Drains in bathroom sinks, bathtubs, and showers can become clogged when deposits, including hair, accumulate in areas such as drains, sinks, and traps, thereby blocking the proper drainage of water. There are various products available to dissolve and remove such deposits. Most common drain cleaning products contain strong caustics such as sodium hydroxide.

The alkali saponifies any fatty material present in the deposit, converting it into a water-soluble soap or softened, water-dispersible material. When clogged by hair, caustic acts as a decomposer, but it is only partially effective as shown from laboratory simulations. On top of that,
Caustic is a poison that harms many conduit materials and damages people who come in contact with it.

That is, if the drain cleaner has to pass through a column of standing water to reach the blockage (which often happens),
It is possible that some of the active ingredient will dissolve in the water away from the blockage site. This part is wasted, thereby reducing the active ingredient concentration in the blockage area.

The harms and shortcomings of these common methods have led to the search for alternative and better ways to clean drain lines clogged with hair-containing deposits. An alternative to this is
was the use of an enzyme-containing composition. Enzymes can convert common substances that clog drains into water-soluble substances that are easier to remove. The disadvantage of using this is its short shelf life, which is often due to interactions between the various components of the enzyme system. This interaction is facilitated at high temperatures (such as those encountered during transportation of enzyme preparations).

Recently, drain cleaner compositions have been created that are site-specific and contain a sequence of active ingredients to enzymatically dissolve proteins. These compositions comprise a plurality of water-soluble beads, each bead consisting of a mixture of at least one active drain cleaning component dispersed in a water-soluble polymer, and prior to dissolution of the polymer, no interacting components are present. unable to react to each other. These compositions have been found to be effective in unclogging drains clogged with hair or hair-containing deposits. However, improvements are desirable to further increase the activity of proteolytic enzymes in drainage pipes.

According to the present invention, the hair-containing deposits are clogged with hair-containing deposits comprising an amount of a proteolytic enzyme, a disulfide reducing agent, and an alkali metal bisulfite compound sufficient to enhance the enzyme activity. A composition for cleaning drains, comprising a plurality of water-soluble beads, each of the beads comprising an enzyme, a reducing agent-free bisulfite compound, or a combination thereof, dispersed in a water-soluble polymer. A composition is disclosed in which the enzyme and the reducing agent are substantially unable to interact with each other prior to dissolution of the polymer, and the composition is maintained at a pH that promotes hair modification.

In these compositions, the disulfide reducing agent serves to crosslink cysteine and hair proteins and break the disulfide bonds that form the crystal structure. Cross-linked crystal structures are highly resistant to single proteolytic enzymes, but once the disulfide bonds are broken, proteolytic enzymes can act to break the covalent backbone of the protein (i.e., the protein's peptide (hydrolyzes bonds)

The inventors have discovered that the efficiency of these compositions is increased by adding an alkali metal bisulfite compound to the compositions. A preferred compound is sodium bisulfite. It has been theoretically demonstrated that, in a concentration range of , these bisulfite compounds alter the proteolytic enzymes in the composition, increasing their activity. Generally, the amount of bisulfite added to enhance enzyme activity is approximately
ooi to about 0.1 weight percent. If a bisulfite compound is added at a concentration outside this range, it usually has no appreciable effect on enzyme activity or appears to inhibit the activity. When the activity enhancer is sodium bisulfite, it is preferably added at about 0.04% by weight of the final composition.

In one embodiment of the invention, the bisulfite compound is incorporated into the composition by mixing with alkaline protease before being dispersed in a water-soluble polymer and formed into beads. Typically, the weight ratio of protease to alkali metal bisulfite will range up to about 10:1:1000:1, more preferably up to about 50:1:500:1.

When a bisulfite compound is mixed with an alkaline protease, it not only increases the activity of the protease (but also serves as an antimicrobial agent). Containing beads show signs of microbial growth over time, but the addition of an alkali metal bisulfite compound, such as sodium bisulfite, can prevent or slow microbial growth on the beads during storage.

The protease 9 bisulfite compound and disulfide reducing agent may be contained in separate beads or together in the same bead. In the latter case, the active ingredients may be layered in the bead in such an order that it is possible to determine the order in which the ingredients arrive at the mass. For example, by mixing proteolytic enzymes and bisulfite compounds together,
An inner layer is formed with the polymer and then coated with an outer layer containing a reducing agent. When the beads are placed in the water that has collected in the drain, the outer layer of the beads dissolves first, releasing the disulfide reducing agent toward the clog. When the reducing agent acts on the cysteine bonds in the hair, the rest of the beads dissolve and release the enzyme, which can attack the hair. Alternatively, the disulfide reducing agent, proteolytic enzyme 9 bisulfite and optional components (e.g. buffers) can be combined on the same beads.
It can also be uniformly dispersed throughout the polymer.

An example of a suitable water-soluble polymer is polyethylene glycol (PEG) with a molecular weight of about 6,000 to about 20,000; higher molecular weight PEGs can be used to connect two or three smaller polymer chains with an epoxy linker. Manufactured by Generally, the amount of polymer in each bead is about 40~
About 99% by volume, more preferably about 60 to about 80%. The remaining portion consists of the active ingredient and water. The actual polymer concentration in the various beads will depend on the nature of the component, i.e. whether it is an enzyme, a detergent, or a reducing agent.
It also depends on whether it is necessary or desirable to produce a final product in which the different components react in an orderly manner, ie, one after the other, in the drain.

Enzymes and other components may be liquid or solid. - By way of example, the enzyme source may be a published liquid medium or a dry enzyme powder. In either case, the polymer is melted and then mixed with the liquid or solid components of the drain cleaning composition. Beads or pellets can then be formed in a variety of ways. For example, the polymer component mixture can be formed into droplets and then solidified again.

The Pharmacia K 16 column was connected to a heating water bath and the temperature was adjusted accordingly (see Table ■). The active ingredient was then added to the PEG and mixed well. For enzyme beads, the enzyme solution is added just before bead production and then added to
The mixture was injected into the column with stirring for ~2 minutes. Additional water was added as indicated in the table. To make Enzyme-3BS beads, the same general method was used, except that SBS was added to the enzyme solution before mixing it with PEG. In the case of SDS beads, the mixture was stirred slowly to prevent foaming of the detergent, which causes foam problems in the column. Sodium thioglycolate (Na-
TGA) was synthesized by adding sodium hydroxide to thioglycolic acid. p of the final composition of the drain pipe section
Excess base was added so that H was adjusted to increase enzyme activity. As a first step, NaOH pellets were ground and added slowly (on ice) to the liquid thioglycolic acid and mixed until the addition was complete. The mixture was ground again and stored in a plastic container to maintain the stability of the compound until use in bead production. To prepare the beads, Na-TGA was mixed with an appropriate amount of water (see Table ■). The resulting mixture was ground and added to PEG. Prepolymer solutions of each component were individually injected into the column, and a lid was attached to the top of the column. Then pump the air with a Masterflex pump (pump head size 701).
4 and using matching tubing) through the central valve of the top lid to create internal air pressure. The prepolymer solution is then forced onto the column bottom cap, connected to a stainless steel valve, and exited through a needle of appropriate gauge as shown in Table 3. The column bottom lid tubing connector, valve and needle are wrapped with heating tape and adjusted to the same temperature as the column. Air flow rate is adjusted to ensure individual bead production. The droplet from the needle is received on a rotating disk and beads are formed. Cool air is directed over the beads to promote rapid solidification. Alternatively, a cooled surface can be used. The beads then pass through a stationary scraper. The scraper removes the beads from the rotating disk and deposits them in a collection container.
