【発明の属する技術分野】
本発明は、ウヤク、ロコン、シツリシから選ばれる１種以上の植物抽出物を含有するメラニン生成抑制剤、並びに医薬品・医薬部外品又は化粧品分野の各種の美白化粧料組成物への応用に関するものである。

その利用分野は、メラニン生成抑制剤として、又は美白化粧料組成物として、各種の内用・外用製剤類（動物用に使用する製剤も含む）全般において利用でき、具体的には、アンプル状、カプセル状、丸剤、錠剤状、粉末状、顆粒状、固形状、液状、ゲル状又は気泡状の1)医薬品類、2)医薬部外品類、3)局所用又は全身用の皮膚用美白化粧品類、4)頭皮・頭髪に適用する薬用又は／及び化粧用の製剤類、5)浴湯に投じて使用する浴用剤、6)その他、液臭・防臭防止剤や衛生用品、衛生綿類、ウエットティシュなどが上げられる。

【従来の技術】
人の皮膚色を決定する因子として、表皮中のメラニン量，毛細血管の血流量，食物由来の色素（カロチン），角質層の厚さなどがあるが、これらの内、最も重要な因子としての一つにメラニン色素がある。いわゆる、肝斑（シミ）、雀卵斑（ソバカス）、日焼け後の皮膚の色素沈着は、皮膚内に存在する色素細胞の活性化によりメラニン色素の生成が著しく亢進したものであり、中高年齢層や女性にとっては肌の大きな悩みの一つとなっている。

このメラニン色素は表皮基底層及び毛根部，外毛根鞘に存在する色素細胞（メラノサイト）内の小器官であるメラノソームで生成される。メラニン色素の生成過程は、色素細胞（メラノサイト）内でチロシンにチロシナーゼが活性作用して、チロシンが酸化され、ドーパ、ドーパキノンに変換、更に自動酸化し、ド−パクロム、５，６一ジヒドロキシインドールを経て重合し、最終的にはメラニン色素になる。生成されたメラニンは、色素細胞（メラノサイト）の樹枝状突起から基底細胞に分泌され、基底細胞が分裂、有棘細胞となると共に上昇し、角質層に達した後、角質層の剥離と共に脱落して行くとされている。

従って、肌の肝斑（シミ）、雀卵斑（ソバカス）や色黒を防止又は改善するためには、メラニンの生成過程を阻害すること。或いは既に生成されたメラニンを淡色化させることが考えられる。そこで、これらの考えに基づき、従来から種々の美白成分が提案されている。例えば、コウジ酸及びその誘導体、アスコルビン酸及びその誘導体、トコフェロール及びその誘導体、グルタチオン、過酸化水素、過酸化亜鉛、ハイドロキノンモノベンジルエ−テル、プラセンターエキス、シルク抽出物や植物エキス（アルニカ、アルテア、アロエ、オウゴン、エンメイソウ、カミツレ、甘草、クチナシ、ゲンノショウコ、シコン、ショウマ、シラカバ、センキュウ、ゲンノショウコ、サイコ、山茶花、当帰、トウキンセンカ、ニワトコ、紅花、ニンニク、ハトムギ、レイシ、ログウッド）などが知られている。

【発明が解決しようとする課題】
しかしながら、前記した美白成分については、処方系中での安定性や溶解性が悪く、澱や分解による異臭、着色が生じ、又、生体レベルにおける効果も充分ではなく、更に皮膚にかぶれを起こすなど安全性の面でも問題があり、満足すべきものではなかった。

【課題を解決するための手段】
こうした事情に鑑み、本発明者らはメラニン生成抑制効果がある有用な植物を開発のテーマとし、その結果、ウヤク、ロコン、シツリシから選ばれる１種以上の植物抽出物が、メラニン色素の生成を有意に抑え、美白効果を有することを確認、更に肌のシミ、ソバカスを軽減し、又、乾燥肌の改善、肌にツヤ・張りも与えることができる安全性の高いメラニン生成抑制剤とそれを含有した美白化粧料組成物を提供することをもって、本発明を完成するに至った。

【発明の実施の形態】
尚、本発明で使用する「ウヤク：烏薬，天台烏薬」とは、クスノキ科（Lauraceae）、クロモジ属（Lindera）の植物：テンダイウヤク「Lindera strychnifolia(Sieb.et Zucc.)F.Vill. （=Benzoin strychnifolium(Sieb.et Zucc.)O.Kuntze）」の根茎又は根皮、根を用いる。

「シツリシ」とは、ハマビシ科（Zygophyllaceae）、ハマビシ属（Tribulus）の植物ハナビシ（Tribulus terrestris（L.））の果実を用いる。

本発明で使用するウヤク、ロコン、シツリシの抽出物とは、各々の植物体の各種部位（全草、花、頭花、花穂、果皮、果実、茎、葉、枝、枝葉、根茎、根皮、根など）をそのまま或い粉砕後搾取したもの。又は、そのまま或いは粉砕後、溶媒で抽出したものである。

抽出溶媒としては、水、アルコール類（例えば、メタノール、無水エタノール、エタノールなどの低級アルコール、或いはプロピレングリコール、1,3-ブチレングリコールなどの多価アルコール）、アセトンなどのケトン類、ジエチルエーテル、ジオキサン、アセトニトリル、酢酸エチルエステルなどのエステル類、キシレン、ベンゼン、クロロホルムなどの有機溶媒を、単独で或いは２種類以上の混液を任意に組み合わせて使用することができ、又、各々の溶媒抽出物が組み合わされた状態でも使用できる。

又、ウヤク、ロコン、シツリシの各植物抽出物は、応用するメラニン生成抑制剤、美白化粧料組成物の剤型・形態により乾燥、濃縮、或いは希釈などを任意に行い調整すれば良い。

尚、製造方法は特に制限されるものはないが、通常、常温〜常圧下での溶媒の沸点の範囲であれば良く、抽出後は濾過又はイオン交換樹脂を用い、吸着・脱色・精製して溶液状、ペースト状、ゲル状、粉末状とすれば良い。更に多くの場合は、そのままの状態で利用できるが、必要ならば、その効力に影響のない範囲で更に脱臭、脱色などの精製処理を加えても良く、脱臭・脱色などの精製処理手段としては、活性炭カラムなどを用いれば良く、抽出物質により一般的に適用される通常の手段を任意に選択して行えば良い。

本発明のウヤク、ロコン、シツリシの各植物抽出物は、メラニン生成抑制剤、美白化粧料組成物へ配合でき、その配合量としては特に規定するものではないが、メラニン生成抑制剤、美白化粧料組成物の種類、品質、期待される作用の程度によって若干異なり、通常、０．００１重量％以上（以下、重量％で表わす）好ましくは０．２〜２０％が良い。尚、配合量が０．００１％より少ないと効果が充分期待できない。

尚、本発明のメラニン生成抑制剤、美白化粧料組成物は、前記の必須成分に加え必要に応じ、本発明の効果を損なわない範囲内で、医薬品類、医薬部外品類、化粧品類などの製剤に使用される成分や添加剤を任意に選択・併用して製造することができる。

(13)植物又は動物系原料由来の種々の添加物
これらは、添加しようとする製品種別、形態に応じて常法的に行われる加工（例えば、粉砕，製粉，洗浄，加水分解，醗酵，精製，圧搾，抽出，分画，ろ過，乾燥，粉末化，造粒，溶解，滅菌，ｐＨ調整，脱臭，脱色などを任意に選択、組合わせた処理）を行い、各種の素材から任意に選択して供すれば良い。

尚、抽出に用いる溶媒については、供する製品の使用目的、種類、或いは後に行う加工処理などを考慮した上で選択すれば良いが、通常では、水，水溶性有機溶媒（例えば、エタノール，プロピレングリコール，１,３−ブチレングリコールなど）の中から選ばれる１種若しくは２種以上の混液を用いるのが望ましい。但し、用途により有機溶媒の含有が好ましくない場合においては、水のみを使用したり、或いは抽出後に除去しやすいエタノールを採用し、単独又は水との任意の混液で用いたりすれば良く、又、搾取抽出したものでも良い。

尚、植物又は動物系原料由来の添加物を、全身用又は局所用の外用剤、化粧品類に供する場合、皮膚や頭髪の保護をはじめ、保湿，感触・風合いの改善，柔軟性の付与，刺激の緩和，芳香によるストレスの緩和，細胞賦活（細胞老化防止），炎症の抑制，肌質・髪質の改善，肌荒れ防止及びその改善，発毛，育毛，脱毛防止，光沢の付与，清浄効果，疲労の緩和，血流促進，温浴効果などの美容的効果のほか，香付け，消臭，増粘，防腐，緩衝などの効果も期待できる。

（試験１）メラニン生成抑制作用試験
本発明者は、Ｂ１６メラノーマ細胞をもとにメラニン生成抑制作用を評価することとした。

「試験方法及び評価方法」
24時間前培養したB16メラノーマ細胞を新鮮な培地に移し、これに試料を0.5％添加し、2日間培養する。次に培養処理した細胞をトリプシンで処理し、1N-NaOH，10％DMSO溶液で加熱溶解後、420nmにおける吸光度を測定する。尚、試料の代わりに、ブランクとして蒸留水を用い、又、同時に試料を添加した細胞をＭＴＴ還元法（Tim Mosmann；Journal of Immunological Methods p55−63(1983)参考）にて活性を測定し、ブランク細胞のＭＴＴ還元活性を１単位として、試料負荷細胞のＭＴＴ還元活性を算出した。試料のメラニン量を、吸光度／ＭＴＴ還元活性（１単位）として算出し、ブランク細胞のメラニン生成量に対する抑制率（％）を数１により求めた。結果は図１〜２に示した。
