Abstract

PURPOSE: To regenerate silica gel simply and sufficiently, by washing silica gel used in developing and separating an isoprenoid high-molecular oxygen-containing hydrocarbon by a certain combined solvent.
CONSTITUTION: Silica gel used in developing and separating an isoprenoid high- molecular oxygen-containing hydrocarbon (e.g.; vitamin K) by a column chromatography is regenerated as described hereinafter. That is, this silica gel is washed by a mixed solvent system of (A) an org. solvent having high eluting capacity of the aforementioned exygen-containing hydrocarbon (e.g., diethyl ether) and (B) an org. solvent having low eluting capacity (e.g.; hexane) for regeneration. In this case, it is necessary to adjust the amount of the solvent A to 50W80vol% against a total solvent amount. After the washing is finished, the silica gel is subjected to the replacement treatment so as to be used in a succeeding development and separation by using the solvent B or a solvent used in development. As a result, the aforementioned regenerating treatment can be carried out over, ten times or more without taking the trouble of drawing out silica gel and carrying out the treatment such as heating.
