Abstract
NEW MATERIAL:The titled compound of formula I (Ar is aryl; R1 is H or lower alkyl; R2 is H or alkyl).
EXAMPLE: Ethyl α-(3-benzoylcyclohexylidene)propionate.
USE: Intermediates for the preparation of ketoprofen having lenitive and anti-inflammatory activities. Ketoprofen or its analogs are prepared in high yield in a short process.
PROCESS: A compound of formula II [R4 is disubstituted amino, group of OR5 or SR6; (R5 is protecting group of OH; R6 is protecting group of thiol)] is made to react with 2-cyclohexenone in the presence of a basic catalyst (e.g. NaOH, KOH etc.) and then hydrolyzed to give a compound of formula III. The compound obtained is made to react with a compound of formula IV (R3 is lower alkyl) in the presence of a base to afford a compound of formula V, which is then made to react with a compound of CuX2 (X is halogen) to give the titled compound of formula I.
