技術分野
本発明は、糖類を用いたエンドトキシンの除去技術に関し、詳しくは、糖類を含有するエンドトキシンの除去用組成物ならびに該組成物を用いたエンドトキシンの除去方法および抽出方法に関し、医療用具、医薬品製造器具、医薬品製造装置などの固体表面からエンドトキシンを除去するために有用な技術に関する。
背景技術
エンドトキシンは主としてグラム陰性菌の細胞壁の外膜に存在するリポ多糖の一種であり、発熱性物質(Pyrogen)として一般によく知られている物質である。
注射剤やカテーテルなどのエンドトキシン汚染により体内へ発熱性物質が混入すると、発熱、ショック、広汎性血管内凝固(DIC:Disseminated Intravascular Coagulation)などを発症する可能性があるので、注射剤等の医薬品、注射器やカテーテル等の医療用具、さらには医薬品製造器具及び装置等のエンドトキシン汚染は、厳しく管理する必要がある。
多くの医療用具や医薬品製造器具及び装置等は、種々の合成樹脂、ガラス、金属及び天然繊維等から構成されており、エンドトキシンの付着又は吸着により汚染される可能性が高い。
通常、エンドトキシンは250℃で30分以上または170℃で120分以上の加熱処理で不活性化できるので、ガラスや金属などは、このような加熱処理によりエンドトキシンを除染できる。また、エンドトキシンは水酸化ナトリウム含有エタノールまたはジメチルスルホキシドなどの有機溶媒中30℃にて不活性化するので、ガラスなどはこのような媒体で洗浄することによってもエンドトキシンを除去できる。しかし、このような加熱処理または有機溶媒処理が適用できない材質で構成された医療用具や医薬品製造器具及び装置等またはこのような加熱処理が不可能な構造の医療用具や医薬品製造器具及び装置等については、他の除染方法によらなければならない。
これらの医療用具や医薬品製造器具及び装置等に含まれるエンドトキシンの除去には、例えば、エンドトキシンを含まない水や生理食塩水で洗浄する方法が考えられるが、この洗浄方法では、水や生理食塩水を大量に必要とし、しかも医療器具や医薬品製造器具及び装置に付着又は吸着したエンドトキシンの除去が不完全である可能性が危惧されていた。
特許第2884975号公報には、容器、器具等の固体表面に付着または吸着されたエンドトキシンを効率よく抽出する方法として、水や生理食塩水などの代わりにアルブミン、ヒト血清アルブミン、グロブリンなどのタンパク溶液を用いて、容器、器具等を洗浄して溶液中にエンドトキシンを抽出する方法が記載されている。
しかし、血液タンパクは生体由来成分であるので、医療用具や医薬品製造器具及び装置等に存在するエンドトキシンを除去するための洗浄には望ましくない。このような用途には、上記のような生体由来成分を含む溶液を用いるのではなく、調製や使用がより簡便な組成物を用いることが望ましい。
このような調製や使用がより簡便な組成物を用いたエンドトキシンの洗浄または除去技術は、例えば、有効期限が短いために需要に応じた高頻度の製造が要求される医薬品、例えば短半減期放射性核種を用いた放射性医薬品の製造に好適である。特に、半減期が数時間以内の超短半減期陽電子放出核種を用いる陽電子放射断層画像(Positron Emission Tomography、以下PETという)診断法に用いられる薬剤の製造にはより適した技術である。
本発明は、医療用具や医薬品製造器具及び装置等の固体表面に付着又は吸着したエンドトキシンを除去する技術であって、生体由来でない化学的な試薬であって且つ滅菌処理できる成分を用い、水や生理食塩水よりも効率良くエンドトキシンを除去可能な方法を提供することを目的とする。
発明の開示
PET診断法の代表的薬剤である2−[18F]−フルオロ−2−デオキシ−D−グルコース(以下18F−FDGという)はHamacherらの方法(Hamacher,K.et al.,J.Nuclear Medicine,27:235−238,1986)により製造されることが多いが、本発明者らは、この製造法にしばしば用いられるイオン遅滞樹脂に付着又は吸着したエンドトキシンを除去する方法について鋭意研究した結果、水や生理食塩水では通常除去されないエンドトキシンが、糖類を成分として含む溶液によって極めて効率良く溶出または抽出等によって除去され、医療用具や医薬品製造器具及び装置等の固体表面に付着又は吸着したエンドトキシンの除去に利用できることを見出し、本発明を完成するに至った。
かくして、本発明の一局面によれば、糖類を有効成分として含有することを特徴とする、固体表面に付着又は吸着したエンドトキシンの除去用組成物が提供される。
したがって、本発明の除去用組成物の溶液を、エンドトキシンの除去が必要とされる医療用具や医薬品製造器具及び装置の他、各種の固体表面に接触せしめて回収することにより、エンドトキシンを効果的に除去することができる。
したがって、本発明の別の局面によれば、エンドトキシンを除去すべき固体表面と前記組成物の溶液とを接触せしめて、該固体表面に付着又は吸着したエンドトキシンを除去することを特徴とするエンドトキシンの除去方法が提供される。
本発明の除去用組成物の溶液を上記固体表面に接触せしめる方法は、当該固体表面の種類に応じて適宜選択すればよく、例えば、本発明の除去用組成物の溶液に固体表面を浸漬したり、該溶液で固体表面を洗浄したり、該溶液を適当な方法で滅菌された布に含浸させ、当該布で固体表面を拭うなどの方法が考えられる。
また、本発明の組成物の溶液を用いて上記のような浸漬、洗浄、拭取操作等を行なうことによって、医療用具や医薬品製造器具または装置等の固体表面に付着または吸着したエンドトキシンを上記溶液中に移行せしめ、該溶液中にエンドトキシンを溶出もしくは抽出できるので、本発明の組成物は、該固体表面のエンドトキシンによる汚染の程度を検出および測定する際の検出液の獲得に有用である。
したがって、本発明のさらに別の局面によれば、エンドトキシンが付着または吸着した固体表面と前記組成物の溶液とを接触せしめて、該固体表面から脱離または脱着したエンドトキシンを含有する前記組成物を得ることを特徴とするエンドトキシンの抽出方法が提供される。
以下、本発明の実施の形態について説明する。
本発明の組成物で使用される糖類は、単糖類もしくはオリゴ糖類またはその誘導体が挙げられる。また、これらの糖類は、その塩または水和物でもよい。また、糖類はD体またはL体のいずれでもよい。具体的には、単糖類は、一般的には六炭糖が好ましく、例えば、グルコース、ガラクトース、マンノース、フルクトースなどの他、グルコサミン、ガラクトサミン、グルコサミン塩酸塩、ガラクトサミン塩酸塩、N−アセチルグルコサミン、N−アセチルガラクトサミンなどのアミノ糖、その誘導体および塩が挙げられる。オリゴ糖としては、二糖から六糖の大きさのものが挙げられ、好ましくは、マルトース、スクロース、ラクトースなどの二糖である。このうち、特に好ましい糖類はグルコース、グルコサミン塩酸塩、N−アセチル−D−ガラクトサミンおよびマルトースであり、最も好ましくはグルコースである。
本発明の除去用組成物は、使用時には適当な溶媒に溶解した溶液として調製されるが、溶媒に予め溶解させた溶液の形態で提供してもよく、また、使用時に溶媒中に溶解して使用できるように凍結乾燥させた形態で提供してもよい。また、本発明の組成物の凍結乾燥品と溶媒とを組み合わせたキットとして提供してもよい。
溶媒としては、生理学的、薬学的または化学的に許容可能なものであれば特に限定されず、例えば水、生理食塩水、リンゲル液のような電解質の組成物、注射用液の他、各種緩衝液が挙げられる。緩衝液としては、リン酸緩衝液、クエン酸緩衝液等の他、フタル酸、リン酸、ホウ酸、クエン酸、コハク酸、酒石酸、乳酸、酢酸、塩化アンモニウム、炭酸、トリス(ヒドロキシメチル)アミノメタンおよびこれらの塩より構成される緩衝液が挙げられる。
また、本発明の組成物は、必要に応じて、生理学的、薬学的または化学的に許容可能な各種の添加剤を含有することができる。添加剤としては、例えば、酸、塩基などの各種pH調製剤、各種緩衝溶液、デキストラン10、デキストラン40、デキストリン、シクロデキストリン、乳酸ナトリウム、各種アミノ酸、メタノール、エタノール、ジメチルスルホキシド、N,N−ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン等などが挙げられる。
本発明において、溶液中の糖類の濃度は、糖および溶媒等の種類、または固体表面へのエンドトキシンの付着や吸着の程度等によって異なり必ずしも一定とすることはできないが、通常は、0.1〜15g/100ml(以下、w/v%と記す)、好ましくは0.5〜10w/v%程度である。濃度が低すぎるとエンドトキシンの除去効果が低下し、濃度が高すぎると除去操作後に残留した糖類を洗浄するために多量の洗浄液が必要となり、操作が煩雑になる。
本発明の組成物の溶液は、糖類を必要に応じて添加剤とともに溶媒に溶解することによって調製できるが、その性質上、エンドトキシンを含まないか失活させたものとして調製しなければならない。本発明の組成物からエンドトキシンを除く方法としては、糖類溶液から限外ろ過膜またはエンドトキシン吸着体などによりエンドトキシンを除去する方法や、限外ろ過膜などによりエンドトキシンを予め除去した水を用いて糖類溶液を調製する方法などが挙げられる。また、滅菌された市販の糖類溶液、例えば、日本薬局方ブドウ糖注射液(5w/v%、10w/v%等)をそのまま用いることもでき、この場合、特別な調製を要さないので好都合である。また、かかる市販の糖類溶液を、適宜、上記溶媒を用いて無菌的に希釈して用いてもよい。
かくして、適当な固体を本発明の組成物の溶液中に浸漬させたり、該固体の表面を該溶液で洗浄することにより、該固体の表面に付着または吸着したエンドトキシンは該表面から脱離または脱着して該溶液中に移行するため、効果的に除去される。また、浸漬または洗浄に用いた溶液を回収することによって、固体表面に付着していたエンドトキシン量を検出又は測定することもできる。すなわち、本発明の組成物は、エンドトキシン検出液としても用いることができる。
上記のようにして回収された溶液を、自体公知のエンドトキシン測定法、例えば、カブトガニの血球成分抽出液(以下、AL溶液と略記する。)を用いるリムルステスト法等に測定用サンプルとして供することにより、固体表面のエンドトキシンによる汚染の度合を正確に測定できる。AL溶液としては、例えば、リムルス属(Limulus)、タキプレウス属(Tachypleus)などのカブトガニの血球から抽出されたもので、エンドトキシン又はβ−1,3−グルカンとの反応により凝固が生じるものを使用できる。一般には、生化学工業(株)及び和光純薬工業(株)等から市販されている凍結乾燥品をもとに調製したものが使用可能である。リムルステスト法によってサンプル中のエンドトキシン濃度を測定する方法としては、日本薬局方に記載されているような通常用いられる方法であれば特に限定されることなく使用可能である。
本発明のエンドトキシン除去または抽出方法の対象となる固体表面としては、医療用具、医薬品製造器具、医薬品製造装置などの表面が挙げられる。医療用具としては、例えば、ディスポーザブル注射針、ディスポーザブル注射筒、ディスポーザブル輸血器具及び輸液器具、ディスポーサブル採血用器具、人工心肺用ディスポーサブルセット、医療用人工血管、人工心臓弁、心臓ペースメーカー、透析型人工腎臓装置のほか、エンドトキシン試験に使用するディスポーサブル実験器具(滅菌済みピペット、滅菌済み試験管、滅菌済みピペットチップ等)などが含まれる。医薬品製造器具および医薬品製造装置としては、例えば、イオン交換樹脂等の精製や分離に用いる樹脂、逆浸透膜、ナノろ過膜、限外ろ過膜、精密ろ過膜、電気透析膜などの精製や分離に用いる膜などが挙げられる。また、本発明は、有効期限が短いために需要に応じた高頻度の製造が要求される医薬品の製造器具および装置からエンドトキシンを除去するために有用であり、特に、2−[18F]−フルオロ−2−デオキシ−D−グルコースの合成装置及び/または器具に付着または吸着したエンドトキシンを効果的に除去するために好都合である。
実施例
以下に、実施例を挙げて本発明を更に具体的に説明するが、本発明はこれら実施例により何ら限定されるものではない。なお、以下の実施例において、特に断らない限り、糖類溶液の濃度表示は「w/v%」とする。
試験に用いた試薬および器具
注射筒、三方活栓及びカラム等は市販の滅菌済使い捨て器具を用いた。ガラス器具は洗浄後注射用水を通し、250℃、90分乾熱滅菌処理したものを用いた。
糖類の溶液については市販品を適宜希釈するか、市販品特級試薬を注射用水に溶解し、下記実施例でイオン遅滞樹脂に通過させるエンドトキシン除去用溶液として用いた。これらの溶液の調製はすべて無菌的な操作によって行った。
日本薬局方に基づいたエンドトキシン試験用試薬は、以下のものを用いた。
エンドトキシン標準溶液(10000EU/ml):エンドトキシン10000標準品(日本薬局方標準品)へエンドトキシン試験用水適量を加え溶解し、エンドトキシン標準溶液(10000EU/ml)を調製し、これを原液とし、適宜希釈して用いた。なお、「EU」はエンドトキシン単位を意味する。
ゲル化法:リムルスES−II テストワコー(和光純薬製)
比濁法:リムルスES−II テストワコーまたはリムルスES−J テストワコー(和光純薬製)
イオン遅滞樹脂はバイオラッド製イオン遅滞樹脂AG11A8を用いた。
実施例1
グルコース溶液によるイオン遅滞樹脂の洗浄−1
オートクレーブ滅菌(120℃、20分)したカラムに、イオン遅滞樹脂を無菌的に充填した。カラムは2本作成した。
2本のカラムに注射用水100mlを通し、コンディショニングを行なった。その後、1本のカラム(以下カラム1とする)には注射用水10mlを通し、もう一本のカラム(以下カラム2とする)には0.175w/v%グルコース溶液10mlを通した。それぞれのカラムからの各溶出液についてエンドトキシン試験(ゲル化法)を行なった。ゲル化法は日本薬局方の方法に従い実施した。結果を表1に示す。
表1のカラム2の結果から、0.175w/v%グルコース溶液をカラムに通すことによって、エンドトキシンが溶出されていることが確認された。一方、注射用水を通したカラム1の溶出液からはエンドトキシンは検出されなかった。このことから、注射用水100mlを用いたコンディショニングではカラムから溶出されなかったエンドトキシンが、この後に0.175w/v%グルコース溶液10mlを通すことによって溶出されたといえる。
実施例2
グルコース溶液によるイオン遅滞樹脂の洗浄−2
オートクレーブ滅菌(120℃、20分)したカラムに、イオン遅滞樹脂を無菌的に充填した。カラムは2本作成した。
1本のカラム(以下カラム3とする)に注射用水を通し、もう1本のカラム(以下カラム4とする)に0.175w/v%グルコース溶液を通した。各溶出液は100ml、200ml、500ml及び1000ml溶出された時点でサンプリングし、それぞれサンプリング番号1,2,3及び4とした。サンプリングした溶出液について比濁法によってエンドトキシン濃度の測定を行なった。比濁法は日本薬局方の方法に従い実施した。結果を表2に示す。
表2に示すように、カラム4の結果から0.175w/v%グルコース溶液を流すことによって、最初のフラクション中(サンプリング番号1)にエンドトキシンが溶出されていることが確認された。また、エンドトキシンのカラムからの抽出は比較的速く、サンプリング番号2から4においては、ほとんどエンドトキシン濃度が認められないレベルになっていることが観察された。一方、カラム3の結果が示すように注射用水を流しても、エンドトキシンは溶出されなかった。
参考例
エンドトキシン混入イオン遅滞樹脂カラムの作成
ビーカーにイオン遅滞樹脂(AG11A8)20gをとり、注射用水適量を加え膨潤させ、さらに、上記の如く調製したエンドトキシン標準溶液(10000EU/ml)を0.4ml添加した。これを一昼夜攪拌しながら4℃で保管した。このイオン遅滞樹脂適量をオートクレーブ滅菌(120℃、20分)したカラムに無菌的に充填した。
実施例3
イオン遅滞樹脂中に含まれるエンドトキシンの除去効果の測定:イオン遅滞樹脂からのエンドトキシンの除去操作−1
参考例にしたがって作成したイオン遅滞樹脂カラムを2本用意し、1本のカラム(以下カラム5とする)には注射用水5mlを、もう一本のカラム(以下カラム6とする)には10w/v%グルコース溶液5mlを通過させた。それぞれのカラムからの各溶出液を滅菌済みのビーカーに0.5mlずつ合計10フラクション分取した。これらのフラクションのそれぞれをフラクション番号1から10とした。
カラム中に残留したエンドトキシンを確認するための洗浄操作
次いで、前記カラム5および6に10w/v%グルコース溶液7.5mlを通過させた。それぞれのカラムからの各溶出液を滅菌済みの試験管に0.5mlずつ合計15フラクション分取した。これらのフラクションのそれぞれをフラクション番号11から25とした。
各フラクションのエンドトキシン濃度測定
カラム5及び6から上記のように得られたフラクション番号4,5,6,7,10,13,16,19,22及び25の溶液について、それぞれエンドトキシン濃度の測定を実施した。エンドトキシン試験は日本薬局方の方法に従い比濁法にて行った。
エンドトキシン濃度測定値の補正
溶出液についてのエンドトキシン試験の測定値は、阻害補正を行った。阻害補正は以下のようにして行った。各々の糖類溶液にエンドトキシン標準溶液希釈液(0.5EU/ml)をサンプル液が0.25EU/mlとなるように添加し、この溶液のエンドトキシン濃度を測定した。一方、注射用水にもエンドトキシン標準溶液希釈液(0.5EU/ml)を同様に添加し、この溶液のエンドトキシン濃度を測定した。エンドトキシン標準溶液希釈液(0.5EU/ml)を加えた各糖類溶液のエンドトキシン濃度のそれぞれについて、エンドトキシン標準溶液希釈液(0.5EU/ml)を加えた注射用水のエンドトキシン濃度を百としたときの割合を百分率(%)を求め、これを阻害率(%)とした。各フラクションのエンドトキシン濃度をこの阻害率で補正した。注射用水のエンドトキシン試験の測定値については、補正は行なわなかった。
表3に、カラム5および6のそれぞれの各フラクション4、5、6、7および10中に含まれるエンドトキシン濃度(補正後の値)を示す。
表3から、10w/v%グルコース溶液をカラムに通した場合、溶出するエンドトキシン量はフラクション4から7までのエンドトキシン濃度の変化が示すように、フラクションごとに急速に減少していった。このことから10w/v%グルコース溶液によって、速やかにエンドトキシンが除去されてゆく様相が観察された。
この結果より、注射用水を用いるよりも10w/v%グルコース溶液を用いてカラム中のエンドトキシンを溶出させるほうが、より速やかにエンドトキシンを溶出できることが確認された。
カラム内になお残留しているエンドトキシンの有無を示すために、表4に、カラム5および6のそれぞれの各フラクション13,16,19,22及び25中に含まれるエンドトキシン濃度(補正後の値)を示す。
フラクション1〜10の溶出液に注射用水を用いたカラム5では、その後の10w/v%グルコース溶液を溶出液としたフラクション11〜25にエンドトキシンが溶出されたので、フラクション1〜10の注射用水によるエンドトキシン除去が充分でなかったといえる。
一方、フラクション1〜10の溶出液に10w/v%グルコース溶液を用いたカラム6では、その後の10w/v%グルコース溶液を溶出液としたフラクション11以降にほとんどエンドトキシンが溶出されなかった。
このように、上記イオン遅滞樹脂中のエンドトキシンを除去するための溶液として、注射用水を用いるよりも、10w/v%グルコース溶液を用いたほうが、より有効にエンドトキシンを溶出できることが確認された。
実施例4
イオン遅滞樹脂中に含まれるエンドトキシンの除去効果の測定:イオン遅滞樹脂からのエンドトキシンの除去操作−2
参考例で作成したイオン遅滞樹脂カラム(以下カラム7とする)へ、1w/v%グルコース溶液5mlを通過させた。このカラムからの溶出液を滅菌済みのビーカーに0.5mlずつ合計10フラクション分取した(フラクション番号1から10)。
各フラクションのエンドトキシン濃度測定
得られたフラクションのうち、フラクション番号4,5,6,7及び10の溶液について、それぞれエンドトキシン濃度の測定を実施した。エンドトキシン試験及び測定値の阻害補正は実施例3の方法と同様に行った。その結果を表5に示す。
表5より、1w/v%グルコース溶液をカラムに通すことによって、イオン遅滞樹脂カラム中のエンドトキシンが各フラクションへ溶出され、その量はフラクション4から7までのエンドトキシン濃度の変化が示すように、フラクションごとに急速に減少してゆき、上記イオン遅滞樹脂カラムから速やかにエンドトキシンが除去、抽出されてゆく様相が観察された。
実施例5

Endotoxin concentration measurement of each fraction
Among the obtained fractions, the endotoxin concentration was measured for the solutions of fraction numbers 4, 5, 6, 7 and 10. The endotoxin test and the inhibition correction of the measured values were performed in the same manner as in the method of Example 3. The results are shown in Table 7.
Example 7
Measurement of removal effect of endotoxin contained in ion retarding resin: operation for removing endotoxin from ion retarding resin-5
Two ion-retarding resin columns prepared according to the reference example were prepared, and 5 ml of 1 w / v% N-acetyl-D-galactosamine solution was added to one column (hereinafter referred to as column 11). (Hereinafter referred to as column 12) was passed 5 ml of a 10 w / v% N-acetyl-D-galactosamine solution. A total of 10 fractions of 0.5 ml of each eluate from each column were collected in a sterilized beaker. Each of these fractions was designated fraction numbers 1-10.

Among the obtained fractions, the endotoxin concentration was measured for the solutions of fraction numbers 4, 5, 6, 7 and 10. The endotoxin test and the inhibition correction of the measured values were performed in the same manner as in the method of Example 3. The results are shown in Table 8.
From Table 6, Table 7 and Table 8, 1 w / v% D-(+)-glucosamine hydrochloride solution, 1 w / v% maltose solution, 10 w / v% maltose solution, 1 w / v% N-acetyl-D-galactosamine Even when the solution and the 10 w / v% N-acetyl-D-galactosamine solution were used as endotoxin removal solutions, the same results as the glucose solutions shown in Tables 1 to 5 were obtained. That is, by passing these solutions through the column, the endotoxin in the ion-retarding resin column is eluted into each fraction, and the amount rapidly decreases with each fraction as shown by the change in the endotoxin concentration from fraction 4 to 7. As a result, it was observed that endotoxin was quickly removed and extracted from the ion-retarding resin column.
Furthermore, 10 w / v% maltose solution is used for removing endotoxin than 1 w / v% maltose solution, and 10 w / v% N-acetyl-D-galactosamine solution is used than 1 w / v% N-acetyl-D-galactosamine solution. When used as a solution, the endotoxin elution profile was rapid, and it was also observed that the rate of decrease was faster in the higher concentration solution.
Industrial applicability
According to the present invention, it has become clear that endotoxin adhering to or adsorbing to a solid surface can be removed using a saccharide solution. Therefore, when the sterilized saccharide solution is brought into contact with the surface of a medical device or a pharmaceutical production device and apparatus, endotoxin adhering to or adsorbing on the surface can be efficiently removed. According to the present invention, it is possible to effectively and easily remove even those that were difficult to elute or extract by the conventional removal method, and further, complicated post-treatment after the removal is unnecessary, and the pharmaceutical manufacturing site This is useful.
