Description

The present invention relates to a flow cytometer or a cell sorter intended for biological fine particles such as cells and bacteria. In particular, the present invention relates to an apparatus and flow cell structure for enabling a disposable flow cell.

A flow cytometer is a scattered light that is generated when a suspension containing particles such as fluorescently labeled cells or bacteria flows through a flow cell, irradiates the channel with irradiation light, and the particles pass through the irradiation region. It is a device that quantifies the size of particles and determines the type of particles by measuring fluorescence and fluorescence. Furthermore, the flow cytometer functions as a cell analysis unit of a cell sorter device for separating cells and the like by type.

In the flow cytometer, the laser is squeezed to irradiate the flow path in the flow cell. As the irradiation beam diameter is reduced, the irradiation energy density can be increased, the signal intensity is increased, and the detection sensitivity can be increased. However, there is a large difference in signal intensity between the fine particles passing through the central portion of the irradiation beam and the fine particles passing through the bottom portion of the beam. In order to prevent this, it is necessary to implement either one of the following: 1) widen the laser beam beyond the channel width, and 2) narrowly limit the position where the microparticles pass around the sample flow at high speed. There is. In the former case, there is a drawback that the detection sensitivity is lowered because the irradiation energy density is lowered to widen the beam. Therefore, the latter is generally adopted. However, in this case, the sheath flow requires a larger amount of liquid than the sample flow, and is currently connected to an external tank of several liters or more. For this reason, replacement of the entire liquid feeding system is impossible at present. As described above, in the flow cytometer including a large volume tank in the liquid feeding system, it is impossible to easily replace the whole liquid feeding system.

Therefore, a method that enables a disposable flow cell has been proposed. In Japanese Patent Application Laid-Open No. 2004-85323 (Patent Document 1), the flow velocity of a fluid is sent by chip exchange by forming means on the flow cell chip by utilizing the flow generated by gravity due to the difference in the height of the introduced liquid surface. The entire liquid can be replaced. However, due to the difference in gravity due to the height difference of about 1 cm on the chip, the flow rate cannot be increased, and it takes 1 hour or more to measure a micro sample of about 10 microliters. Japanese Patent Laid-Open No. 2003-302330 (Patent Document 2) describes a method for achieving stable liquid feeding by applying the same pressure to two sheath liquid ports and a sample liquid port on the upstream side. However, there is a problem that the balance of the sheath flow changes when the two sheath liquid levels are different.

If the flow cell used is a quartz flow cell, it is expensive and not suitable for disposable use. On the other hand, a method for producing a resin microfluidic chip has been disclosed. Japanese Unexamined Patent Publication No. 2006-81406 (Patent Document 3) in which a substrate resin film and an injection molded resin member having a concave portion on the surface are bonded together, or the substrate resin film, spacer resin film, and another substrate resin film Are bonded in this order, and are disclosed as a microfluidic device having a polymerase chain reaction channel.

However, in flow cytometer flow cells that use laser light with a high irradiation energy density, if the material of the flow cell is an inexpensive transparent resin, self-fluorescence of the flow cell occurs even when visible light with a wavelength of 400 nm or more is irradiated. However, there is a problem that it becomes background noise light of fluorescence detection. The presence of this autofluorescence becomes a factor that degrades the sensitivity of fluorescence detection of the flow cytometer. In the case of glass, autofluorescence occurs when irradiated with light having a wavelength shorter than 400 nm.

Furthermore, in a general flow cytometer, the sample liquid is flowed at a high speed together with a large amount of sheath flow. Therefore, the sample liquid once measured is diluted with the sheath liquid and is discarded because it is contaminated with other sample liquids. It is common. However, this is a major drawback when measuring a few precious samples.

Accordingly, there is a need for a disposable flow cell in which the liquid feeding system including the sample liquid tank, the sheath liquid tank, the liquid feeding pipe, and the flow cell can be easily replaced without having to wash each time the sample is exchanged.

In the present invention, in view of the above situation, a flow cytometer apparatus and a flow cell that can replace the entire liquid feeding system by using only a disposable flow cell and replace only the flow cell, and can collect a sample in a contamination-free state. It is possible to realize an apparatus and a flow cell that simultaneously satisfy the advantages described above.

Embodiments of the present invention will be specifically described below, but the present invention is not limited to these specific examples, and it goes without saying that various modifications or improvements can be made without departing from the spirit of the present invention. Yes.

FIG. 1 is a schematic view for explaining a fine particle measuring apparatus of the present invention. 1 is a flow cell main body substrate, which is made of an acrylic transparent resin. A concave flow path pattern is formed on the back side of the substrate by injection molding, and a sheet having a thickness of about 100 μm is pasted on the flow path. Is forming. The channel cross section is typically 80 micrometers wide and 25 micrometers deep. Reference numeral 2 denotes a flow path through which the sample solution flows. Reference numeral 31 denotes a sample liquid reservoir, and the surrounding 4 is a reservoir for the sheath liquid 6. The sample liquid is separated from the sheath liquid by a partition wall. The sheath liquid is connected to a pair of flow paths that merge from both sides of the flow path of the sample liquid 3 (see FIG. 3). As shown in FIG. 1, the sheath liquid 6 connected to the pair of flow paths inside the water tank 4 has a common liquid level. Air 29 pressurized by a pump 9 is connected to the water storage tank 4 through a pipe 8 by a cap 7. Air pressure values are typically 2 to 20 kilopascals. Inside the reservoir 4, a common air pressure is applied to the sheath liquid and the sample liquid, and since the sheath liquid has a common liquid surface, an equal pressure is always applied to the pair of flow paths. To converge at the center of the flow path (see FIG. 4). Since the downstream side of the flow path is symmetric with the upstream flow path pattern, the sample liquid and the sheath liquid are separated again by the downstream branch flow path after passing through the laser irradiation region 34 (see FIG. 4). The The central branch flow path in which the separated sample liquid is collected is connected to the water storage tank 32, and the pair of branch flow paths in which the sheath flows on both sides are collected is connected to the water storage tank 5 (see also FIG. 3). Reference numerals 10 and 11 in FIG. 1 denote laser light sources having wavelengths of 473 nm and 640 nm, respectively. The laser light emitted from these light sources is aligned substantially coaxially using the dichroic mirror 28 and is irradiated to the flow path of the flow cell substrate 1. Scattered light and fluorescence generated when particles flowing in the flow path pass through the irradiation light are collected by the lens 12, 473 nm light is reflected by the dichroic mirror 16, and transmitted light is directly removed by the beam stopper, Scattered light is detected by the photodiode 18. Light having a wavelength region shorter than 550 nm is reflected by the dichroic mirror, light having a wavelength of 510 nm to 550 nm is transmitted by the bandpass filter 20, and fluorescence is detected by the photomultiplier tube 21. Light having a wavelength shorter than 750 nm is reflected by the dichroic mirror 22, light from 680 nm to 720 nm is transmitted by the bandpass filter 23, and fluorescence is detected by the photomultiplier tube 24. The light is reflected by the mirror 25, the fluorescent light having a wavelength longer than 750 nm is transmitted by the band pass filter 26, and the fluorescent light is detected by the photomultiplier tube 27. In this way, scattered light and fluorescence from the particles are detected, and the magnitude of the signal intensity is recorded for each detected particle.

FIG. 2 is a schematic diagram for explaining a particle measuring apparatus according to another embodiment of the present invention. The apparatus shown in FIG. 2 is different from the embodiment shown in FIG. 1 in that a cap is attached to the downstream water storage tank 5 and air 33 having a constant pressure lower than the atmosphere is connected. In the water storage tank 4 on the upstream side of the flow cell, the sample liquid 3 is replenished to the water storage tank 31 in a non-contact manner by dropping using a liquid feed by a metering pump. In this way, by feeding a sample solution by applying a negative pressure to the downstream water storage tank, a larger amount of the sample solution can be sent for a long period of time for measurement.

In the flow cell of the present invention, in order to reduce the autofluorescence of the flow cell, the thickness of the flow cell material (substrate) where the laser beam is transmitted is made thinner than the surroundings. The region 30 in FIGS. 1 and 2 is a region in which the thickness of the flow cell is thinner than the other regions. For example, the other region is 1.5 mm thick, but the region is 0.5 mm or less. Yes.

FIG. 3 is a schematic diagram of a flow cell according to the present invention. The lower panel of FIG. 3 represents a plan view of the flow cell. The sheath liquid 6 is introduced into the flow path from the openings 40 and 41, and merges with the sample liquid 3 from the sample water storage tank 31 downstream. The merged liquid passes through the flow path including the region 30 and is further divided into the respective flow paths downstream. The sample liquid 3 is stored in the water storage tank 32 through the central flow path. And are discharged from the openings 42 and 43 to the water storage tank 5. A flow path in the region 30 is set as a laser irradiation region. A depression is formed on the flow cell substrate on the side opposite to the side where the flow path pattern is formed, and the total thickness of the substrate thickness + flow path depth + sheet thickness in that region is set to the thickness of other regions (for example, , 1.5 mm), for example, 0.5 mm or less. As a result, the auto-fluorescence of the flow cell derived from the 473 nm laser irradiation was reduced to about 1/3 compared with the case where the thickness was the same as that of other regions (eg, 1.5 mm thickness).

FIG. 4 is a schematic diagram for explaining the flow of the sample liquid in the flow path inside the flow cell according to the present invention. 4 in FIG. 4 is a laser irradiation region in the flow path. The flow path 50 through which the sample liquid 3 from the sample water storage tank 31 flows is combined with the flow paths 51 and 52 through which the sheath liquid 6 from the water storage tank 4 flows to form a flow path 53. The flow channel 53 again branches downstream into a flow channel 54 through which the sample liquid 3 flows and flow channels 55 and 56 through which the sheath liquid 6 flows.

FIG. 5 is a schematic view showing a further embodiment of the flow cell of the present invention in which a plurality of flow path patterns shown in FIG. 3 are formed on a substrate. Thus, by forming a plurality of flow cell patterns on one substrate, the efficiency of fine particle separation can be increased.
