Abstract
(a)該中実繊維の横断面において、内接円の直径をDi、外接円の直径をDoとすると、異形度Do/Diが1.20〜8.50;
(b)繊維の細孔比表面積が30m2/g以上
上記多孔質繊維を繊維束として28vol%以上含んでなる吸着材料。
上記吸着材料がケース軸方向にストレート形状に配列され、プラスチックケーシングの両端に被処理液の入口ポート、出口ポートが取り付けられてなる浄化カラム。
被処理液中の除去対象物質の効率的な吸着が可能な多孔質繊維、および多孔質繊維を内蔵した浄化カラムを提供する。
Description
異形断面形状の中実繊維であって、次の(a)〜(b)を満たす多孔質繊維、である。
(a)該中実繊維の横断面において、内接円の直径をDi、外接円の直径をDoとすると、異形度Do/Diが1.20〜8.50;
(b)繊維の細孔比表面積が30m2/g以上。
上記多孔質繊維を繊維束として28vol%以上含んでなる吸着材料、である。
上記吸着材料がケース軸方向にストレート形状に配列され、プラスチックケーシングの両端に被処理液の入口ポート、出口ポートが取り付けられてなる浄化カラム、である。
本発明の多孔質繊維は、多孔質繊維中の孔の直径が25μm以下であり、繊維の中心部領域における平均孔径に対する繊維の外表面近傍領域における平均孔径の比率が、0.50倍以上3.00倍以下であることが好ましい。
(d)多孔部と緻密層が連続している;
(e)緻密層が多孔部よりも繊維の外表面近傍に存在し、繊維最外表面から多孔部までの距離T1が0.001μm以上、30μm以下である。
本発明の多孔質繊維は、中実繊維がストレート形状であることが好ましい。
まず、多孔質繊維を十分に湿らせた後に液体窒素に浸し、細孔内の水分を液体窒素で瞬間的に凍結させる。その後、速やかに繊維を折り、繊維断面を露出させた状態で、0.1torr以下の真空乾燥機内で凍結させた水分を除去して乾燥試料を得る。その後、スパッタリングにより、白金(Pt)や白金−パラジウム(Pt−Pd)などの薄膜を試料表面に形成して、観察試料とする。該試料の断面を走査型電子顕微鏡((株)日立ハイテクノロジーズ製、S−5500)にて観察する。ここで、繊維断面の中心点8を通る半径を任意に選択し、図2のように、この半径の線分を均等な長さに5分割する点を通る同心円5(図2)を描き、中心点を含む領域を中心部領域6とし、最も外周部に近い側を外表面近傍領域7とする。中心部領域、外表面近傍領域、それぞれに存在する孔の円相当径を求め、それぞれの領域における平均孔径を得る。それぞれの領域における平均孔径の算出に際しては、走査型電子顕微鏡(5万倍)で2μm×2μmの範囲を任意に20箇所選択し、撮影した写真の中に孔全体が含まれるものについて測定し、平均の孔径を算出するものとする。孔径の測定においては、電子顕微鏡像を印刷したものの上に透明シートを重ね、黒いペンなどを用いて孔部分を黒く塗りつぶす。その後、透明シートを白紙にコピーすることにより、孔部分は黒、非孔部分は白と明確に区別し、画像解析ソフトにて孔径を求める。
Dxyの下限としては、好ましくは0.2以上、より好ましくは0.4以上、さらに好ましくは0.6以上である。Dxyの上限としては、好ましくは6.0以下、より好ましくは4.0以下、さらに好ましくは2.5以下となる。延伸開孔法などで作製した繊維は、繊維長手方向に特徴的な配向構造をもつため、一般的にDxyが非常に高い構造となるため、好ましいとはいえない。
孔径が10nm以上の孔を特定し、その孔が存在しない層を緻密層として、孔径10nm以上の孔から繊維表面までの最短距離を緻密層の厚みとする。すなわち、径が10nm以上の孔のうち、繊維表面に近い順に発見される5点をピックアップし、各々繊維表面に接する平面に対して垂線を引き、その垂線上における繊維表面と孔径10nm以上の孔との距離を各々求める。同様の測定を10枚の画像で行い、計50点の測定データの平均値について小数点第3位を四捨五入し、これを繊維の表面近傍緻密層の厚みとする。
また、表面緻密層厚みを縮小することで、繊維表面の開孔面積も増大できる。多孔質繊維の表面開孔率は0.5%以上であることが好ましく、より好ましくは1.5%以上、特に好ましくは2.0%以上である。開孔率が高くなると、処理液中の被吸着物質が繊維内部の吸着サイトに拡散しやすいため好ましい。一方で、上限としては、好ましくは30%以下であり、さらに好ましくは16%以下、特に好ましくは12%以下である。開孔率上限が上記好ましい範囲である場合には、繊維強度の低下や、表面粗さの増大を招くことはない。また、細孔内部で発生した微粒子などが繊維外部へ流出しやすくなることもない。
内接円占有度の下限としては、0.10以上であり、より好ましくは、0.20以上、さらに好ましくは0.30以上、特に好ましくは0.50以上となる。
T1/T2の上限としては0.030以下が好ましく、さらに好ましくは0.020以下、特に好ましくは0.010以下となる。T1/T2の上限が上記好ましい範囲であると、吸着サイトである多孔部が相対的に減少せず、吸着サイトが飽和しにくく、繊維の吸着孔率が低下するおそれはない。

本発明における多孔質繊維は、処理液の入口ポートと出口ポートをもつケーシングに内蔵することで浄化カラムとして用いることができる。ケーシングの形状としては、両端が開放端であり、例えば四角筒体、六角筒体等の角筒体や円筒体が挙げられ、中でも円筒体、特に断面が真円状の筒体が好ましい。これはケーシングが角をもたないことで、角部での血液の滞留を抑制できるためである。また、両側を開放端とすることで、処理液の流れが乱流になりにくく圧力損失を最小限に抑えることができる。また、ケーシングはプラスチックや金属等により構成される器具であることが好ましい。プラスチックの場合は、例えば機械的強度、熱安定性に優れる熱可塑性樹脂が用いられる。このような熱可塑性樹脂の具体例としては、ポリカーボネート系樹脂、セルロース系樹脂、ポリエステル系樹脂、ポリアリレート系樹脂、ポリイミド系樹脂、環ポリスルホン樹脂、ポリエーテルスルホン樹脂、ポリオレフィン系樹脂、ポリスチレン樹脂、ポリビニルアルコール樹脂、及びこれらの混合物が挙げられる。これらの中でもケーシングに求められる成形性、放射線耐性の点においてポリプロピレン、ポリスチレン、ポリカーボネートおよびそれらの誘導体が好ましい。特に、ポリスチレン、ポリカーボネートなどの透明性に優れた樹脂は、たとえば血液などの灌流時に内部の様子を確認できるため安全性の確保に好都合であり、放射線耐性に優れる樹脂は滅菌時に放射性照射する場合に好ましいためである。樹脂は、金型による射出成形や、素材を切削加工することにより製作される。中でもコストや成型性、重量、血液適合性などの観点からプラスチックが好適に用いられる。

なお、ケーシング胴部の断面積については、ケーシングにテーパーがある場合は、ケーシング中央における断面積とする。
[多孔質繊維の作製]
質量平均分子量が40万のシンジオタクティックPMMA(以下、syn−PMMA)を31.7質量部、質量平均分子量が140万のsyn−PMMAを31.7質量部、質量平均分子量が50万のアイソタクテイックPMMA(以下、iso−PMMA)を16.7質量部、パラスチレンスルホン酸ソーダを1.5mol%含む分子量30万のPMMA共重合体20質量部をジメチルスルホキシド376質量部と混合し、110℃で8時間撹拌し紡糸原液を調製した。得られた紡糸原液の92℃での粘度は1,880poiseであった。得られた紡糸原液を、92℃に保温された図6に示す形状であって表1に示す寸法の吐出口をもつ口金から、1.1g/minの速度で空気中に吐出し、空中部分を380mm走行させた後、凝固浴に導き、浴内を通過させて中実繊維を得た。凝固浴には水を用いており、水温(凝固浴温度)は43℃であった。それぞれの繊維を水洗後、保湿剤としてグリセリンを70重量%含む水溶液から成る浴槽に導いた後、温度を84℃とした熱処理浴内を通過させて余分のグリセリンを除去した後に16m/minで巻き取った。
図7に示す形状であり、表1に示す寸法の吐出口をもつ口金を用いた以外は、実施例1と同様の条件で繊維を作製した。結果を表1,2に示した。

図6に示す形状であり、表3に示す寸法の吐出口をもつ口金を用いた以外は、実施例1と同様の条件で繊維を作製した。結果を表3,4に示した。
図10に示す形状であり、表5に示す寸法の吐出口をもつ口金を用い、表5に示す凝固浴温度とした以外は実施例1と同様の条件で繊維を作製した。結果を表5,6に示した。
[カラムの作製]
実施例5で得た繊維を束ね、内径56mm、軸方向長さ58mmのポリカーボネート製円筒状ケーシング内に、繊維の充填率が53%となるようにストレート形状で内蔵した。次にこのカラムの両側端面の被処理液の流出入口に、ケーシング内径と同等の径にカットした目開き円相当直径84μm、開口率36%のポリプロピレン製メッシュフィルタを装着した。最後に、ケーシング端部には被処理液の流入口、流出口をもつヘッダーと呼ばれるキャップをとりつけた。
カラムの吸着性能評価として、β2−MGのクリアランスを測定した。β2−MGは、長期透析合併症である透析アミロイドーシスの原因タンパク質であることが知られている。
循環を1時間行った後ポンプを停止した。
I式において、CO=β2−MGクリアランス(mL/min)、CBi=Bi液におけるβ2−MG濃度、CBo=Bo液におけるβ2−MG濃度、QB=Biポンプ流量(mL/min)である。結果を表7に示した。
実施例11で得た繊維を用いた以外は、実施例19と同様の方法でカラム化し、カラムの吸着性能測定を行った。結果を表7に示した。
比較例1で得た繊維を用いた以外は、実施例19と同様の方法でカラム化し、カラムの吸着性能測定を行った。結果を表7に示した。
実施例19と同様の方法でカラムを作製した。カラムは、RO水10Lで洗浄した後、抗酸化剤としてエタノール1,000ppmを含む水溶液を充填し、線量25kGyのγ線を照射した。その後、カラムを解体して繊維を取り出し、付着血小板数を評価した。結果を表8に示した。
成人健常者の血液を採血後、直ちに抗凝固剤としてヘパリンナトリウム注射液(エイワイファーマ(株)製)を100U/mLになるように添加した。前記血液を、採血後30分以内に、カラムを解体して取り出した繊維に接触させ、37℃で2時間振盪させた。その後、繊維を生理食塩水で洗浄し、2.5容積%グルタルアルデヒド(ナカライテスク(株)製)生理食塩水溶液で血液成分の固定を行い、続いて蒸留水で洗浄した。電子顕微鏡用試料台に前記繊維を貼り付け、常温0.1torr以下にて10時間減圧乾燥した。その後、スパッタリングにより、白金/パラジウムの薄膜を中空糸膜表面に形成させて、試料とし、中空糸膜内表面をフィールドエミッション型走査型電子顕微鏡((株)日立ハイテクノロジーズ製S−800)にて、倍率1,500倍で試料の表面を観察し、1視野中(4.3×103μm2)の付着血小板数を数えた。これを繊維表面50箇所について測定し、その平均値を求めた。
実施例22
抗酸化剤としてエタノール500ppmを含む水溶液を充填した以外は、実施例21と同様の方法でカラムを得、付着血小板数を評価した。結果を表8に示した。
実施例23
抗酸化剤としてエタノール200ppmを含む水溶液を充填した以外は、実施例21と同様の方法でカラムを得、付着血小板数を評価した。結果を表8に示した。
抗酸化剤としてエタノール100ppmを含む水溶液を充填した以外は、実施例21と同様の方法でカラムを得、付着血小板数を評価した。結果を表8に示した。
抗酸化剤としてブタノール1,000ppmを含む水溶液を充填した以外は、実施例21と同様の方法でカラムを得、付着血小板数を評価した。結果を表8に示した。

Claims (20)
- 中実の異形断面形状であって、次の(a)〜(b)を満たす多孔質繊維。
(a)該中実繊維の横断面において、内接円の直径をDi、外接円の直径をDoとすると、異形度Do/Diが1.20〜8.50;
(b)繊維の細孔比表面積が3m2/g以上 - 平均細孔半径が0.8nm以上、90nm以下である請求項1記載の多孔質繊維。
- 細孔比表面積が30m2/g以上である請求項1または2記載の多孔質繊維。
- 次式で表される内接円占有度が、0.10以上である請求項1〜3のいずれか一項記載の多孔質繊維。
内接円占有度= 繊維横断面の内接円の面積/繊維横断面積 - 多孔質繊維中の孔の直径が25μm以下であり、繊維の中心部領域における平均孔径に対する繊維の外表面近傍領域における平均孔径の比率が、0.50倍以上3.00倍以下である請求項1〜4のいずれか一項記載の多孔質繊維。
- 連通孔からなる網状の構造を有する多孔部と、該多孔部に比べて緻密な構造をもつ緻密層とを有する繊維断面構造であり、次の(d)〜(e)を満たす請求項1〜5のいずれか一項記載の多孔質繊維。
(d)多孔部と緻密層が連続している;
(e)緻密層が多孔部よりも繊維の外表面近傍に存在し、繊維最外表面から多孔部までの距離T1が0.001μm以上、30μm以下である - 円相当直径(μm)をT2としたときに、T1、T2が次式を満たす請求項1〜6のいずれか一項記載の多孔質繊維。
T1/T2≦0.030 - 中実繊維がストレート形状である請求項1〜7記載のいずれか一項記載の多孔質繊維。
- 円相当直径T2が10μm以上、1,000μm以下である請求項1〜8のいずれかに記載の多孔質繊維。
- 表面の開孔率が0.5%以上、30%以下である請求項1〜9のいずれかに記載の多孔質繊維。
- 細孔径分布指数が1.0以上、2.8以下である請求項1〜10のいずれかに記載の多孔質繊維。
- 多孔部と緻密層の構成材料について、双方が、双方に共通の素材を45vol%以上含む請求項6〜11のいずれかに記載の多孔質繊維。
- 陰性荷電を有する請求項1〜12のいずれかに記載の多孔質繊維。
- 非晶性の高分子材料を含む請求項1〜13のいずれかに記載の多孔質繊維。
- 非晶性の高分子を含み、前記非晶性高分子にエステル基含有ポリマーが含まれる請求項1〜14のいずれかに記載の多孔質繊維。
- 表面にヒト血液を接触させたとき付着する血小板数が30個/(4.3×103μm2)以下である請求項1〜15のいずれかに記載の多孔質繊維。
- 請求項1〜16のいずれか一項記載の多孔質繊維を繊維束として28vol%以上含んでなる吸着材料。
- 医療用途である請求項17に記載の吸着材料。
- β2−マイクログロブリンの繊維体積あたりの吸着量が0.005mg/cm3以上である請求項18に記載の吸着材料。
- 請求項16〜19のいずれかに記載の吸着材料が、プラスチックケーシングの軸方向にストレート形状に配列され、前記プラスチックケーシングの両端に被処理液の入口ポート、および出口ポートが取り付けられてなる浄化カラム。
