a 産業上の利用分野 本発明は、側鎖のカルボキシル基に細胞毒性物
質を多数結合すると共に、主鎖の片末端には活性
ジスルフイド結合を含む反応性基を有する、反応
性に富んだ重合体の製造法に関する。そして本発
明の目的とするところは、腫瘍細胞等の標的物に
結合能を有する抗腫瘍抗体等と、制ガン剤等の細
胞毒物を結合して標的指向型制ガン剤(抗腫瘍
剤)等を製造するに際し、両者を有効かつ効率良
く結合させるために用いることができる重合体を
提供することになる。 b 従来技術 ある種の細胞だけを選択的に殺すことを目的と
して、その標的細胞と特異的に結合しうる免疫グ
ロブリンを種々の細胞毒性物質と結合させる試み
がなされてきた。例えば、免疫グロブリンにp−
ビス(2−クロロエチル)アミノ−L−フエニル
アラニン等を結合した複合体(特開昭51−61640
号)、免疫グロブリンにメトトレキセート等を結
合した複合体(特開昭56−65829号)、免疫グロブ
リンにクロラムブシル等を結合した複合体(特開
昭56−65828号)免疫グロブリンにマイトマイシ
ンーC等を結合した複合体(特開昭55−92325
号)、免疫グロブリンにダウノマイシンを結合し
た複合体(特開昭51−144723号)等が公知であ
る。 更に、特開昭51−126281号には、抗腫瘍免疫グ
ロブリンと、1分子当り制ガン剤を5〜500分子
共有結合している重合体担体(例えば、ポリダル
タミン酸)を、アミド結合によつて結合させて抗
腫瘍剤を得たことが開示されている。 これらの方法で得られた細胞毒性複合体は、腫
瘍細胞と選択的に結合し腫瘍細胞に毒性を発揮す
ることが期待されるものであり、非常に興味のあ
る薬剤である。しかしながら細胞毒性物質を直接
免疫グロブリンに結合する場合は、免疫グロブリ
ンに多数の細胞毒性物質を結合すると、免疫グロ
ブリンの抗原認識活性が低下してしまうので、か
かる困難を回避するためには、少数の細胞毒性物
質を結合するにとどめざるをえない。 一方、重合体を細胞毒性物質の担体として用い
る場合は、上記の難点を改善することができると
考えられる。しかし、特開昭51−126281号記載の
方法は、重合体担体に多数の細胞毒性物質を結合
する反応と、重合体−制ガン剤結合体に免疫グロ
ブリンを結合する反応が同一な反応のため、多数
の免疫グロブリンが重合体担体に結合してしま
い、そのため得られる複合体が均一なものとなり
得ないのみならず、治療剤として用いるのが不適
当な高分子量物質も含む、といつた問題を生じる
のである。 c 発明の目的 本発明者等は、かかる先行技術の欠点を解決す
べく鋭意研究を行なつた結果、免疫グロブリンと
の結合反応に供する反応基を1コだけ含有し、か
つ、それとは異なる、細胞毒性物質を結合するた
めの、反応基を多数含む重合体担体を用意し、先
づ多数の反応基によつて多数の細胞毒性物質を該
重合体担体に結合した後に、免疫グロブリンとの
反応基によつて免疫グロブリンと結合する、とい
う手順を踏むことによれば、治療剤として用いる
のが不適当な高分子物質を含まず、かつ、多数の
細胞毒性物質を結合した免疫グロブリン−細胞毒
性物質複合体を製造し得ることを見い出した。本
発明は、かかる抗腫瘍剤あるいは又その他の標的
指向型薬剤を製造する際に最適に使用できる、細
胞毒性物質を結合した反応性重合体を提供するも
のである。 d 発明の構成 本発明は、下記式〔〕 〔式〔〕において、Sは硫黄原子を表わす。

ジチオスレイトール、2−メルカプトエタノール
がある。チオール化合物は、重合体中のジスルフ
イド結合に対し1〜100倍モル量用いられる。反
応温度は−5゜〜70℃、反応時間は5分〜10日間が

は、重合体との反応は通常、水溶液中で行なわれ

た、重合体の水溶液に活性ジスルフイド化合物又
はそのアセトン溶液又はジオキサン溶液等を添加
混合した反応系で行なうこともできる。反応温度
は−5゜〜70℃、反応時間は1分〜24時間が適当で
ある。反応終了後、反応液をゲル過または透析
などに付すことにより式〔−1〕で表わされる
細胞毒性物質を結合した反応性重合体を精取す
る。必要なら凍結乾燥品として得ることができ
る。 本発明の原料物質である、式〔〕で表わされ
る末端の活性基を有している反応性重合体は、例
えばL−グルタミン酸、L−アスパラギン酸等の
重合体又は、それらと他のアミノ酸との間の共重
合体のアミノ末端(N−末端)に、チオール基又
は、保護チオール基導入剤を反応させ、しかる後
に、直接又は保護基をはずして得られたチオール
含有重合体に、活性ジスルフイド化合物を反応さ
せることにより、容易に得られるものである。チ
オール基導入剤として好的に用いることができる
物質としては、例えばN−サクシンイミジル3−
(2−ピリジルジチオ)プロピオネート

詳述する。 e 発明の効果 本発明で得られる細胞毒性物質を結合した反応
性重合体は、分子中に反応性に富んだチオール基
又は活性ジスルフイド結合を有しているので、か
かる基又は結合の反応性を利用して抗腫瘍免疫グ
ロブリン又はそのフラグメントと直接又は適当な
架橋剤を用いて結合させることができる。かくし
て得られる抗体一細胞毒性物質結合体は腫瘍細胞
等に選択毒性を発揮することが期待されるもので
ある。 以下、参考例および実施例により本発明を詳述
する。 参考例 1 N−末端に反応性基を含有するポリ−L−グル
タミン酸(ナトリウム塩)(3〜)の製造。 1−1 N−末端にジスルフイド結合を導入した
重合体の製造。 ポリ−L−グルタミン酸のナトリウム塩(平均
分子量21000)226.5mgの0.1Mリン酸ナトリウム
緩衝液(PH7.5)(15ml)中溶液に、N−サクシイ
ミジル3−(2−ピリジルジチオ)プロピオネー
ト(以下SPDPと省略する)68.8mgのエタノール
(6ml)溶液を撹拌下、2度に分けて加え、1.5時
間室温下り反応させた。反応液をセロフアン透析
チユーブに入れ、0.01Mリン酸緩衝液(PH7.5)
に対して4゜で2日間透析して低分子物質を除去す
ると、ポリ−L−グルタミン酸分子のアミノ末端
に3−(2−ピリジルジチオ)プロピオニル基が
アミド結合により導入されて生成したジスルフイ
ド結合を有する重合体を含有する溶液50mlが得ら
れた。 1−2 ジスルフイド結合の導入率 参考例1−4)の結果より、生成した重合性に
含有されかつ単離できるジスルフイド基導入重合
体量は35.4mg(15.6重量%)である。 1−3 N−末端にチオール基を有する反応性重
合体(2〜)の製造。 1−1)によつて得られた、アミノ末端に3−
(2−ピリジルジチオ)プロピオニル基を導入し
た、ジスルフイド結合を有する重合体を含有する
溶液50mlに、ジチオスレイトール17mgの0.1Mリ
ン酸ナトリウム(PH0.6)緩衝液2.0ml中溶液を加
え、室温下に80分間反応させた。次いで塩酸酸性
とし、生じた沈澱を遠心分離した。得られたポリ
−L−グルタミン酸の、沈澱物(末端にチオール
基を有するポリ−L−グルタミン酸と有しないポ
リ−L−グルタミン酸を含む。)は、0.01NHCl
で洗浄した。 一方、チオプロピルセフアロース6Bゲル
(Thiopropyl Sepharse6B、フアルマシア社
製)15mlを0.1Mリン酸ナトリウム(PH6.0)−
1mMエチレンジアミンテトラ酢酸(以後EDTA
と省略する)(PH6.0)緩衝液40mlに分散し、得ら
れた分散溶液に、前記で得られたポリ−L−グル
タミン酸を同一緩衝液10mlに溶解して得られる溶
液を加え、室温下、窒素雰囲気中でゆるやかに1
夜撹拌した。これで末端にチオール基を有するポ
リ−L−グルタミン酸が樹脂に結合する。次いで
樹脂をロ別し、0.01Mリン酸緩衝液PH7.5で充分
洗浄した。 かくして得られた樹脂を0.1Mトリスー塩酸−
1mMEDEA(PH8.5)緩衝液50mlに分散し、2−
メルカプトエタノール1.17gを加え、室温下窒素
雰囲気中で10時間ゆるやかに撹拌した。これで末
端がチオール基のポリ−L−グルタミン酸(2〜)
が樹脂から遊離する。 樹脂を別し、0.01Mトリス−塩酸−
0.1mMEDTA(PH8.5)緩衝液でよく洗浄し、次い
で数を氷冷下塩酸でPH1.8とし、生じた末端に
チオール基を有するポリ−L−グルタミン酸
(2〜)の沈澱を遠心分離した。生成物の末端基量
は参考例1−4)、1−5)の結果より
1.984μmoleであつた。 1−4 N−末端に活性ジスルフイド基を有する
反応性重合体(3〜)の製造 1−3)で得られた末端にチオール基を有する
ポリ−L−グルタミン酸の沈澱を、再び0.4Mリ
ン酸ナトリウム−1mMEDTA(PH7.5)緩衝液1
mlに溶解し、得られた溶液を2−ピリジルジスル
フイド(以下2−PDSと省略する)23mgのエタ
ノール(4ml)溶液を0.1Mリン酸ナトリウム−
1mMEDTA(PH6.0)10mlに加えて得られた溶液
に加え、室温下で30分間反応させた後、(ポリ−
L−グルタミン酸の末端が活性ジスルフイド基と
なる)反応液をセロフアンチユーブに入れ、
0.01Mリン酸ナトリウム(PH7.5)に緩衝液に対
して6時間、純水に対して1日透析した。回収液
を減圧で30mlに減少し、凍結乾燥すると、末端に
2−ピリジルジチオ基が導入されたポリ−L−グ
ルタミン酸(ナトリウム塩)(3〜)の綿状固体
35.4mgが得られた(原料である(1)よりの重量収率
15.6%)。末端基量は1−5)の結果より
1.984μmoleであつた。従つて平均分子量は35.4÷
1.984×103=1.78×104、平均ユニツト数は1.74×
104/151=118個と計算される。 1−5 (3〜)の重合体末端2−ピリジルジチオ
基定量 一定量のサンプルを精秤し(1.895mg)3.00ml
の0.1Mリン酸ナトリウム緩衝液(PH7.2)に溶解
し、ジチオスレイトールの小片を加え、遊離した
2−メルカプトピリジンに由来する吸収
(343nm、分子吸光係数ε=8080)を測定した
(A=0.286)。精秤されたサンプル中の末端基量
は0.1062μmole、従つて、同サンプル中の末端活
性ジスルフイド含有ポリ−L−グルタミン酸分子
の分子量は17.800、又同分子中のグルタミン酸
(ナトリウム塩)のユニツト数は、1ユニツトの
質量数が151であることより、118と計算された。
従つて参考例1−3)、1−4)で得られた末端
チオール型及び末端2−ピリジルジチオ型重合体
に含有される末端基量は、35.4/1.895×0.1062= 1.984μmoleである。 実施例 1 側鎖にダウノマイシンを結合し、末端に反応性
基を含有する重合体の製造。 1−1 側鎖にダウノマイシンを結合し、N−末
端に2−ピリジルジチオ基を含有するポリ−L
−グルタミン酸(ナトリウム塩(4〜)の製造。 参考例1−3)で得られたN−末端に2−ピリ
ジルジチオ基を含有するポリ−L−グルタミン酸
(ナトリウム塩)(分子量17800)(3〜)25mg
(1.40μmole)を5%食塩水6mlに溶解し、これ
にダウノマイシン塩酸塩11.2mgと、1−エチル−
3−(3−ジメチルアミノプロピル)カルボジイ
ミド塩酸塩(以下、EDCI・HClと省略する)
38.1mgを加え、室温下にて一夜反応させた。反応
液をセロフアンチユーブに入れ、水に対して4゜で
充分透析して低分子物質を除去した。透析内液を
減圧で10.5mlに濃縮すると、目的物である末端に
2−ピリジルジオ基を含有し、側鎖にダウノマイ
シンを結合したポリ−L−グルタミン酸(ナトリ
ウム塩)(4〜)の水溶液が得られた。 生成物は490nmにダウノマイシンに由来する吸
収極大を示したことより、側鎖にダウノマイシン
を結合していることが明らかとなつた。490nmの
吸光度の測定より重合体に結合したダウノマイシ
ンの量は1.16×10-5moleであつた。又、1−2)
の結果より、重合体に含有される2−ピリジルジ
チオ基の量は1.36×10-6moleである。従つて重合
体1分子に結合したダウノマイシン分子の数は、
1.16×10-5mole/1.36×10-6mole=8.53個と計算
された。 1−2 ダウノマイシンを側鎖に結合し、N−末
端にチオール基を含有するポリ−L−グルタミ
ン酸(ナトリウム塩)(5〜)の製造。 1−1)で得られた側鎖にダウノマイシンを結
合し末端に2−ピリジルジチオ基を含有するポリ
−L−グルタミン酸(4〜)の溶液10.5mlに、
0.1Mリン酸緩衝液(PH7.5)中1mMジチオスレイ
トール溶液2.0mlを添加し、室温下に30分間反応
せしめた。遊離した2−メルカプトピリジンに由
来する343nmにおける吸光度増加量を測定して求
めた末端基量は、1.36×10-6moleであつた。 反応液を純水に対し4゜で充分透析することによ
り低分子物質を除くと、側鎖にダウノマイシンを
結合し、末端がチオール基に変換したポリ−L−
グルタミン酸(5〜)の水溶液が得られた。 1−3 ダウノマイシンを側鎖に結合し、N−末
端に3−カルボキシ−4−ニトロフエニルジチ
オ基を含有するポリ−L−グルタミン酸(6〜)
の製造。 1−2)で得られた側鎖にダウノマイシンを結
合し、末端にチオール基を含有するポリ−L−グ
ルタミン酸(5〜)(末端基量0.27×10-6mole、グ
ウノマイシン量2.5×10-6mole)の水溶液5mlに、
0.1Mリン酸緩衝液(PH7.6)中1mMの5,5′−ジ
チオビス(2−ニトロ安息香酸)溶液3.0mlを加
えた。室温下15分反応した後、反応液を4゜で純水
に対して充分透析すると、目的物であるダウノマ
イシンを側鎖に結合し末端に3−カルボキシ−4
−ニトロフエニルジチオ基を含有する、ポリ−L
−グルタミン酸(6〜)の水溶液12mlが得られた。 水溶液の一部にジチオスレイトールを加える
と、遊離した5−メルカプト−2−ニトロ安息香
酸にもとずき、412nmにおいて吸光度の増大が生
じたことより(6〜)の末端構造を確認した。 実施例 2 側鎖にマイトマイシンCを結合し、末端に反応
性基を含有する重合体の製造。 2−1 側鎖にマイトマイシンCを結合し、N−
末端に2−ピリジルジチオ基を含有する重合体
(8〜)の製造 N−末端に2−ピリジルジチオ基を含有するポ
リ−L−グルタミン酸(ナトリウム塩)(分子量
16500)(7〜)20mg(1.212μmole)を5%食塩水
5mlに溶解し、これにマイトマイシンC9.0mgと
EDCI・Cl25mgを加え、室温下に1夜反応させた。
反応液をセロフアンチユーブ中水に対して4゜で充
分透析して低分子物質を除き、目的物である末端
に2−ピリジルチオ基を含有し、側鎖にマイトマ
イシンCを結合したポリ−L−グルタミン酸(ナ
トリウム塩)(8〜)の水溶液8.5mlを得た。実施例
2−2)の結果より、得られた重合体は末端基量
で1.1μmoleであり、9.8μmoleのマイトマイシン
Cを結合していることが判明した。 生成物は360〜nmにマイトマイシンCに由来す
る吸収を示したことにより、ポリ−L−グルタミ
ン酸の側鎖にマイトマイシンCが結合しているこ
とが明らかになつた。360nmの吸光度の測定よ
り、重合体は9.8×10-6moleのマイトマイシンC
を結合していることが判明した。又、2−2)の
結果より、重合体に含有される2−ピリジルジチ
オ基の量は1.1×10-6moleである。従つて重合体
1分子に結合したマイトマイシンCの数は、9.8
×10-6/1.1×10-6=9.0個と計算された。 2−2 側鎖にマイトマイシンCを結合し、N−
末端にチオール基を含有するポリ−L−グルタ
ミン酸(ナトリウム塩)(9〜)の製造。 実施例2−1)で得た、側鎖にマイトマイシン
Cを結合し、末端に2−ピリジルジチオ基を含有
するポリ−L−グルタミン酸(8〜)の溶酸8.3ml
に、0.1Mリン酸緩衝液(PH7.6)中20mM2−メル
カプトエタノール溶液1.5mlを添加し、室温下に
30分間反応せしめた。遊離した2−メルカプトピ
リジンに由来する343nmにおける吸光度増加量よ
り、末端基量を求めると1.10×10-6moleであつ
た。反応液を純水に対し4゜で充分透析して、低分
子物質を除くことにより精製し、マイトマイシン
Cを側鎖に結合し、末端にチオール基を含有する
ポリ−L−グルタミン酸(9〜)の水溶液を得た。 2−3 側鎖にマイトマイシンCを結合し、N−
末端に5−ニトロ−2−ピリジルジチオ基を含
有するポリ−L−グルタミン酸(ナトリウム
塩)(10〜)の製造。 実施例2−2)で得られた、側鎖にマイトマイ
シンCを結合し、末端にチオール基を含有するポ
リ−L−グルタミン酸(9〜)(末端基量
0.1μmole、マイトマイシンC量8.9μmole)の水
溶液2mlに、2,2′−ジチオビス(5−ニトロピ
リジン)の0.1Mリン酸緩衝液(PH7.5)中、
10mM溶液0.5mlを加え、室温下30分間反応せし
めた。反応液を4℃にて純水に対して充分透析
し、目的物である側鎖にマイトマイシンCを結合
し、末端に5−ニトロ−2−ピリジルジチオ基を
含有するポリ−L−グルタミン酸(ナトリウム
塩)(10〜)の水溶液3.2mlを得た。生成物の一部
にジチオスレイトールを加えると、遊離した5−
ニトロ−2−メルカプトピリジンに由来する
386nmにおける吸光度の増大が生じたことによ
り、(10〜)の末端構造を確認した。 実施例 3 側鎖にメルフアランを結合し、末端に反応性基
を含有する重合体の製造。 3−1 側鎖にメルフアランを結合し、N−末端
に2−ピリジルジチオ基を含有するL−グルタ
ミン酸とL−アラニンの共重合体(ナトリウム
塩)(12〜)の製造 N−末端に2−ピリジルジチオ基を含有する、
L−グルタミン酸とL−アラニンの共重合体(ナ
トリウム塩)(構成当量比30.3:1)(分子量
13900)(11〜)10mgを3mlの水に溶解し、4゜にて
EDCI・HCl12.5mgを加え、2分間撹拌したのち、
p−〔ビス(2−クロロエチル)アミノ〕L−フ
エニルアラニン(メルフアラン)10mgを3mlに溶
解して得られた水溶液を滴下し、反応液を4゜にて
12時間撹拌した。反応液をセロフアンチユーブに
入れ、水に対して4゜で充分透析して低分子物質を
除くことによつて精製し、目的物である末端に2
−ピリジルジチオ基を含有し、側鎖にメルフアラ
ンを結したL−グルタミン酸とL−アラニンの共
重合体(ナトリウム塩)(12〜)の水溶液7.4mlが
得られた。 生成物は259nmと302nmにメルフアランに由来
する吸収極大を示したことより、側鎖にメルフア
ランを結合していることが明らかとなつた。
259nmの吸光度の側定より、重合体に結合したメ
ルフアラン量は、4.92×10-6moleであつた。又3
−2)の結果より、重合体に含有される2−ピリ
ジルジオ基の量は、0.70×10-6moleである。従つ
て、重合体1分子に結合したメルフアラン分子の
数は、4.92×10-6/0.70×10-6=7.0と計算され
た。 3−2 側鎖にメルフアランを結合し、N−末端
にチオール基を含有するL−グルタミン酸とL
−アラニンの共重合体(ナトリウム塩)(13〜)
の製造。 実施例3−1)で得られた、側鎖にメルフアラ
ンを結合し、N−末端に2−ピリジルジチオ基を
含有するL−グルタミン酸とL−アラニンの共重
合体(ナトリウム塩)(12〜)の溶液7.4mlに、
0.1Mリン酸緩衝液(PH7.5)中1mMジチオスレイ
トール溶液2.0mlを添加し、室温下に30分間反応
せしめた。遊離した2−メルカプトピリジンに由
来する343nmにおける吸光度の増加量より末端基
量を定量した結果、0.70×10-6moleであつた。 反応液を純水に対し4゜で透析することにより生
成物を精製し、目的物である側鎖にメルフアラン
を結合し、N−末端にチオール基を含有する共重
合体(ナトリウム塩)(13〜)の水溶液を得た。 3−3 メルフアランを側鎖に結合し、N−末端
に3−カルボキシ−4−ニトロフエニルジチオ
基を含有するL−グルタミン酸とL−アラニン
の共重合体(ナトリウム塩)(14〜)の製造。 3−2)で得られた、側鎖にメルフアランを結
合し、末端にチオール基を含有するL−グルタミ
ン酸とL−アラニンの共重合体(ナトリウム塩)
(13〜)(末端基量0.7μmole、メルフアラン量
4.92μmole)11.7mlに、0.1Mリン酸緩衝液(PH
7.6)中5mM5,5′−ジチオビス(2−ニトロ安息
香酸)溶液2mlを加え、室温下に15分間反応させ
た。反応液を4゜で純水に対して充分透析すると、
目的物であるメルフアランを側鎖に結合し、末端
に3−カルボキシ−4−ニトロフエニルジチオ基
を含有するL−グルタミン酸とL−アラニンの共
重合体(ナトリウム塩)(14〜)の水溶液16.3ml
が得られた。生成物の一部にジチオスレイトール
を反応せしめたところ、遊離ある5−メルカプト
−2−ニトロ安息香酸に由来する412nmにおける
吸光度の増大があつたことから、末端基の構造を
確認した。 実施例 4 側鎖にアラーCを結合し、末端に反応性基を含
有する重合体の製造。 4−1 側鎖にアラーC誘導体を結合し、N末端
に4−ニトロ−3−カルボキシフエニルジチオ
基を含有するポリ−L−グルタミン酸(ナトリ
ウム塩)(16〜)の製造。 N−末端に4−ニトロ−3−カルボキシフエニ
ルジチオ基を含有するポリ−L−グルタミン酸
(ナトリウム塩)(分子量13100)(15〜)10mgを、
0.1Mリン酸緩衝液(PH7.5)3mlに溶解し、これ
に5′−(2−アミノエチルホスホリル)−1−(β
−D−アラビノフラノシル)シトシン(アラーC
誘導体)47mgと、EDCI・HCl50mgを加えて溶解
し、12時間撹拌して反応せしめた。反応液をセロ
フアンチユーブに入れ、水に対して4゜で充分透析
して、低分子物質を除くことによつて精製すると
目的物質である側鎖にアラーC誘導体を結合し、
末端に4−ニトロ−3−カルボキシ−フエニルジ
チオ基を含有したポリ−L−グルタミン酸(ナト
リウム塩)(16〜)の水溶液4.7mlが得られた。 生成物は272nmにアラーCに由来する吸収極大
を示したことより、側鎖にアラーC誘導体を結合
していることを確認した。272nmの吸光度の測定
より、結合したアラーC誘導体の量は15.9×
10-6moleであつた。又、4−2)の結果より、
重合体に含有される4−ニトロ−3−カルボキシ
フエニルジチオ基の量は、0.68×10-6moleであ
る。従つて、重合体1分子に結合したアラーC誘
導体の分子の数は15.9×10-6/0.68×10-6=23.4
個と計算された。 4−2 側鎖にアラーC誘導体を結合し、N−末
端にチオール基を含有するポリ−L−グルタミ
ン酸(ナトリウム塩)(16〜)の製造。 4−1)で得られた、側鎖にアラーC誘導体を
結合し、N−末端に4−ニトロ−3−カルボキシ
フエニルジチオ基を含有するポリ−L−グルタミ
ン酸(16〜)の溶液4.7mlに0.1Mリン酸緩衝液中
1mMジチオスレイトール溶液1.0mlを添加し、室
温下に30分間反応せしめた。遊離した5−メルカ
プト−2−ニトロ安息香酸に由来する412nmにお
ける吸光度の増大量を測定することにより、末端
基量を求めた結果、0.68×10-6moleであつた。反
応液を純水に対して、4゜で充分透析することによ
り、低分子物質を除き、側鎖にアラーC誘導体を
結合し、末端がチオール基に変換したポリ−L−
グルタミン酸(17〜)の水溶液を得た。 4−3 アラーC誘導体を側鎖に結合し、N−末
端に2−ピリジルジチオ基を含有するポリ−L
−グルタミン酸(18〜)の製造。 4−2)で得られた、側鎖にアラーC誘導体を
