Claims (1)
- 【特許請求の範囲】 1.ヒトp75TNF−R遺伝子のプロモーター配列。 2.ヒトp75TNF−R遺伝子の、約2.0kbpの配列内に含まれ、第1エ キソンの5′上流に位置する5′上流プロモーター配列ならびに第1および第2 エキソンの間の第1イントロンに位置し、約1.9kbpの配列内に含まれるプ ロモーター配列から選ばれる請求の範囲第1項記載のプロモーター配列。 3.ヒトp75TNF−Rをコードするゲノムクローンの5′上流プロモーター 配列であり、2.0kbpのNcoIフラグメントからなる請求の範囲第2項記 載の配列。 4.図2Aに記載のヌクレオチド配列の少なくともヌクレオチド1335から1 527にわたる配列からなる請求の範囲第3項記載の配列。 5.ヒトp75TNF−R遺伝子をコードするゲノムクローンのイントロンプロ モーター配列であり、1.9kbpのSmaIフラグメントからなる請求の範囲 第2項記載の配列。 6.図2Bに記載の配列の少なくともヌクレオチド1569から1768にわた る配列からなる請求の範囲第5項記載の配列。 7.図2Bに記載の配列の少なくともヌクレオチド1569から1827にわた る配列からなる請求の範囲第6項記載の配列。 8.イントロンプロモーター配列の上流の請求の範囲第5項記載の1.9kbp のSmaIフラグメントの配 列内に位置する転写抑制領域を含む配列。 9.図2Bに記載の配列のヌクレオチド1から1569にわたる配列からなる請 求の範囲第7項記載の転写抑制領域。 10.請求の範囲第1〜8項のいずれかに記載の配列内に含まれる配列モチーフ。 11.転写因子に結合可能であって、該転写因子がプロモーターに存在するモチー フと結合可能な因子または転写抑制領域に存在するモチーフと結合可能な因子の 群から選ばれる請求の範囲第9項記載の配列モチーフ。 12.請求の範囲第9項または第10項記載の少なくとも1つの配列モチーフとさ らに、片側または両側にそれを並列する配列部分および/または少なくとも1つ のほかの配列モチーフとそれとを連結する配列部分とを含むモチーフ領域。 13.所望のモチーフの上流および/または下流の望ましくない配列部分の従来の 手段による欠失、えられたモチーフを、任意には適切な制御配列とともに、ベク ターへ挿入すること、該ベクターを適切な原核生物株へ挿入すること、該株を培 養することおよび該モチーフを単離することからなる配列モチーフの製造法。 14.所望のモチーフ領域の上流および/または下流の望ましくない配列部分の従 来の手段による欠失、えられたモチーフを、任意には適切な制御配列とともに、 ベクターへ挿入すること、該ベクターを適切な原核生物株へ挿入すること、該株 を培養することおよび該モチーフ領域を単離することからなるモチーフ領域の製 造 法。 15.請求の範囲第12項または第13項記載のモチーフまたはモチーフ領域と結 合する因子の単離法であって、プローブとしてモチーフまたはモチーフ領域から なる配列を用いてヒト遺伝子ライブラリーをスクリーニングすることからなる単 離法。 16.請求の範囲第12項または第13項記載のモチーフまたはモチーフ領域と結 合する因子の単離法であって、クロマトグラフィーマトリックスに付着するモチ ーフ領域を用いるアフィニティ精製からなる配列を用いてヒト遺伝子ライブラリ ーをスクリーニングすることからなる単離法。 17.ヒト遺伝子ライブラリーがゲノムライブラリーである請求の範囲第14項記 載の方法。 18.ヒト遺伝子ライブラリーがcDNAライブラリーである請求の範囲第14項 記載の方法。 19.プローブとして、請求の範囲第9項または第11項記載のモチーフもしくは モチーフ領域または請求の範囲第14項から第16項のいずれかに記載の方法に よりえられる因子を用いてクローン化される遺伝子。 20.請求の範囲第9項記載のモチーフからなる医薬組成物。 21.請求の範囲第11項記載のモチーフ領域からなる医薬組成物。 22.TNFの有害な効果を抑制するための請求の範囲第19項記載の医薬組成物 。 23.TNFの有害な効果を抑制するための請求の範囲第20項記載の医薬組成物 。 24.p75TNF−R遺伝子配列によってコードされるタンパク質であって、該 タンパク質が第1エキソンと第2エキソンとの間の第1イントロンに位置するプ ロモーター配列の制御下で発現され、かつ該タンパク質が第1イントロン配列内 のその3′領域で始まるp75TNF−R遺伝子を有する配列によってコードさ れるタンパク質。 25.前記タンパク質がp75TNF−Rの不完全な形態であり、前記タンパク質 が図2Bに示された配列のヌクレオチド1768で始まる配列によってコードさ れる請求の範囲第23項記載のタンパク質。 26.請求の範囲第23項または第24項記載のタンパク質からなる医薬組成物。
