Abstract

PURPOSE:To readily obtain the titled substance useful as a raw material for cosmetics at a low cost, by heat-treating an extract of a cucumber fruit, removing the formed precipitates and adding sodium citrate. CONSTITUTION:A cucumber fruit is sufficiently crushed and pressed to remove tissual residues and provide a green suspendible extract, which is then heat- treated at 40-55 deg.C for 10-100min (preferably at 45-50 deg.C for 10-30min) to accelerate formation of insoluble pectin and aggregation of chloroplast and afford a clear light yellow extract by a method for filtration, etc. Sodium citrate is then added to the extract so as to provide 0.2-2.0% final concentration to provide a stable cucumber extract having a high concentration of superoxide dismutase (>=200units/ml) without any change with time. The superoxide dismutase is capable of inhibiting toxicity of active oxygen in living bodies and contriving improvement in skin tissues.

りキュウカンバエキスの品質評価については各種の生物
活性で示すことが妥当と考えられていた。しかし、生物
活性の測定には多くの場合繁雑な操作を必要とし、実際
上実行し難いのが現状であった。

カル自身及びこれから生ずる他の活性酸素による障害を
防ぐ機能をもつことから、近年慢性関節リュウマテ、変
形性関節炎、尿路感染症及び放射線療法時の副作用等の
治療に利用する試みがなされている。SODは従来、ウ
シ赤血球より大量に精製され、その性質が明らかにされ
ている他、植物中でもホウレンソウ、リョクトウ、コム
ギ胚芽、サイトウ等に存在が確認されているが、これま
でキュウリ果実中のSODに関する報告は全くなされて
いない。

本発明者らはかかる現状に鑑み、キュウカンバエキス中
の有効成分について、しみ、そばかす、肌荒れなど、皮
膚に対する有効性を指標として鋭意研究の結果、複数の
有効成分の存在を確認したが、その中でSODが重要な
成分の一つであることを発見した。SODは他の有効成
分と協調し、過酸化詣質産生の主原因となるスーパーオ
キシド・ラジカルを消去し活性酸素の毒性を強く抑制す
ることにより、皮膚組織の改善を図るものと考えられる
。また、SODはその含量の測定が極めて容易でありキ
ュウカンバエキスの品質評価の指標として有用であるこ
とが明らかとなった。更にSODが熱、化学処理等に不
安定であることから、キュウリ果実より天然のままの状
態で抽出されていることの証明としても価値のある評価
法である。しかしながら、現在市販されているキュウカ
ンバエキス中のSOD含量を測定したところ、10単位
/ m 1以上のSODを含むキュウカンバエキスは皆
無であり、このことは従来のキュウカンバエキスの製造
にあたりSODは全く考慮されず、製造工程中何らかの
原因で失活していたものと考えられた。そこで、本発明
者らは更に研究を進めた結果、SODを高濃度に含有し
高い有効性を有するキュウカンバエキスの製法を発明す
ることに成功した。

本発明においては、まず上記原料を十分破砕せしめた後

の操作により不溶性ペクチンの生成とクロロプラストの
凝集を促進し、濾過等の方法により容易に清澄な淡黄色
のエキスを得ることができる。また、同時にこの操作に
より高分子物質を沈殿として除去し、得られる製品の抗
原性を軽減すると共ににその安定性を向上させることが
できる。なお、この工程中、SODをはじめ他の有効成
分の失活は全く認められない。更にこのエキスに対しク
エン酸ナトリウムを最終濃度0.2〜2.0%に添加す
ることにより、経時変化のなく、安定でかつSODを高
濃度に含有するキュウカンバエキスを製することができ
る。かくして得られたキュウカンバエキスは更にパラベ
ン等の防腐剤を添加、除菌濾過し実際の使用に供される

熱処理を行い生じる不溶物を濾過した後、得られる清澄
なエキス41kgにクエン酸ナトリウム0.4kgを加
え、更にメチルパラベン0.1kg、エタノール2kg
を加え除菌濾過しキュウカンバエキス43kgを得た。
