(iv)分子による光の吸収による検出。方式(i)及び
(ii)において、蛍光検出器は次の要件を満たさねばな
らない。(a)励起光線は、バンドの幅よりも実質的に
大きい幅をもってはならない。これは一般に0.1〜0.5mm
の範囲である。このような幅狭い励起ビームを使用する
ことによってバンドの最大分離を可能にする。(b)励
起波長を変化させて種々異なる染料のそれぞれの最大吸
収に適合させ、或いは4種の蛍光発生剤を励起するが蛍
光放出のいずれとも重ならないような単一の幅狭い高強

素子F3(励起フィルタ)、DM(二色鏡)及びF4(遮光フ
ィルタ)を一緒に切替える。この方法においては、励起
光と観察されたエミッション光との両者を変化させる。
第4図に装置は、単一波長の励起の場合に良好な配置で
ある。この装置は、可動部分を必要としない利点を有
し、4種の染料の全てからの蛍光を同時かつ連続的にモ
ニターすることができる。検出の第3の方法上記(ii
i)は、ゲルの底部から標識分子を溶出させてこれらを
たとえば1,2−ジオキシエタンジオンのような化学発光
を励起させる薬剤と結合させ〔エス・ケー・ギル、アン
ドリッチヒミカ・アクタ、第16(3)巻 第59−61頁
(1983);ジー・ジェー・メルビン、Liq・Chrom、第6
(9)巻 第1603−1616頁(1983)〕かつこの混合物を
4種の別々の波長で放出光を測定しうる検出器に直接流
入させる方法である(この検出器は第4図に示したもの
と同様であるが、励起光源を必要としない)。光学発光
におけるバックグランド信号は蛍光におけるよりもずっ
と低く、その結果、信号対ノイズの比率が高くなりかつ
感度が増大する。最後に、吸光度の測定により測定を行

定よりも本質的に低い感度を有することである。

6により集め、これを適当な組み合せの光学フィルタ108
及び110に通過させて散乱励起光を除去し、光電子増幅
管(PMT)112を用いて検出する。シグナルはチャート記
録紙で容易に検出される。DNA配列決定反応は、フルオ
レセイン標識したオリゴヌクレオチドプライマーを用い
て行なわれる。チャート紙におけるピークは、配列決定
反応で合成されかつ電気泳動によりゲル管で分離された
種々異なる長さを有するフルオレセイン標識されたDNA
の断片に相当する。各ピークは10-15〜10-16モルの程度
のフルオレセインを含有し、これは放射性同位元素の検
出を用いる同等な配列決定用ゲルにおいて1バンド当り
に得られるDNA量にほぼ等しい。これは、蛍光標識が配
列決定反応においてオリゴヌクレオチドプライマーから
除去されず或いは分解されないことを照明する。さら
に、これは検出感度が、この手段によりDNA配列分析を
行なうのに充分であることを示している。
