本発明は、例えば糖やペプチド,蛋白質等の生体関連の混合試料の分離分析に用いられる、液体クロマトグラフと質量分析計とを結合した装置、すなわち液体クロマトグラフ・質量分析計に関する。

【従来の技術】
現在、分析の分野では、生体関連物質の質量分析法の開発が重要視されている。生体関連物質は通常混合物として溶液中に溶け込んでいるため、混合物を分離する手段と質量分析計とを結合する装置の開発が進められている。この方法の代表的な装置として、液体クロマトグラフ・質量分析計(以下、LC/MSと略する)がある。液体クロマトグラフ(以下、LCと略する)は混合物の分離に優れるが物質の同定ができず、一方質量分析計(以下、MSと略する)は感度も高く物質の同定能力に優れるが混合物の分析は困難である。そこで、LCの検出器としてMSを用いるLC/MSは、混合物の分析に対して大変有効である。

MS以外の検出器、例えば紫外線の吸光度の変化により試料を検出する紫外吸光検出器を有するLCでは、分離能力や試料の溶出時間の再現性を向上させるために、難揮発性塩を含む移動相溶媒を用いる場合がある。しかしながら、LC/MSに難揮発性塩を含む移動相溶媒を用いると、この塩がイオンを大気中から真空中へと導入するためのイオン導入細孔の周辺に析出し、イオン導入細孔を閉塞させてしまう。このため、LC/MSにおいて、難揮発性塩を含む移動相溶媒は使用が困難であり、LCの高い分離能力を十分に活かした分析ができなかった。

【課題を解決するための手段】
本発明においては、LCから送られて来る試料溶液を静電噴霧法により霧化し、得られた液滴を気化して得られるガス状の試料分子を化学反応によりイオン化し、この試料分子に関するイオンをMSで分析することにより前記目的を達成する。

図1は、本発明のLC/MSの構成を示す概略図である。LC14で分離された試料は移動相溶媒とともに静電噴霧部15にて噴霧される。噴霧により得られた液滴は気化部5において気化が促進される。気化部5で生成されたガス状の試料分子はイオン化部6において化学反応によりイオン化される。イオン化部6で生成された試料に関するイオンは、イオン導入細孔9a,排気系により排気された差動排気部11,イオン導入細孔を通して9b,排気系により排気された高真空部12に導入される。真空中に導入されたイオンは質量分析部13で質量分析される。

【発明の実施の形態】
図2は、図1に示した構成のより詳細な構造を示す図である。LCから溶出してくる試料溶液は配管1,コネクタ2を介して金属管3に導入される。金属管3と金属ブロック4bとの間に高圧電源8bにより数kVの電圧を印加することにより、試料溶液を静電噴霧させる。噴霧により生成された微小な液滴は加熱された金属ブロック4bにより構成される気化部5へと導入される。金属ブロック4bはヒーター(図示略)により約300℃程度に加熱されている。噴霧により生成された液滴は、金属ブロック4bの開口部を通過する間に、熱により気化される。

気化部5において気化された試料分子はイオン化部6へと導入される。イオン化部6には針状電極7が設けられている。この針状電極7に高圧電源8aにより数kVの高電圧を印加することにより、イオン化部6にコロナ放電を発生させる。

金属管3の先端は、図3に示すように、気化部6の中に設けても良い。また、同じく図3に示すように、溶液を金属ブロック4bに向けて直接噴霧してもよい。試料溶液は金属管3と金属ブロック4bとの間に高圧電源8bから印加される高電圧によって静電噴霧される。金属管3と金属ブロック4bとの間の絶縁は絶縁管19により行う。溶液の沸点より高い温度に加熱された金属ブロック4bに吹き付けられた液滴は瞬時に気化し、ガス状の試料分子が得られる。試料分子がコロナ放電部分に到達すると、コロナ放電により生成されたヒドロニウムイオンなどの一次イオンと化学反応を起こし、試料分子のイオン化が達成される。試料分子に関するイオンはイオン導入細孔9a,排気系10aによって数十Paから数百Pa程度に排気された差動排気部11を介して排気系10bによって10−3Pa程度に排気された高真空部12に取り込まれ、質量分析部13よって質量分析される。

また、図2に示した構成において、静電噴霧で大きな液滴が生成されてしまうと、加熱金属ブロック4bによる気化部5では完全に気化できず、針状電極7によってコロナ放電が発生しているイオン化部6に液滴のまま到達する場合がある。液滴がコロナの生じている部分に到達すると、針状電極7とイオン導入細孔9aとを短絡させ、高圧電源8a等を故障させる可能性がある。

これを防ぐためには、図4に示すように、金属管3の末端と針状電極7によりコロナ放電が発生しているイオン化部6とを遮蔽するように電極21aを配置し、その電極21aに向けて静電噴霧させても良い。この場合、液滴の気化効率を上げるため、電極21aはヒーター22aにより加熱されていることが望ましい。図4に示した構成により、ガス状の分子だけがイオン化部6に運ばれイオン化されるため、液滴が針状電極7に付着することによる短絡が回避される。図4において、電極21aの形状は板状のものだけではなく、メッシュであっても良い。試料分子のイオン化部6への到達効率を上げるため、図3と同様に、イオン化部6に向けてガス20を流しても良い。

図5に、気化手段に赤外線の照射を用いる実施例を示す。試料溶液は金属管3とメッシュ23aとの間に印加される電圧により静電噴霧される。このメッシュ23aは加熱されていることが望ましい。噴霧により得られた液滴は、気化部5へと送られる。気化部5では、電源24に接続されたヒーター22bから放射される赤外線を液滴に照射し、液滴を気化させる。液滴がヒーター22bに直接当たることによりヒーター22bの劣化が起こる場合には、ヒーター22bを保護するためのガラス管25をヒーター22bの内側に設けてもよい。液滴の気化の効率を向上させるため、噴霧用ガス17中の水蒸気は除去されていることが望ましい。また、噴霧用ガス17は室温以上に加熱されていることが望ましい。気化部5において得られたガス状の試料分子は、針状電極7によるコロナ放電でヒドロニウムイオン等が生成されているイオン化部6においてイオン化される。

図2から図5に示した構造により、LC/MSにおいて難揮発性の塩を含む移動相溶媒を使用することが可能であるが、極めて高濃度の塩を使用する場合や分析に長時間を要する場合には、図6に示すように、イオン導入細孔9aの中心軸と垂直の方向に噴霧させても良い。金属管3に導入された試料溶液は、対向する電極21bに向けて噴霧される。この電極21bは溶液の沸点以上に加熱されていることが望ましい。

図7は、図6に示した対向電極21bの代わりにメッシュ23bに向けて噴霧する構成を示す図である。このメッシュ23bは加熱されていることが望ましい。メッシュ23bの背部に受け皿28を設けておくことができ、受け皿28を交換することにより簡便にメンテナンスが行えるほか、受け皿28に溜った溶液を分取し、質量分析法以外の分析手法、例えば蛍光や発光による分析法や免疫学的分析法などを行うことができる。

本発明によれば、従来のLC/MSにおいて使用が困難であった難揮発性塩を含む移動相溶媒を使用することが可能となる。このため、LCの高い分離能力を十分に活用でき、LC/MSの適応範囲が拡がり、より多くの物質の分析が可能となる。
