Abstract

PURPOSE: An enzyme preparation obtained from a microorganism in Nocardia is used to oxidize cholesterol into Δ4-cholestenone and hydrogen peroxide and the formed hydrogen peroxide is measured to determine the cholesterol in biological fluid.
CONSTITUTION: Nocardia NCIB10554 or NCIB10555 is cultured in a medium containing glycerol as a carbon source and cholesterol, then an enzyme preparation having cholesterol oxidase activity is obtained from the culture cells by surfactant extraction, cell crushing and removal of the residue. The resultant preparation and at least one of reagents for determining the hydrogen peroxide such as a tetravalent titanium compound and xylenol orange are brought into contact with a sample containing cholesterol such as serum to determine the hydrogen peroxide formed by the oxidation of the cholesterol in the sample.
