変異p85β遺伝子陽性のトランスジェニックF0マウスの雄より手術によって精子を取り出し、正常なC57BL/6マウスの雌の卵子と人工授精を行ってF1マウスを作製した。方法としては、正常なC57BL/6マウスの雌30匹より630個の卵子を得、CO2インキュベーターで培養した。人工授精を行って得られた80個の2Cellを4匹のレシピエントマウスに移植した。自然分娩及び帝王切開によって48匹のトランスジェニックF1マウスを得ることができた。得られた8匹のマウスの尾よりDNAを抽出し、PCR法によって導入遺伝子の検出を行った。その結果、雌24匹の内15匹が陽性を示し、雄24匹中11匹が陽性であった。

また図4に示すように、変異p85β遺伝子が発現したマウス(P85βM−tg(+))は他の2群(WT及びP85βM−tg(−))と比較して有意に寿命が延長した。
図5に示すように、変異p85β遺伝子が発現したマウス(太線)は正常マウス(細線)と比較して有意に生存率が高かった。
