Abstract

PURPOSE: To easily, stably and inexpensively discriminate the anti-A anti-B agglutinin in a serum to be inspected by adding the dyed red blood cel ghost obtd. from the human-derived red blood cells to the serum to be inspected and discriminating the agglutionating thereof.
CONSTITUTION: The human red blood cells are first collected from a person having the distinct blood group and are subjected to a bromelin treatment. The resulted liquid is centrifugally separated to separate the red blood cells. Red blood cells are throughly washed by adding a physiological soline. The resulted clean red blood cells are dispersed into a suitable hyprotoni soln. and are thereafter subjected to a centrifugal sepn. again by which the red blood cell ghosts are insolated. The red blood cell ghosts are dyed. The dyes which are used for the dyeing of tissues, cells and clinical inspections or are used for food, medical supplies, and cosmetics are adequate. Sealing may be executed before or after or simultaneously with the dyeing. The red blood cell ghosts may be subjected to an immobilization treatment by using a known immobilizing agent. The dyed red blood cell ghosts obtd. in such a manner are added to the serum to be inspected and the agglutination thereof is discriminated, by which the blood group is discriminated.
