Abstract
Description
図1は、本発明の磁性体粒子操作用デバイス(以下、単にデバイスともいう)の一形態を示す模式的斜視図であり、図2A〜図2Cは、図1に示すデバイスのII−II線断面図である。図2Dは、図2BのD−D線断面図である。
容器とは別に、ゲル状媒体および液体等が、独立に提供されてもよい。容器内へのゲル状媒体および液体の装填は、磁性体粒子の操作の直前に行われてもよく、磁性体粒子の操作前に十分な時間をおいて行われてもよい。ゲル状媒体が液体に不溶または難溶である場合には、装填後に長時間が経過しても、両者の間での反応や吸収はほとんど生じない。
上記のように、磁性体粒子を用いた操作では、液体中での磁性体粒子を分散と、他の液体中への磁性体粒子の移動とを繰り返し行うことにより、目的物質の分離、精製、反応、検出等が行われる。例えば、シリカコーティングが施された磁性体粒子を用いて核酸の分離・抽出を行う場合、核酸を含む試料中で磁性体粒子を分散させ、磁性体粒子の表面に核酸を固定した後、磁性体粒子を洗浄液中へ移動させる。洗浄液中で磁性体粒子を分散させ表面に付着した夾雑タンパク質等を除去した後、磁性体粒子を核酸溶出液中へ移動させる。磁性体粒子を核酸溶出液中へ移動させる。核酸抽出液中で磁性体粒子を分散させることにより、粒子表面に固定されていた核酸を溶出液中に回収することができる。

Claims (10)
- 液体およびゲル状媒体が装填された磁性体粒子操作用デバイスであって、
第1の液体が収容された第1液体収容部と、第2の液体が収容された第2液体収容部と、第3の液体が収容された第3液体収容部と、第1のゲル状媒体が収容された第1ゲル状媒体収容部とを備え、
前記第1液体収容部、前記第2液体収容部および前記第3液体収容部はそれぞれ前記第1ゲル状媒体収容部に接続されており、
前記第1のゲル状媒体によって、前記第1の液体、前記第2の液体および前記第3の液体が隔てられている、磁性体粒子操作用デバイス。 - 第4の液体が収容された第4液体収容部をさらに備え、
前記第4液体収容部は前記第1ゲル状媒体収容部に接続されている、請求項1に記載の磁性体粒子操作用デバイス。 - ゲル状媒体が収容されたゲル状媒体収容部として、前記第1ゲル状媒体収容部のみを備える、請求項1または2に記載の磁性体粒子操作用デバイス。
- 第4の液体が収容された第4液体収容部と、第2のゲル状媒体が収容された第2ゲル状媒体収容部とをさらに備え、
前記第3液体収容部および第4液体収容部はそれぞれ前記第2ゲル状媒体収容部に接続されており、
前記第2のゲル状媒体によって、前記第3の液体および前記第4の液体が隔てられている、請求項1に記載の磁性体粒子操作用デバイス。 - 前記第1液体収容部、前記第2液体収容部、前記第3液体収容部および前記第1ゲル状媒体収容部は、同一平面上に形成された外壁面を有する、請求項1〜4のいずれか1項に記載の磁性体粒子操作用デバイス。
- 前記デバイス内において移動されるべき磁性体粒子が前記デバイス内に装填されている、請求項1〜5のいずれか1項に記載の磁性体粒子操作用デバイス。
- 請求項1〜6のいずれか1項に記載の磁性体粒子操作用デバイスを作製するためのキットであって、
第1の液体を収容する第1液体収容部と、第2の液体を収容する第2液体収容部と、第3の液体を収容する第3液体収容部と、第1のゲル状媒体を収容する第1ゲル状媒体収容部とを備え、前記第1液体収容部、前記第2液体収容部および前記第3液体収容部がそれぞれ前記第1ゲル状媒体収容部に接続されている容器;および
前記第1ゲル状媒体収容部に収容されるべきゲル状媒体を含む、磁性体粒子操作デバイス作製用キット。 - 前記第1液体収容部、前記第2液体収容部および前記第3液体収容部にそれぞれ収容されるべき液体をさらに含む、請求項7に記載の磁性体粒子操作デバイス作製用キット。
- 液体、ゲル状媒体および磁性体粒子が装填されたデバイス内において、前記磁性体粒子を移動させるための磁性体粒子の操作方法であって、
前記デバイスは、第1の液体が収容された第1液体収容部と、第2の液体が収容された第2液体収容部と、第3の液体が収容された第3液体収容部と、第1のゲル状媒体が収容された第1ゲル状媒体収容部とを備え、
前記第1液体収容部、前記第2液体収容部および前記第3液体収容部はそれぞれ前記第1ゲル状媒体収容部に接続されており、
前記第1のゲル状媒体によって、前記第1の液体、前記第2の液体および前記第3の液体が隔てられており、
前記磁性体粒子の操作方法は、
磁場操作により、前記第1液体収容部内の磁性体粒子が、前記第1ゲル状媒体収容部へ移動させられるステップ;
磁場操作により、前記第1ゲル状媒体収容部内の磁性体粒子が、前記第2液体収容部へ移動させられるステップ;
磁場操作により、前記第2液体収容部内の磁性体粒子が、前記第1ゲル状媒体収容部へ移動させられるステップ;および
磁場操作により、前記第1ゲル状媒体収容部内の磁性体粒子が、前記第3液体収容部へ移動させられるステップを有する、磁性体粒子の操作方法。 - 前記第1液体収容部、前記第2液体収容部、前記第3液体収容部および前記第1ゲル状媒体収容部は、同一平面上に形成された外壁面を有し、
前記磁性体粒子は、前記外壁面に沿って移動させられる、請求項9に記載の磁性体粒子の操作方法。
