In recent years, a dry-type slide has been developed which can quantitatively analyze a specific chemical component or a formed component contained in the sample solution simply by supplying a small drop of the sample solution (Japanese Patent Publication No. 53-53- 21677, JP-A-55-164356, etc.) has been put to practical use. The use of these slides makes it possible to analyze sample liquids more easily and quickly than conventional wet analysis methods, so its use is particularly useful in medical institutions and laboratories that need to analyze many samples. It is preferable.

In order to analyze the chemical components in the sample solution using such a slide, after the sample solution is weighed and adhered to the slide, it is kept at a constant temperature (incubation) for a predetermined time in an incubator (incubator). Color reaction (dye formation reaction) is performed, and then the irradiation light for measurement containing the wavelength preselected by the combination of the sample component and the reagent contained in the reagent layer of the slide is irradiated to this slide to measure its reflection optical density. With this, quantitative analysis of the above chemical components and the like is performed.

When carrying out such an analysis, a predetermined amount of the sample liquid to be spotted and supplied to the reagent layer of the slide must be accurately weighed and spotted. This is because when the amount of the sample liquid is different from the predetermined amount, the reflection optical density is different and the analysis accuracy is also lowered. Therefore, various pipettes and the like have been devised so that a predetermined amount can be spotted correctly when the sample liquid is spotted and supplied. Such a pipette is, for example, one in which a tip is attached to the tip of the pipette, a predetermined amount of sample liquid is sucked into the tip, and then this predetermined amount of sample liquid is spot-supplied on the reagent layer of the slide. is there. In many such pipettes, a piston / cylinder mechanism is used to suck a predetermined amount of sample liquid into the chip and discharge the sample liquid. In order to aspirate the sample solution into the chip and discharge it to the reagent layer using such a pipette, first insert the tip of the chip into the sample solution, and then use the piston / cylinder mechanism etc. The liquid is sucked and held, and then the tip of the chip is positioned on the reagent layer of the slide, and the sample liquid in the chip is spot-supplied onto the reagent layer by a piston / cylinder mechanism or the like. However, in this case, when the tip of the tip is inserted into the sample solution and then pulled out for spotting, the sample solution adheres to the outer periphery of the tip of the tip, and this adhered sample solution is also spotted on the reagent layer. Is worn
There is a problem that an error occurs in the amount of the sample liquid spotted and the measurement accuracy decreases.

For this reason, in the past, after the sample solution was sucked into the chip by a pipette, the sample solution adhering to the outer periphery of the tip was wiped off, and then the sample solution was supplied by spotting. However, there is a problem in that the wiping work is required and the work efficiency is not improved so much, and it is difficult to automate the wiping work when performing automatic spotting.

(Problems to be Solved by the Invention) Therefore, when the sample solution is sucked into the chip, the present applicant controls the vertical position of the chip and the sample solution surface to insert the tip of the chip into the sample solution. The amount of sample adhering to the tip tip outer circumference is always kept constant, so that the amount of sample to be supplied is always constant. I proposed a spotting device. In this case, in order to keep the insertion amount constant, it is necessary to accurately grasp the position of the tip of the chip and the position of the liquid surface of the sample liquid. However, the tips are often replaced each time for reasons of preventing mixing of sample liquids and hygiene, and in addition, the dimensional accuracy of the tips is often not very high, and the shape is However, it is difficult to accurately grasp the position of the tip attached to the tip of the pipette, and it is difficult to control the insertion amount of the tip. Also, regarding the liquid surface position of the sample liquid, there is a problem that it is difficult to detect the liquid surface position, for example, when bubbles are generated on the liquid surface.

(Means for Solving Problems) From the above, according to the present invention, when the nozzle member provided at the lower end of the suction member is vertically moved relative to the sample liquid, the lower end of the nozzle member is A means for detecting that the lower end of the nozzle body or the lower end of the spotting tip attached to the nozzle body is in contact with the sample liquid is provided, and the insertion amount of the lower end of the nozzle member into the sample liquid is accurately grasped by this means. It is an object of the present invention to provide such a liquid level detection device as possible. As a means for this, the device of the present invention arranges a suction member above the sample container containing the sample liquid so as to be vertically movable by the vertical position control means, and to the nozzle member provided at the lower end of this suction member. The sample liquid can be sucked by the communicating suction means, while the nozzle member is held by the vibration transmission plate capable of vibrating the suction member, and the vibration is applied to the nozzle member by the vibration applying means through the frequency control circuit. Vibration is applied while changing the output voltage, and the extreme value of the output voltage or current value corresponding to the frequency change is detected from the output voltage or current value of the vibration applying means at this time, and the change in the extreme value is read to read the nozzle member. The contact between the sample liquid and the sample liquid is detected, and the liquid surface position of the sample liquid is detected. There are no restrictions on the type of sample liquid and the shape of the nozzle member in this device, and various sample liquids and nozzle members are used, but a prescribed sample liquid is spot-supplied on the reagent layer of the chemical analysis slide. In the automatic spotting device for, when the nozzle member (nozzle body or a spotting tip attached to the lower end of the nozzle body) sucks a predetermined amount of the sample liquid, the contact between the nozzle member and the sample liquid is detected,
It is particularly effective for detecting the liquid surface position and controlling the amount of insertion of the lower end of the nozzle member into the sample liquid.

(Operation) According to the liquid level detection device of the present invention configured as described above,
When the lower end of the nozzle member (nozzle body or spotting tip) comes into contact with the liquid surface, the vibration of the nozzle member imparted by the vibration imparting means receives a resistance due to the sample liquid, so that the output voltage or current of the vibration imparting means changes. This change can be detected as a change in the extreme value of the output voltage or the current with respect to the frequency of the applied vibration, and thus the contact between the nozzle member and the sample liquid can be reliably detected. By doing so, even if bubbles are generated on the surface of the sample liquid, there is a large difference between the resistance due to the bubbles and the resistance due to the liquid, and therefore contact with the liquid surface can be reliably detected. For this reason,
From the position where the contact between the nozzle member and the sample liquid is detected, if the nozzle member is moved down by a predetermined amount, it is easy to insert the lower end of the nozzle member into the sample liquid by a certain depth.

When the fog photometry is performed, the chemical analysis slide 1 is pushed out by the pushing lever 12 onto the carrying / inserting means 40 moved to the position shown by the chain double-dashed line X in FIG. 2 and held there. The conveying / inserting means 40 moves rightward on the rail 49 and moves to a position below the spotting position (position indicated by solid line Y). Then, by the automatic spotting device 50,
The sample in the sample container 52 is placed in the spotting tip 35 at the suction position.
(The suction operation into the spotting tip and the structural details of the spotting arm 30 will be described later), and spotting onto the reagent layer of the slide 1 transported to the position indicated by Y at the spotting position. Supplied. After that, the conveying / inserting means 40 moves in the lateral direction (direction of arrow C) on the rail 49 to face the predetermined storage chamber 21 of the incubator 20, and the slide 1 is inserted into the storage chamber 21. The slide 1 which is kept at a constant temperature (incubation) in the incubator 20 is irradiated with irradiation light and its reflected optical density is measured by a reading head moved below the storage chamber, and a chemical analysis of the sample is performed. When these measurements are completed, the chemical analysis slide 1 is discharged from the storage chamber 21 into the tray 29 by the carrying / inserting means 40. Hereinafter, by repeating the above operation, it is possible to automatically and continuously carry out chemical analysis using a large number of chemical analysis slides.

The electric signal to the vibrator 34 is sent from the controller 90. The operation of the controller 90 will be described with reference to the block diagram of FIG. The electric signal to the oscillator 34 is
This is a signal output from the sweep generator 91 via the voltage controlled oscillator 92, whereby the vibrator 34 oscillates while changing the frequency so as to sweep within a predetermined frequency range (for example, 0 to 1 KHz). That is, the sweep generator 91, the voltage controlled oscillator 92, and the oscillator 34 constitute a frequency control circuit as shown in the figure. This oscillator
The vibration of 34 is transmitted to the nozzle body 33 and the spotting tip 35 via the vibration transmission plate 32, and these also vibrate. In addition,
Since the hose 36 is flexible, there is almost no vibration transmission from the hose 36 to the arm body 31. When the vibrator 34 vibrates in this manner, the voltage (V) across the fixed resistor 93 arranged in series with the vibrator 34 in the frequency control circuit.
Alternatively, the current (I) flowing through this resistance is measured, and this is amplified by the amplifier 94 and sent to the extreme value detecting means 95. The extreme value detecting means 95 detects the above voltage (V) or current (I) which is the output voltage or current of the vibrator 34 corresponding to the above swept frequency, and the relationship between the two is, for example, FIG. Is indicated by a solid line (in the figure, the relationship between voltage (V) and frequency is shown, but since voltage and current are in a proportional relationship, current may be used instead of voltage). That is, the vibration transmitting plate 32, the nozzle body 33, and the spotting tip 35.
From the relationship with the natural frequency of, the voltage (V) has a maximum value Q and a minimum value P for a certain frequency.

At the extreme points P and Q, the lower end of the spotting tip 35 is the sample container.
If it is far from the sample liquid surface of 52, the spotting arm 30
Is constant regardless of the vertical movement of the sample liquid, but when the lower end of the spotting tip 35 comes into contact with the liquid surface of the sample liquid, the resonance point changes due to the resistance of the sample liquid. For example, as shown by the broken line in FIG. The value of the voltage (V) with respect to changes, and the extreme values P and Q are P ′,
Move to the position of Q '. Therefore, the extreme value detecting means 95 stores the extreme values P and Q when the lower end of the spotting tip 35 is away from the liquid surface, and the tip vertical position control means 60 is stored.
The extreme value while the spotting arm 30 is being moved downward is continuously detected by the comparison circuit 96 and the detected extreme value and the stored extreme value are compared.
In comparison, the presence / absence of contact between the lower end of the spotting tip 35 and the liquid surface is detected by the presence / absence of the change in the extreme value, and the liquid surface position is detected. In response to the detection of the presence or absence of the contact with the liquid surface, the motor operation control circuits 97 and 98 cause the pulse motor 61 of the vertical position control means 60 and the suction / discharge means 80.
An operation control signal is sent to the piston drive motor 81, and vertical movement control of the spotting arm 30 and suction / discharge control of the sample liquid to the spotting tip 35 are performed.

The operation of the automatic spotting device configured as described above will be described. First, the sample container 52 containing the sample liquid is placed, and the oscillator 34 is oscillated by a signal from the frequency control circuit of the controller 90. Next, the tip vertical position control means 60 is operated to move the spotting arm 30 in the suction position and the spotting tip 35 attached to the tip of the arm 30 downward.
When the spotting tip 35 is moved down by a predetermined distance and the lower end of the spotting tip 35 comes into contact with the liquid surface of the sample liquid in the sample container 52, the output voltage (V) of the vibrator 34 is changed as shown in FIG. Extreme values P, Q
Changes to P ', Q'. Therefore, by detecting this change, the contact between the spotting tip 35 and the sample liquid can be reliably detected. In addition, depending on the insertion depth of the lower end of the spotting tip 35 into the liquid, the extremal value P ′ after the change from the original extremal values P and Q,
Since the amount of change in Q'is different, it is also possible to detect the insertion depth by this amount of change. With this configuration, it is possible to reliably detect the contact between the lower end of the spotting tip 35 and the sample liquid even when the liquid surface is difficult to see due to bubbles on the liquid surface.

Next, in this way, the spotting tip 35 is moved downward by a predetermined amount h from the position in contact with the liquid surface of the sample liquid,
The lower end of the is inserted into the sample solution by a predetermined depth h. In addition,
The predetermined depth h is preferably as shallow as possible in order to reduce the amount of sample liquid attached to the outer periphery of the lower end of the spotting tip 35. After this, the suction / discharge means 80 is operated to move the piston 84
The sample liquid is sucked into the space 35a of the spotting tip 35 by a predetermined amount by moving a.

As described above, when a specified amount of the sample liquid is sucked into the space 35a in the spotting tip 35, the tip vertical position control means 60 moves the spotting arm 30 upward to lift the spotting tip 35. Thereafter, the tip moving means 70 is used to move the spotting arm 30.
Is rotated about the rotation shaft 31b, and the spotting tip 35 is moved from the suction position to the spotting position. Then, the spotting tip 35 is moved downward by the tip vertical position control means 60, and when the tip thereof is located near the upper portion of the reagent layer portion of the slide 1, the sample liquid in the spotting tip 35 is caused by the action of the suction / discharge means 80. Are slowly discharged to form a liquid bulb at the lower end of the spotting tip 35. Then, the spotting tip 35 descends as it is, and the liquid bulb is spotted and supplied onto the reagent layer portion of the slide 1. After that, if the spotting tip 35 is moved upward, the spotting of the sample solution is completed.

(Effects of the Invention) As described above, according to the present invention, the nozzle member held on the suction body via the vibration transmission plate is applied with the vibration of the predetermined frequency swept by the vibration applying unit (vibrator). Further, since the extreme value of the output voltage (current) of the vibration applying means corresponding to this swept frequency is detected, the lower end of the nozzle means becomes the sample liquid if the change in the extreme value is detected. It is possible to detect whether or not they have come into contact with each other, and it is possible to reliably detect the liquid surface. Particularly, in this case, since it is not necessary to detect the initial position of the lower end of the nozzle means and the initial position of the liquid surface, there is no limitation on the size and shape of the sample container and the amount of liquid to be put in this container, and When the spotting tip is used as the means, there is an advantage that there is no limitation on the shape and size of the spotting tip.
