본 실험예에서는 실시예 1 ~ 8에서 제조한 기능성 식품에 대한 주요 기능성을 동물실험으로 평가하였다. 기능성 평가로 6주령의 S·D계 웅성 랫트(160g)를 구입하여 1주일간 고형 사료(삼양 사료)로 적응시켰다. 사육실은 온도 20℃±5, 습도 60%를 유지하며 12시간 간격으로 점등 및 소등하였다. 실험동물의 구분은 대조군과 실험군으로 나누었으며 대조군은 담배연기의 흡입과 세균감염을 시키지 않은 건강한 상태의 랫트로 하였다. 실험군의 랫트는 밀폐된 상자(L90 × W45 × H45 ㎝)내에 넣어 1일간 담배연기를 흡입시킨 다음 기관지 조직에 폐렴 세균을 주입시켜 기관지 질환을 유도한 후 1일간 반응을 진행시켰다. 기능성 평가의 기준은 병리학적 소견으로 해석하였다. 기관지 조직에 질환이 유발되어 염증이 생기게 되면 건강한 상태(5 × 103∼ 2.5 × 104)보다 백혈구 수가 증가되는 백혈구과다증(2.5 × 104이상) 증상이 나타나며 이 증상의 치료효과로 본 발명의 기능성 평가를 하였다. 기관지 질환의 유발 유무를 확인하기 위해 치료직전에 건강한 대조군의 랫트와 실험군의 랫트로부터 혈액을 채취하여 백혈구 수를 측정하였다. 기관지 질환이 유도된 실험군은 체중이 비슷한 랫트를 무작위로 분류하여 대조군과 실험군으로 나누었으며 대조군에는 실험군과 동일한 스트레스를 주기 위해 체중 100g당 0.5 mL(60㎏당300 mL 기준)의 증류수를 경구투여하였으며 실험군에는 체중 100g당 0.5 mL(60㎏당 300 mL 기준)의 도라지 추출물인 시료를 5시간마다 경구투여하였다. 백혈구 수의 측정은 5시간 간격으로 3일동안 랫트의 꼬리정맥으로부터 혈액을 채취하여 Turk식 용액인 0.3% 초산에 메틸렌블루가 섞인 염색액으로 희석한 다음 헤모사이토메타 (hemacytometer : Sigma chemical Co.,)를 이용하여 1㎜3의 혈액속 백혈구 수를 계산하였다. 계산방법은 하기와 같다.In this experimental example, the main functionalities of the functional foods prepared in Examples 1 to 8 were evaluated by animal experiments.
