試料中のGAPDH mRNAの測定方法において、GAPDH mRNA内の特定塩基配列の一部と相同的な配列を有する第一のプライマー及び特定塩基配列の一部と相補的な配列を有する第二のプライマーを用いた、下記の工程からなる測定方法であり、ここで第一又は第二のプライマーのいずれか一方はその5'末端にRNAポリメラーゼのプロモーター配列が付加されたプライマーであり、また(i)前記第一のプライマーが配列番号12又は配列番号13で示された塩基配列からなるオリゴヌクレオチドであり、前記第二のプライマーが配列番号25で示された塩基配列からなるオリゴヌクレオチドである測定方法。
(1)RNAを鋳型とする、RNA依存性DNAポリメラーゼ活性を有する酵素による特定塩基配列に相補的なcDNAを合成する工程、
(2)リボヌクレアーゼH(RNase H)活性を有する酵素によるRNA−DNA2本鎖のRNAを分解する工程(1本鎖DNAの生成)、
(3)1本鎖DNAを鋳型とする、DNA依存性DNAポリメラーゼ活性を有する酵素による、特定塩基配列又は特定塩基配列に相補的な配列のRNAを転写可能なプロモーター配列を有する2本鎖DNAを生成する工程、
(4)RNAポリメラーゼ活性を有する酵素による前記2本鎖DNAを鋳型とするRNA転写産物を生成する工程、
(5)該RNA転写産物が、前記(1)の反応におけるcDNA合成の鋳型となることで、連鎖的にRNA転写産物を生成する工程、
(6)前記RNA転写産物量を測定する工程
