conducted, and a method of extracting collagen from animal tissues is also being studied.

In this regard, the Republic of Korea Patent No. 10-0465015 is a high purity type 1 collagen by removing the non-collagen substances through the enzyme treatment to reduce the immunity and extract and remove fatty impurities and insoluble collagen using an organic solvent The manufacturing method is described. However, when the organic solvent is used as in the Republic of Korea Patent No. 10-0465015, there may be a degeneration of collagen and the use of an organic solvent that is harmful to the human body may cause side effects when the separated collagen is applied in vivo. .

In addition, the Republic of Korea Patent No. 10-0676285 in the bone tissue, cartilage tissue, skin tissue, tendon / tendon tissue of the pig powdered or fragmented and acid treatment and then repeatedly treated pepsin to isolate the type 1 collagen primary In order to remove impurities, a method of separating the precipitated collagen by centrifugation after neutralization at a low temperature of 30 to 37 ° C. after repeating the three steps of salt treatment and neutral titration is performed. However, the separation method of the Republic of Korea Patent No. 10-0676285 may be advantageous in terms of removing fat or insoluble substances while preventing the denaturation of collagen, but the salt treatment process and the titration process to neutral are complicated and multiply first. There was a problem that is inadequate to economically separate the type collagen yield.

Because of these problems, it is very complicated and difficult to separate high-purity type 1 collagen from animal tissues while completely eliminating fats, impurity proteins, various insoluble impurities, urea, pepsin or other enzymes, salts, etc. Is considered. On the other hand, Korean Patent Laid-Open Publication No. 10-2002-0029859 discloses a method for producing atelocollagen, which is treated by enzymatic treatment and then purified by a conventional method immediately after pulverizing soft tissues or hard tissues of mammals in an acid solution. In addition, this technique also has the advantage of simplifying the process of the previous stage of the separation and purification process, but the method of increasing the purity by using dialysis and filtration in the subsequent separation and purification process.

In this regard, the extraction process of collagen has been performed using dialysis, multiple filtration, or various pore sizes in the process of increasing the purity by removing various impurities from the collagen separated from animal tissues. I was using a method of overlapping three or four filters. However, the dialysis method was limited in increasing purity due to the limited membrane pore size of 12,000 ~ 14,000 Daltons, and it was not possible to reuse multiple filters with different pore size. The use of expensive filters has the disadvantage of high cost.

Therefore, in the technical field of the present invention, it can be said that there is a need to provide an atelocollagen separation method for efficiently and simply removing impurities to economically separate high-purity atelocollagen from animal tissue.

In order to solve the problems of the prior arts described above, the present inventors provide a separation method that fuses an ultrafiltration and diafiltration method using a reusable filter to provide high purity even through a single process line. A method of extracting collagen was developed.

In addition, the present inventors have developed a method for producing modified atelocollagen, which is dissolved in a neutral solution and has excellent biocompatibility and can be applied to various formulations. Without collagen, it can be applied to various fields such as cosmetics, medicine, and food. In addition, the present inventors have introduced a porous matrix manufacturing technique in which a collagen is made of a two-layer structure (two collagen layers having different porosities) of a hard layer and a porous layer and crosslinked into the above-described technique to improve mechanical strength. The fabrication of the dimensional matrix structure solved the low tensile strength and biodegradation problem of collagen.

(2005.01.13.)Republic of Korea Patent No. 10-0465015 (January 13, 2005) 대한민국 등록특허 제10-0676285호 (2007.01.30.)Republic of Korea Patent No. 10-0676285 (2007.01.30.) (2002.04.20.)Republic of Korea Patent Publication No. 10-2002-0029859 (2002.04.20.)

An object of the present invention is to provide an atelocollagen separation method for efficiently and conveniently removing impurities and economically separating high-purity atelocollagen from animal tissues, and to provide high-purity atelocollagen prepared using the same.

In addition, the present invention is a modified atel collagen production method and modified atel prepared using the improved ease of use that can be applied to a variety of formulations by modifying the water-soluble collagen dissolved in a neutral solution to improve biocompatibility Its purpose is to provide collagen.

In addition, the present invention is a three-dimensional that improves the mechanical strength by making a hard layer and a porous layer (two collagen layers with different porosities) and crosslinking them using atelocollagen and modified atelocollagen as described above. It is an object of the present invention to provide a collagen-based matrix having a matrix structure and a method of preparing the matrix.

First, the terms used in the specification of the present invention will be described.

The term "ultrafiltration" used in the specification of the present invention is a filtration method located in the middle of microfiltration and reverse osmosis, and removes low-molecular substances from the polymer solution, but not a difference in concentration across the filtration membrane. By means of separating and filtering the material using. Filtration membranes used for ultrafiltration have pores of various sizes depending on the material to be separated.

In addition, the term diafiltration used in the specification of the present invention means that in the ultrafiltration, materials larger than the pores of the filtration membrane are concentrated in the relation of the feed, the residue, and the filtrate. Some of the desired substances to be filtered remain in the residue to be concentrated. The ratio of the desired substances to be gradually refined by returning them back to the feed side and adding purified water, etc., and concentrating them again through the filtration membrane. It means how to increase.

Hereinafter, the configuration of the present invention using the terms used in the specification of the present invention as described above.

② The solution obtained through the above step ① (hereinafter referred to as "refined collagen solution") is separated and purified through a single process line using a separation method that combines ultrafiltration and diafiltration. For this purpose, a separation and purification apparatus 100 as shown in FIG. 1 was prepared. The device of FIG. 1 may be manufactured directly or may be configured using a device (model name: CENTRASETTE System) sold by Pall corporation.

(3) Assemble a filtration membrane (not shown) in the filtration module 30 of the separation and purification apparatus 100 of FIG. 1, and wash NaOH remaining in the filtration membrane using purified water.

④ While checking the feed pressure gauge 32 and the residue pressure gauge 34 provided in the filtration module 30 of the separation and purification apparatus 100 shown in FIG. 1, the feed portion 33 into which the "collagen solution to be purified" is introduced. ) And the pressure of the remaining portion 35 from which the "retentate" remaining in the filtration module 30 and returned to the feed tank 10 flows out, and the front part of the filtration membrane is purified with purified water. Wash.

⑤ Applying a pressure of 10 ~ 30psi to the remaining portion 35 of the filtration module 30 by adjusting the pressure control valve 40 of the separation and purification device 100 shown in Figure 1, freely from the feed portion 33 By allowing the incoming purified water to be pressurized, the purified water also washes the back of the filtration membrane. After the cleaning is completed, the pressure of the remaining part 35 is released.

⑥ "정제대상이 되는 콜라겐 용액"을 수용하기 위한 용기와 분리정제된 아텔로 콜라겐 용액을 수용하는 용기를 준비하고 이들 용기들을 도 1의 분리 정제 장치(100)에 호스를 통해 연결한다.⑥ Prepare a container for accommodating the "refined collagen solution" and a container containing the purified purified atelo collagen solution, and connect these containers to the separation and purification apparatus 100 of FIG. 1 via a hose.

⑦ by adjusting the pressure control valve 40 of the separation and purification device 100 of Figure 1 apply a pressure of 10 ~ 30psi to the remaining portion 35 of the filtration module (30). This pressurizes the "refined collagen solution" which flows into the filtration module 30 from the feed tank 10 through the feed part 33 by the pumping action by the feed pump 20, and atelo collagen is pressurized. It is filtered through a filtration membrane. As a result, ultrafiltration of the "collagen solution to be purified" is primarily performed, and the purified atelocollagen is released from the filtration module 30 and received in a container prepared in advance.

⑧ Then, the filtration stops if the rate of the filtrate solution is reduced to approximately 1 g / min or less during the ultrafiltration of step ⑦.

⑨ And the yield and purity of atelo collagen by a single process using the separation and purification apparatus 100 shown in Figure 1 by fusing an ultrafiltration process and a diafiltration process for the convenience and efficiency of the separation and purification of atelo collagen Carry out a separation purification to raise. That is, in the separation and purification apparatus 100 of FIG. 1, the residue which is not filtered through the filtration membrane in the filtration module 30 is transferred to the feed tank 10 at a constant flow rate through the remaining portion 35 of the filtration module 30. If the ultrafiltration rate decreases below a certain speed in step ⑧, the ultrafiltration is stopped and the diafiltration process for the residue returned to the feed tank 10 is subsequently performed. At this time, the same amount of purified water as the ultrafiltered solution is added to the residue returned to the feed tank 10, and diafiltration is performed five times or more using the separation and purification apparatus 100 shown in FIG.

아 the atelo collagen solution obtained by ultrafiltration through step ⑦ and the atelo collagen solution obtained by diafiltration through step ⑨ are adjusted to pH 7.0, and then lyophilized to form a sponge for atelo collagen. Finally get

Using the atelocollagen separation and purification method of this embodiment can solve the problem of expensive purification and inconvenient purification by consuming a large number of expensive filters when multiple filters are overlapped. That is, the ultrafiltration using the reusable filter first purified some of the atelo collagen and based on the filtration rate, the efficiency of the ultrafiltration is judged to be lowered. It can be used as diafiltration device for. In other words, the atelocollagen separation and purification method of the present embodiment is efficient and simple to remove impurities and economically removes atelocollagen from animal tissues through a single cyclic process line using the separation and purification apparatus 100 as shown in FIG. 1. It can be separated and extracted with high purity with good yield.

In addition, pepsin was quantified to confirm whether or not pepsin used for telopeptide remained in the extracted and purified atelocollagen in Example 1. As a result, as shown in Figures 6a and 6b it was confirmed that pepsin is not detected in the atelo collagen separated and purified through the atelocollagen separation method of an embodiment of the present invention.

Therefore, it can be seen that the atelocollagen separation method of the present invention is an effective method for separation and purification with high purity while maintaining the atelocollagen from the animal tissues in an undenatured state.

First, real-time PCR was performed to determine whether HEV (Hepatitis E virus), a pig-derived virus, was detected in atelocollagen extracted and purified in Example 1. Example 3-1: Method for preparing succinylated atelo collagen

Method for producing succinylated atelo collagen of an embodiment of the present invention is as follows.

① Add 0.002 to 0.01 wt% of atelo collagen (both purified and commercially available in Example 1) in 0.1 M acetic acid solution and stirred at 4 ° C. for 1-2 days to atelo collagen. Dissolve.

② 상기 ①번 과정에서 얻은 아텔로 콜라겐 용액에, 상기 ① 과정에서 첨가한 아텔로 콜라겐 1g당 0.8~1.3g 정도의 비율로 숙신산 무수물(succinic anhydride)을 넣고, 10분 동안 0.05~1M NaOH를 이용하여 pH를 9~10근처로 유지시킨다.② In the atelo collagen solution obtained in step ①, succinic anhydride is added at a rate of about 0.8 to 1.3 g per 1 g of atelocollagen added in step ①, and 0.05 to 1 M NaOH is used for 10 minutes. To keep the pH near 9-10.

The solution obtained in step ② is stirred at 4 ° C. for 30 minutes.

Stir the solution obtained in step ⑥ at 4 ° C. for 20 minutes.

⑧ 상기 ⑦번 과정에서 교반한 용액을 0.05~1M NaOH를 이용하여 10분 동안 pH를 9~10근처로 유지시킨다.⑧ The solution stirred in step ⑦ is maintained at around 9 ~ 10 for 10 minutes using 0.05 ~ 1M NaOH.

The solution obtained in step VII was adjusted to pH 4.03 with 3-7M HCl to form succinylation atelocollagen sedimentation and stirred at 4 ° C. for 15 minutes.

(3) Centrifuging the solution stirred in step (b) to obtain succinylation atelocollagen sedimentation.

아 distilled water adjusted to pH 4.03 using 3-7 M HCl at a rate of about 20 mL per 1 g of atelo collagen added in step ① above to atelo collagen sedimentation obtained in step, above, Wash by stirring for minutes.

The succinylated atelocollagen saline obtained by repeating the above-described steps 과 and ⑭ once more is lyophilized at −70 ° C. for 30 hours to finally obtain succinylation atelo collagen.

A reaction scheme showing a reaction in which the atelo collagen is succinylated through the manufacturing process as described above is as follows.

Figure 112011091282840-pat00001

Figure 112011091282840-pat00001

On the other hand, when manufacturing succinylation collagen using the existing method there is a problem that does not dissolve even if the pH of the succinic anhydride is too low or too high. Succinic anhydride dissolves best when the pH is about 9 ~ 10 and does not dissolve when the pH is above 11 In view of these problems, the present inventors have identified a problem that the solubility of the succinic anhydride is decreased when the pH is changed according to the reaction of atelo collagen and succinic anhydride, so that the reaction rate is lowered and the yield is lowered. The process of repeating the pH of 9 to 10 was newly introduced.

That is, in the method for preparing succinylation atelocollagen according to one embodiment of the present invention as described above, after stirring the reaction product of atelocollagen and succinic anhydride for a predetermined time at low temperature, the stirred solution is maintained at pH 9 to 10 for a predetermined time. This facilitates dissolution of succinic anhydride, thereby promoting the succinylation reaction. Thus, according to the succinylated atelo collagen manufacturing method of an embodiment of the present invention, it is possible to improve the yield of succinylated atelo collagen.

Example 3-2 Preparation of Esterified Atelo Collagen

The preparation method of esterified atelo collagen of one embodiment of the present invention is as follows.

① 0.5 to 1 M acetic acid in a dispersion containing 1 to 5% by weight of atelo collagen (both purified and commercially available in Example 1) in 70 to 90% ethanol (or methanol). acetic acid) or 0.1 ~ 0.5M HCl is adjusted to pH 2-4 and stirred at 4 ℃ for 4-10 days.

② Adjust the pH of the atelo collagen dispersion obtained in step ① to 7.4 using 0.1 ~ 0.5M NaOH and then centrifuge to obtain only acupuncture.

③ The acupuncture obtained in step ② is stirred in purified water at a rate of about 10-100 mL per 1 g, and then put in a dialysis membrane to perform dialysis in purified water.

④ After stirring for 16 ~ 24 hours, replace the purified water (dialysis buffer), and then replace the purified water (dialysis buffer) 3 to 12 times every 3 to 5 hours.

⑤ 상기 ③번 과정 및 ④번 과정을 통해 투석된 에스테르화 아텔로 콜라겐 침사를 -70℃에서 30시간 이상 동결건조하고, 동결건조된 에스테르화 아텔로 콜라겐을 얻는다.⑤ Freeze-dried esterified atelocollagen sedimentation of the dialysis esterification atelocollagen through the process of step ③ and step ④ for more than 30 hours, to obtain lyophilized esterified atelo collagen.

A reaction scheme showing a reaction in which the atelo collagen is esterified through the manufacturing process as described above is as follows.

Figure 112011091282840-pat00002

On the other hand, in the conventional method for producing esterified atelo collagen, the dialysis using only purified water is performed to improve the yield and purity, whereas in the method for producing the esterified atelo collagen according to one embodiment of the present invention, ethanol or methanol The atelo collagen dispersion containing the atelo collagen was neutralized, centrifuged to obtain only sedimentation, and then dialysis was performed using a dialysis membrane to improve purity and yield.

Example 4 Evaluation of Physical Properties of Modified Atelo Collagen

In Example 3, the solubility of succinylated atelo collagen (anionized atelo collagen) obtained using the modified atelocollagen production method of one embodiment of the present invention was compared with conventional atelo collagen. As can be seen in the photograph of Figure 9, it can be seen that the succinylated atelo collagen prepared using the modified atelocollagen production method of the present invention shows high solubility at neutral pH (pH 6.0 ~ 7.0).

Thus, the "succinylated atelo collagen in which succinic anhydride is bound to atelo collagen" obtained using the modified atelocollagen production method of one embodiment of the present invention has anionic properties and is lysed at neutral pH. Can improve adhesion, proliferation, and migration. In addition, "esterified atelo collagen in which ethanol (or methanol) is bound to atelo collagen" obtained using the modified atelocollagen production method of one embodiment of the present invention is also dissolved at neutral pH and has a cationic property. It has the advantage that it can be combined more quickly and easily with the cell.

Therefore, in the present invention, the high-purity atelo collagen obtained through ultrafiltration and diafiltration is succinylated and esterified to be solubilized in neutral pH solution so that collagen is not hydrolyzed by peptide or enzymatic treatment of collagen. As it is, there are advantages that can be applied to various fields such as cosmetics, medicine, and food.
