【発明の属する技術分野】
本発明は、毛包再構成系を担持する実験動物、ならびにその作出のための細胞系および使用に関する。

【従来の技術】
特定の目的に向けられた動物反応を安定に再現することのできる多種多様のキメラ動物またはトランスジェニック動物は、主として、特定の疾患のモデルとして、または特定の疾患の病因を解明するために開発されている。例えば、毛髪再構成アッセイ用として、毛芽(特殊なケラチノサイト集塊として新生仔マウスの皮膚に存在する毛包前駆細胞)が、毛髪誘発性の毛乳頭細胞(ラットのヒゲ由来)と一緒に移植されたヌードマウスが開発されている(Lichti et al,J.Invest Dermatol 101:124−129,1993)。

また、新生仔マウスの毛芽に由来する培養ケラチノサイトと毛乳頭細胞とが移植されたマウスでは、その移植領域の組織学的な解析により完全に組織化された上皮と毛包の再構成が確認されている(Kamimura et al,J.Invest Dermatol 109:534−540,1997)。かような再構成毛包はin vivoでの毛髪の誘発についての信頼のおける機能性のアッセイに使用されうることが示唆されている。

他方、毛髪の誘発と相俟って言及することのできる事象として毛の色調を挙げることができるであろう。毛の色調は多くの場合、毛母細胞(hair matrix)の近傍に存在するメラノサイトによって産生され、毛を形成する細胞に与えられるメラニンによりもたらされるものとみなされている。白髪になるのは、かようなメラノサイトがメラニンを産生しなくなるか、メラニンを産生しても極めて少量であるか、またはメラノサイトが減少ないしは消滅することが主要な原因であるといわれている。しかし、毛周期に関連して考慮する場合、退行期(毛の増殖が止まり、毛包の短縮および棍毛の形成)および休止期を通じて毛包から激減または消失したメラノサイトは、次期成長期においてどのように補充され、そして毛の色調を整えることができるのか(どこから来るのか)、等については明らかでない。

【発明が解決しようとする課題】
したがって、毛の色調を整えることのできる手段、換言すれば、ある一定の手段が白髪の防止もしくは抗白髪作用を奏するか否かを評価できるイン・ビボの評価系を提供することは価値があろう。こうして、本発明の目的は、かかる評価系を提供することにある。なお、本明細書にいう、「抗白髪性」または「抗白髪作用」とは、例えば、メラノサイトの増殖促進、遊走能力促進、分化能促進、生存能力の増進、メラニン産生能等からなる群より選ばれる一種以上の活性で評価できる白髪化を抑制しうる特性であり、毛の色によりそれらの活性が示される性質を意味する。

【課題を解決するための手段】
本発明者らは、上記目的を達成すべく検討してきたところ、上述の毛包の再構成が確認されている、所謂、再構成毛包が、同種または異種のいずれの外来のメラノサイトを加えても再構成でき、しかもこうして構成された毛包のメラノサイトは活性な状態に維持できることを見出した。

したがって、本発明によれば、毛乳頭細胞および表皮細胞と、これらの細胞に対して外来のメラノサイトとの組み合わせからなる毛包を再構成するための系が提供される。

別の態様の本発明として、レシピエント動物に上記系が移植され、こうして再構成毛包を担持するキメラ動物が提供される。

また、別の態様の発明として、上記キメラ動物を被験動物として用意し、該被験動物をある一定の手段で処置し、こうして処置された被験動物の再構成毛包におけるメラノサイトの活性をモニターし、未処置対象に対する該活性の変化の程度を該手段のメラノサイトの活性化能と関連付けることを特徴とする、ある一定の手段のメラノサイトの活性化能の評価方法も提供される。

【発明の具体的な態様】
本明細書において、「毛乳頭細胞(dermal papillae)」(以下、DPという場合あり)は、本発明の目的に沿う限り、広く毛包最下部にある毛乳頭細胞およびその周辺の細胞および組織を包含する概念を表わすものとして使用されている。このような組織としては毛乳頭細胞を含有する真皮を挙げることができる。限定されるものでないが、このような細胞は、マウスに由来する細胞を例にすれば、バーシカンプロモーターの下流に適当なレポーター遺伝子(例えば、LacZ遺伝子、グリーン蛍光タンパク質(GFP)の構造遺伝子)をつないだ発現ベクターを導入したトランスジェニックマウス(Ver−LacZ)から生まれた新生仔(生後4日以内に使用)から、レポーター遺伝子の発現を目印にして取得することができる。また、毛乳頭細胞を含有する真皮としては、皮膚から通常の調製方法によって取得される真皮が、一般に、毛乳頭細胞を含むので、そのまま、本発明の毛乳頭細胞として使用することもできる。

他方、「表皮細胞」は、皮膚の表皮または上皮の大部分を構成する細胞であり、真皮に接する1層の基底細胞から生じる。マウスを例にすると、かような表皮細胞としては新生仔(もしくは胎児)に由来する表皮細胞が好ましく使用できるが、ケラチノサイトの形態にある細胞の培養物であってもよい。かような細胞は、それ自体公知の方法により所望のドナー動物の皮膚から調製することができる。

上記の毛乳頭細胞または毛乳頭細胞を含む真皮細胞と表皮細胞は、レシピエント動物に移植可能であれば、それらのドナー動物の種を問うことなく使用できるが、レシピエント動物と同種の動物に由来するものが好ましい。限定されるものでないが、レシピエント動物がマウスである場合、上記両細胞はいずれもマウス由来であることができ、他方、両細胞が同じマウスの系統に由来するものでなくてもよい。したがって、毛乳頭細胞または毛乳頭細胞を含む真皮細胞を、その確認が容易な上述のいずれかのトランスジェニックマウス(例えば、Ver−LacZ系統)から取得し、他方、表皮細胞を、例えば、アルビノ(チロシナーゼの遺伝的欠陥を有する)の性質をもつICR系統およびメラノサイト欠損系統(例えば、Wsh/Wshマウス)からなる群より選ばれるマウスから取得したものを使用してもよい。本発明に従えば、かようなアルビノもしくはメラノサイト欠損の性質をもつ動物由来の表皮細胞の使用が、後述する外来のメラノサイトの挙動をより容易にモニターできるので好ましい。また、上記のトランスジェニックマウスもICR系統およびメラノサイト欠損系統からなる群より選ばれるマウス由来であることもできる。

本明細書の「外来のメラノサイト」における「外来」とは、毛乳頭細胞または表皮細胞の起源と起源が異なることを意味する。したがって、メラノサイトには、毛乳頭細胞または表皮細胞の起源と異種動物に由来するもののみならず同種同系統であっても毛乳頭細胞または表皮細胞を採取した個体と異なる個体に由来するものをも包含する。このことは、仮りに、毛乳頭細胞または表皮細胞調製物に生来のメラノサイトが混在する場合であっても、かようなメラノサイト以外の追加のメラノサイトが、目的の系に必ず含められることを意味する。

しかし、限定されるものでないが、毛乳頭細胞または表皮細胞がマウス由来である場合、メラノサイトは、マウス以外の動物、例えばヒト由来であることが好ましい。このような好ましい組み合わせを用いると、再構成毛包において、メラノサイトの分化状態に関わりなくメラノサイトの分布をトレースすることができる(例えば、ヒトメラノサイトに対する抗体もしくはヒトに対する特異的抗体またはヒトの特異的遺伝子配列もしくはヒトメラノサイトの特異的遺伝子配列の使用)。本発明に従えば、毛乳頭細胞(マウス)、表皮細胞(マウス)およびメラノサイト(ヒト)からなる、所謂、キメラ再構成毛包において、メラノサイトがその活性(例えば、メラニン産生活性)を維持しうることにも特徴がある。また、後述する評価方法においてはヒトメラノサイトに作用しうる手段を探索できる点も本発明に備わる特徴である。

本発明で用いることのできるメラノサイトは、本発明の目的に沿って、メラニン色素の産生能を有するものであれば、いかなる種の動物に由来するものであってもよいが、上記のような特徴を活かすには、上述のとおり、ヒト由来のメラノサイトが好ましい。メラノサイトは、適当な組織、例えば表皮、さらに包皮、等からそれ自体公知の方法で調製することができる。また、市販品を使用することもできる。例えば、ヒト由来の培養メラノサイトは、NHEM細胞の名称の下にCascade社またはクラボウ社等から入手できる。

他方、以上の毛乳頭細胞および表皮細胞は、上述のとおり、それらのレシピエント動物と同種の動物に由来するものが好ましく、本発明の目的上、かかる動物の具体的なものとしては、マウス、ラットを挙げることができる。

本発明の「毛包を再構成するための系」は、上記の毛乳頭細胞および表皮細胞と、外来のメラノサイトとの組み合わせからなる。かような「組み合わせからなる・・・・・系」とは、上記の各細胞を一体として含む場合だけでなく、将来一体として使用する目的で各細胞が個別に保存されるように、例えば、個別の容器に入れられている形態をも包含する意味を有する。したがって、それらが上記の系を再構成する目的で使用される限り、本発明の範囲内にある。また、「毛包を再構成する」とは、レシピエント動物において、発毛または育毛を支持しうる器官として役立ちうる再構成毛包(またはキメラ毛包)をもたらしうることを意味する。このような器官には、毛乳頭細胞、毛母細胞、毛包上皮系細胞・組織(例えば、毛根鞘)、脂腺、毛包をとりまく結合組織等が含まれるであろう。

本発明に従う、上記の系は、適当なレシピエント動物に移植した場合に、再構成毛包を担持するキメラ動物を提供しうる。かかるキメラ動物は、毛包内に活性な(好ましくは、異種動物由来)メラノサイトを有するので、メラノサイトの機能またはメラノサイトの活性に影響を及ぼしうる手段(薬物および環境等を含む)の評価をする上で有用である。レシピエント動物は、該動物に移植される系に含まれる各細胞の起源に拘わりなく、免疫系が抑制された動物であることが好ましい。また動物種としては、実験動物として使用しうるものであり、本発明の目的に沿うものである限り、いかなる動物であってもよいが、好ましくは、マウス、ラット等を挙げることができる。このような動物のうち、免疫系が抑制されているものとしては、マウスを例にすれば、ヌードマウスのように、胸腺欠損のごとき形質をもつものが挙げられる。なお、本発明の目的を考慮すれば、特に好ましいレシピエント動物としては、市販のヌードマウス(例えば、Balb−c nu/nu系統)、スキッドマウス(例えば、Balb/c−SCID)、ヌードラット(例えば、F344/N Jcl−rnu)を挙げることができる。このようなレシピエント動物を使用し、そして表皮細胞としてアルビノの性質を持つマウス(例えば、ICR系統)またはメラノサイト欠損マウス(例えば、Wsh/Wsh)由来のものを使用して得られる再構成毛包は、該毛包に含まれるメラノサイトの活性のトレースが容易である点で特に好ましい。

本発明に従えば、上記のメラノサイトがヒト由来であっても、上記の特に好ましいレシピエント動物、表皮細胞を使用することによって、再構成毛包からメラニン色素を含む毛が誘発および発育する。また、毛包の周辺領域にもメラニン色素の産生が観察できる。さらに、移植後6週目の毛の実体顕微鏡観察によると、その間に毛の色が、例えば灰色から白に変わることはなく、メラニンを産生するメラノサイトが長期間、再構成毛包に存在していることが確認されている。さらにまた、移植に際して、ヒト由来のメラノサイトの混合量が多いと毛の色が、一般に、濃くみえることから、再生された毛の色は毛乳頭細胞周囲のヒト由来のメラノサイトのメラニン産生量に依存するものとみなされる。

かような、本発明に従う、毛包を再構成するため系をレシピエント動物に移殖する方法は、それ自体公知の移殖方法によることができる。例えば、ヌードマウスの背部に該系を移殖する場合、その背部の直径約1cmの円に対し、毛乳頭細胞は、約50万〜1000万個、好ましくは約100万〜約400万個、マウス表皮細胞は、約100万〜4000個、好ましくは約1000万〜約2000万個使用し、培養ヒトメラノサイト(DARK)は、約50万〜1000万個、好ましくは、約100万個〜500万個、使用することができる。

以上のような再構成毛包を担持するキメラ動物は、再構成毛包から、長期にわたってメラニンを含む毛を誘発し、そして育成しうるので、毛の成長および毛の色を濃くするのに関与する細胞もしくは組織、または器官、例えば、メラノサイト、毛乳頭、毛母細胞等が正常に働いているものとみなされる。したがって、本発明に従うキメラ動物は、
1)メラノサイトを活性化させることにより、毛の色を濃くする作用のある薬物のスクリーニング、
2)毛乳頭を活性化させることにより、毛の成長を促し、毛全体の色を濃くする作用のある薬物のスクリーニング、
3)毛母細胞を刺激することにより毛の成長を促し、毛全体の色を濃くする作用のある薬物のスクリーニング、
4)毛乳頭を刺激することにより毛包内のメラノサイトを活性化させ、毛の色を濃くする作用のある薬物のスクリーニング、
5)毛母細胞を刺激することにより毛包内のメラノサイトを活性化させ、毛の色を濃くする作用のある薬物のスクリーニング、
