The medium was replaced with 200 μL of Opti-MEM (registered trademark) the next day for a 24-well plate seeded with 0.9 × 10 3 TOV112D cells (human ovarian cancer cell line). Next, 100 μL of the liposome dispersion prepared in Examples 4 to 6 diluted with Opti-MEM (registered trademark) so as to be 3 times the treatment concentration was added to the 24-well plate (total volume 300 μl). Thereafter, the cells were cultured for 24 to 48 hours in a 5% CO 2 incubator.

(3)定量PCR反応
逆転写反応とPCR反応はQUANTIFAST PROBE RTPCR KIT(QIAGEN社、登録商標)を用いて行った。用いたsiRNA遺伝子に対するプライマー/プローブはTaqMan Gene expression assay(ABI、登録商標)を使用し、Mx3000P(アジレント・テクノロジー株式会社、登録商標)を用いて定量PCRを実施した。PCRのコンディションは50℃ 30分;95℃ 15分;94℃ 15秒;60℃ 30秒 (40 サイクル)とした。内部標準はTaqMan Encogeneous Control Human ACTB(ABI、登録商標)を使用した。得られたデータは△△CT法を用いトランスフェクション未処理に対する相対定量でmRNA残存率として算出した。
(3) Quantitative PCR reaction Reverse transcription reaction and PCR reaction were performed using QUANTIFAST PROBE RTPCR KIT (QIAGEN, registered trademark). 上記の結果から、実施例4〜6に示す本発明の化合物と核酸を含むリポソーム分散液は、いずれも高いmRNAの産生抑制効果を示し、細胞外(血中)で核酸分子を安定に保持することができ、速やかに細胞質中で核酸を放出して標的細胞内で効率よく核酸本来の機能を発揮させることができることがわかった。すなわち、本発明の化合物を含むリポソームは、核酸キャリアとして有用である。   From the above results, the liposome dispersions containing the compounds of the present invention and nucleic acids shown in Examples 4 to 6 all show a high mRNA production inhibitory effect and stably hold nucleic acid molecules outside (in the blood).
