車エビ(Penaeus monodon)を30℃±2℃で0〜5時間曝露し、毎時間試料を採集した。車エビの殻、頭部、及び尾を取り除き、車エビの胴体領域約10gを0.1M、pH7.4のリン酸緩衝液100mlとブレンドした。次いで、いかなる予備処理又は抽出ステップもなしで、本開発のバイオセンサーを使用して試料を分析した。0.35ボルトで操作されたGPESソフトウエアを備えたAutolab PGSTAT 12 Potentiostat/Galvanostatを使用して測定を実施した。0.1M、pH7.4のリン酸緩衝液を含むビーカー内で試料を試験した。
小型化バイオセンサーの構築では厚膜技術を適用した。というのは、この技術によって、固体状態の機械的に強固な平面センサーを構築することが可能になるからである。これは、スクリーン印刷プロセスによって基板上に厚膜を順次堆積させて実施される。バイオセンサーの構造は、絶縁性担体又は基板上に順次堆積したペースト層を備える。本発明の小型化バイオセンサーを、半自動化DEK−J202RS厚膜プリンタを使用する多段スクリーン印刷法によって製造した。モノフィラメントポリエステル(SEFAR(登録商標)PET 1000)ステンシル(規格90−48W)の高弾性率メッシュを、スクリーンメッシュに固定された正方形パッドに平行なストレートで短く広い導電体として設計した。これによって、円形パッドを有する曲がった狭い導電体の設計に比較してより良好な導電率がもたらされる。印刷ペーストに対するエマルジョン厚さが12μm±2μmで、ステンレス鋼スクリーンメッシュ(地金厚78μm)を45°でプリントストロークに載せた。3つの電極をポリエステル基板上にスクリーン印刷した(50×60mm)。印刷プロセスの前に、ポリエステルシートをオーブンで130℃5時間焼くことによって、次の加熱ステップ中にホイルが収縮するのを防止した。各印刷サイクルによって、単一片のポリエステル基板上に、カーボンペースト(Screen Technology、BBI 440)作用電極及び対電極、並びに塩化銀(AgCl)ペースト(Dupont、B166)参照電極を備える3つの小型化スクリーン印刷電極が製造された。銀層(Dupont、B111)を基底トラック層として印刷することによって基板状のペーストの導電率及び接着性を増進させた。各層を印刷した後、ペーストをオーブンで110℃10分間乾燥することによって溶媒を揮発させた。
Claims (13)
- スクリーン印刷作用電極と、対電極と、参照電極とを備える、海産食品又は魚組織中のヒスタミンを測定するための電流測定バイオセンサーであって、ジアミンオキシダーゼが滴下コーティングを介してスクリーン印刷作用電極の表面上にポリ(2−ヒドロキシエチルメタクリレート)(光HEMA)とともに固定化され、かつ前記バイオセンサーが、0.30ボルト〜0.50ボルトの範囲である低動作電圧下で海産食品又は魚組織中のヒスタミンのレベルを測定できることを特徴とする電流測定バイオセンサー。
- スクリーン印刷作用電極がカーボンペースト系である、請求項1に記載のバイオセンサー。
- 対電極が白金棒である、請求項1又は2に記載のバイオセンサー。
- 参照電極が銀/塩化銀(Ag/AgCl)電極である、請求項1〜3のいずれか一項に記載のバイオセンサー。
- ジアミンオキシダーゼが、
a)比1:4に従ってジアミンオキシダーゼ溶液をポリ(2−ヒドロキシエチルメタクリレート)(光HEMA)と混合するステップと、
b)スクリーン印刷作用電極の表面上にこの混合物を滴下コーティングするステップと、
c)電極をUV曝露ユニット内で窒素ガス流下300秒間光硬化させるステップと
によって、固定化される、請求項1〜4のいずれか一項に記載のバイオセンサー。 - 電極すべてが基板上にスクリーン印刷され、ジアミンオキシダーゼが滴下コーティングを介してポリ(2−ヒドロキシエチルメタクリレート)(光HEMA)とともに固定化され、ヘキサシアノ鉄(III)酸カリウムが、スクリーン印刷作用電極の表面上に電着されることを特徴とする、請求項1〜5のいずれか一項に記載のバイオセンサー。
- スクリーン印刷作用電極及び対電極がカーボンペースト系である、請求項6に記載のバイオセンサー。
- 電極が、ポリエステル基板上にスクリーン印刷される、請求項6又は7に記載のバイオセンサー。
- バイオセンサーが、基底トラック層として印刷された銀層をさらに備える、請求項6〜8のいずれか一項に記載のバイオセンサー。
- スクリーン印刷作用電極を、脱イオン水に溶解した0.1Mヘキサシアノ鉄(III)酸カリウム溶液中で、少なくとも15回0.2vs −1 で、撹拌しながらサイクルさせるステップを含む、サイクリックボルタンメトリ法によって、ヘキサシアノ鉄(III)酸カリウムが電着される、請求項6〜9のいずれか一項に記載のバイオセンサー。
- a)ヒスタミンを含む疑いのある試料を請求項1〜10のいずれか一項に記載のバイオセンサーにかけるステップと、
b)0.30ボルト〜0.50ボルトの範囲である電圧で、6.4〜8.4の範囲であるpHで電流を測定することによってヒスタミンの存在を示す出力信号を提供するステップと
を含む、海産食品又は魚組織中のヒスタミンを測定するための方法。 - 電圧が、0.35ボルトである、請求項11に記載の方法。
