Description
1.1.微生物
本発明は、キチンと、グルコース、マンノース及び/又はガラクトースを含むポリマー、また、それらの誘導体の、ピキア・パストリス野生型株、その変種及び突然変異体の発酵による生産に関する。
1.2.発酵培地
ピキア・パストリス発酵は、炭素源、窒素源及び無機塩を含む液体培地で行われる。
1.3.発酵条件
本発明は、ピキア・パストリス発酵を用いた、細胞壁多糖の生産のためのあらゆる手段又は手順に関する。多糖は例えば、キチン、グルコース、マンノース及び/又はガラクトースを含むポリマー又はその誘導体産物である。より具体的には、本発明は、P.パストリスの生育が高い細胞密度と微生物の細胞壁中の高いキチン/グルカン含量に達するのに有利に働くいくつかの方法を記述する。
2.発酵産物の抽出と精製
本発明に記載された多糖を抽出し、精製するために使用される手順は以下のとおりである。
表1に記載の組成の14.25Lの発酵培地に、750mLのピキア・パストリスDSMZ70877の培養液を接種した。発酵は、パイロットスケールのバイオリアクター(LP351、50L、BioEngineering, スイス)中で、制御された温度とpH(それぞれ30℃及び5.0)で行われた。空気の流速は、全稼働中、30L/分に保たれ、圧力は0.10バール(bar)に維持された。

最高撹拌速度(1000rpm)に達すると(約44時間の発酵)、pO2は基質の制限により20%に調節された。供給溶液の添加速度は、pO2が20%以下に落ちないように連続的に調節された。
実施例1に記載の手順がピキア・パストリスDSMZ70877を増殖させるために使用された。純粋なグリセロールが、86%グリセロールを含む、バイオディーゼル産業由来のグリセロール副産物によって置き換えられた。供給溶液の流速は、新しい基質組成を考慮して修正された。両方の培養の間、細胞の乾燥重量の漸進的変化を、図1に表わしている。
Claims (10)
- キチン及びグルカンを含むキチン‐グルカン複合体及び、
グルコース、マンノース、及びガラクトースのうち少なくともいずれか1つを含むバイオポリマーの生産方法であって、
以下の工程を含む:
窒素源として、アンモニア、水酸化アンモニウム、アンモニウム塩、及び有機窒素化合物から選択される少なくとも1つを含み、
さらに、炭素源として、アルコール、糖、有機酸、ポリオール、脂肪酸、及びアミノ酸のうち少なくともいずれか1つを含む、食品、産業廃棄物、又はバイオディーゼル産業により生じたメタノール豊富な副産物、グリセロール豊富な副産物、又はグリセロール−メタノール混合物である副産物を含み、
pH5.0とpH7.0の間で発酵中のpHを調整するアルカリ、アンモニア、及びアンモニウム塩を含む培養液中で、
ピキア・パストリスを発酵させる工程を含むことを特徴とする方法。 - 請求項1記載の方法であって、
培養液中の炭素源は、バイオディーゼル産業により生じたアルコール、糖、有機酸、ポリオール、脂肪酸、及びアミノ酸のうち少なくともいずれか1つを含む副産物であることを特徴とする方法。 - 請求項1又は2記載の方法であって、
バイオディーゼル産業により生じた副産物が、少なくとも86%グリセロール、又は
100%メタノールであることを特徴とする方法。 - 請求項1〜3いずれか1項記載の方法であって、
バイオディーゼル産業により生じた副産物が、少なくとも99%グリセロールであることを特徴とする方法。 - 請求項1〜4いずれか1項記載の方法であって、
発酵中の温度が20〜40℃に調節されていることを特徴とする方法。 - 請求項1〜5いずれか1項記載の方法であって、
発酵中の温度が30℃に調節されていることを特徴とする方法。 - 請求項1記載の方法であって、
発酵中のpHは、アルカリ、アンモニア、又はアンモニウム塩を培養液に添加することによって調節されていることを特徴とする方法。 - 請求項1記載の方法であって、
発酵中のpHがpH5.0に調節されることを特徴とする方法。 - 請求項1〜8いずれか1項記載の方法であって、
発酵中に得られるバイオポリマーが、キチンーグルカン複合体に加えて、グルコース、マンノース、ガラクトースのうち少なくともいずれか1つを含む水溶性の多糖を含むことを特徴とする方法。 - 請求項1〜9いずれか1項記載の方法であって、
発酵中に得られたバイオポリマーが、グルコース、マンノース、ガラクトースのうち少なくともいずれか1つを含む多糖を細胞の乾燥重量の最大45%含むことを特徴とする方法。
