うな現在の治療方法では、急速に成長する癌細胞を選択
的に殺す薬剤が中心となっている。残念ながら、骨髄あ
るいは消化管上皮のような正常な重要組織とは違って、
腫瘍の多くはあまり早い速度では成長しない。もう1つ
の治療アプローチとして、腫瘍細胞表面の特異な化学構
造を標的とし、最も頻繁に採用される例では、この標的
分子と選択的に結び付く抗体を用いる方法が挙げられ
る。この種の治療アプローチの1つに、抗体と植物毒素
または細菌毒素のような細胞殺傷性物質とを結合させた
ものがある。この抗体・毒素錯体、言い替えると免疫毒
素類(immunotoxins)は、組織培養及び動物実験による
腫瘍系モデルにおいて選択的な腫瘍細胞殺傷効果を示す
ことが認められている(Pastan et al.,Cell 47:641
−648(1986))。
を遊離しようと多くの試みがなされたが、選択的に腫瘍
細胞のみに結び付いて、他の正常な重要組織には結び付
かない抗体として同定されたものは、現在でもなお極め
て小数である。従って、免疫主導の治療を行ってゆく上
で、このような腫瘍特異性を持つ抗体を遊離することは
重要である。
(Kohler and Milstein)よって初めて発表され(Nat
ure 156:495−497(1975))、さらにコプロフスキー
ら(Koprowski et al)の特許(米国特許番号第4,17
2,124号)で開示されたものがあり、これにより、抗体
を純粋な形で遊離して治療薬にすることが出来るように
なった。しかしながら、二つの理由からこれまでに遊離
された多くの抗体を使用することができなかった。腫瘍
細胞に反応するモノクロナール抗体は、重要なヒトの正
常組織にも反応してしまうのが第一の理由。第二の理由
としては、多くの遊離抗体は表面成分に結び付くが、こ
の表面成分がトキシン共役系がエンドサイトーシスによ
り細胞内に入り込むのを能率的に仲介しないことが挙げ
られる。本発明は、選択的にある種のヒト腫瘍に結び付
くが、多くの正常な重要組織のには結び付かないモノク
ロナール抗体、K1に関する。このモノクロナール抗体
は、免疫毒素の標的用成分として添加されると、毒性物
質を効果的に細胞内には入り込ませることが出来ること
が認められている。

1))。しかしながら、K1モノクロナール抗体が細胞表

ープとは全く異なる。さらに、K1と反応する抗原は、卵
巣癌患者の血漿に流れ込まない。血漿に流れ込まないこ
とから、血流中に注入された直後でも循環している抗原
により中和されることないので、K1は、免疫療法に好適
の抗体である。

ハイブリドーマ細胞系は、1990年10月10日、寄託番号AT
CC HB 10570としてメリーランド州ロックビルのアメ
リカンタイプカルチャー コレクションにブタペスト条
約に基づいて国際寄託されている。

処理をした別のウェルにヨウ素化した放射性K1を入れた

掲のウェルとを一緒にインキュベートした。表8と同じ
ように、OVCAR−3培養基の上澄み液(濃縮物を1:3で希
釈したもの、測定値は2で除す)では、OC125の結合が
標識を付けたOC125によって検出された。これとは対照
的に、結合したK1加えてインキュベートした上澄み液で
は、ヨウ素化K1を用いて検出を図ったが、検知できなか
った。精製CA125抗原の標識サンプルは、OC125系とは反
応することが認められたが、K1系とは反応しなかった。
患者の血清サンプルはOC125系とは各種強度のシグナル
を示したが、K1系とのシグナルは認められなかった。K1
系は、トリプシン処理によりOC125が破壊されたOVCAR−
3細胞のトリプシン塩(trypsinate)では弱いシグナル
を検出したが、OC125系ではシグナルは認めなかった。
これら二つの標識を付けた抗体のアリコートを作ってチ
ェックしたところ、各種水準の充分な放射能が認めら
れ、充分な感度を有することが認められた。

だクローンは、癌との反応性は高いが、ヒトの正常細胞
とは反応しない。抗体は、マウスで腹水生産したクロー
ンから作ったり、クローンの培養上澄み液を採取して、

していることはすでに立証されている。これらの細胞に
は、例えばOVCAR−2、OVCAR−3、OVCAR−4、1874、H
TB77、HeLa S3、KB、AGS、HTB103(例えば、Kato II
I)がある。しかしながら、これらの細胞系のあるもの
(例えば、1847、AGS、Kato III)は、OC−125と反応
しない。

た、ある種の細胞系がK1と均質な反応をしているので、

示す可能性も考えられる。一つの癌の細胞全部と均質な
反応をすることは、一区分の癌細胞ばかりではなく、一
つの癌のあらゆる細胞を撲滅して免疫療法を成功させる
大きな要因となる。

を認識することを明確に示している。

から、K1に反応する抗原は卵巣癌細胞から流れ出すこと
がなく、免疫療法で使用する抗体としてK1がOC125より
も遥かに適していることが分かる。さらにまた、精製さ
れたCA125抗原は、K1と反応しないことも注目に値す
る。CA125はOC125に反応する標準抗原である。

た。その結果では、K1とOC125とを一緒に添加すると、
広い濃度範囲(1−50μg/ml)において、それぞれの抗

った(下記表9)。この事実から、エピトープはお互い
に物理的に充分に離れていて、両方がそれぞれ独立して
同時に占有されることが出来ると考えられる。

K1は免疫療法に好適である。二つの抗原が全く異なる分
子に存在するのであれば、K1の方が抗原の発現を司る遺
伝子を単離して、潜在的に有用な新しい試薬を作り出し
易いかもしれない。

エピトープの構造の一部に多量の炭水化物があるせいで
あろうと思われる。このような炭水化物の付加物は、CA
125の発現を司る遺伝子を単離するために使わなくては
ならない細菌系中にはない。K1抗原の遺伝子は、細菌の
発現系で失われるような炭水化物の付加物またはその他
の翻訳後の変更を必要としないかもしれないので、K1抗
体は、K1に反応する抗原の構造上の遺伝子を単離するの
に効用があるかもしれない。この様な遺伝子を発現する
ことによって、純粋な抗原を作ること、抗原分子上の異
なるエピトープに対して第二世代のモノクロナール抗体
を単離すること、および異なる癌サンプル中のこの抗原
の遺伝子を遺伝学的手法でスクリーニングするために用
いる試薬を用意することが可能となる。
リプシンで処理すると細胞からK1エピトープを取り除く
ことが出来ることが分かった。また、細胞を過ヨウ素酸
塩で処理すると、OC125との反応性は低くなるが、K1と
の反応性は僅かながら高くなる。細胞抽出物にタンパク
質イムノブロット(immunoblots)をしたもの、培養し
たOVCARの上澄み液、精製したCA125抗原についてSDS・
ゲル・ブロットを行ったところ高分子量(>200キロダ
ルトン)のOC125と反応するスミアを示すが、同じ領域
でK1とは反応しなかった。従って、K1抗原の真の化学的
性質はどんなものかの点は明らかになっていない。しか
しながら、K1抗原の化学的性質または構造は、CA125と
は明かに異なっている。

来る。

つくる際の標的メカニズムとして用いることが出来る。
抗体の不変領域をヒトその他の種の不変領域に変換する
ことを含む遺伝学的手法を用いて、抗体の構造を操作す
ることが出来る。従って、免疫療法の複合体は、天然の
ままの抗体か、遺伝学的手法で変形した抗体の何れかを
含有することとなる。さらに、クローン化されたK1抗体
の可変部遺伝子を用いて融合タンパクを開発することが
出来る。
リンGの不変領域を置換すると、免疫系が害されていな
い患者において、抗体が主導する細胞の殺傷を仲介する
上で効用が高いヒト・マウス・キメラ抗体を作ることが
出来、免疫毒素と較べて毒性の低い免疫療法となるであ
ろう。
メジーングなど免疫診断検定の標的メカニズムとして使
うこともできる。また、K1と反応する抗原の発現は、癌
の病因の診断や、転位の組織分布のための免疫組織化学
的な病理診断手段として有用である。具体的には、組織
サンプル(例えば、生検)または細胞調製物(例えば、
パパニコラウ・スミア、卵巣含浸出物など)を用いて免
疫組織化学的な病理診断を行うことが出来る。この他に
も、外科診査の際にK1抗体に放射能標識をつけて腫瘍の
位置診断を行うこともできる。
の実施例を理由として本発明を限定してはならない。
HT−29、MDA−MB−469、DU145、HTB20およびHTB33は、
従来から開示されていた(Hay et al.,American Typ
e Culture Collection Catalog of Cell Lines
and Hybridomas,6th Ed.(1988))。マウスに、ヒト
の正常な腎臓の膜に対する耐性を与えた(Matthew et
al.,J.Immnunol.Methods,100:73−82(1987))。続
いて、このマウスにOVCAR−3細胞で免疫性を与えた(W
illingham et al.,Proc.Natl.Acad.Sci.USA84:2474−
78(1987))。但し、培養細胞は過ヨウ素酸塩で処理し
た。過ヨウ素酸塩で処理するのはアンチ・マウス免疫グ
ロブリンGを細胞表面に結びつけて、マウス抗体を運ん
でいるリンパ球に対する免疫抗原の標的特性を向上させ
るために行うものである。免疫生を与えられたマウスか
ら脾臓を摘出懸濁し、懸濁した脾臓からOVCAR−3と反
応する細胞を選び出し、続いてパンニング法(panning
method)により融合させた。選び出された脾臓細胞
を、AG8マウス骨髄腫細胞(AG8 mouse myeloma cell
s)と融合させて、クローンを形成した。2週間後に、
生存OVCAR−3細胞にローダミンによる間接蛍光抗体法
を行うスクリーンファスト(ScreenFast)(Life Tech
nologies,Ind.,Gaithersburg,MD)大規模スクリーニン
グ室を使用してクローンのスクリーニングを行った。こ
の様にして選ばれたクローンに対して、ヒトの癌よび正
常細胞から冷温によって作ったサンプルにペルオキシダ
ーゼ免疫組織化学法を実施して、第二次スクリーニング
を行った。一つのクローン、K1が選び出されたが、この
K1は卵巣の嚢胞腺癌とは反応したが、正常な肝臓、腎
臓、結腸、小腸、骨髄、小脳、肺、心臓または大脳皮質
には反応しなかった。このクローンは、もともとは、免
疫グロブリンM(IgM)抗体クローンであったが、パン
ニング・イソタイプ・スウィッチ法(panning−isotype
switching method)(Changら、発表準備中)を用い
てクローン細胞継代培養をした後、免疫グロブリンG
(InG)変異体に転換したものである。
タイプを検定したところ、マウス免疫グロブリンG1k(I
gG1k)であった。免疫グロブリンGのクローンをマウス
の腹水で生産するか、あるいは培養基の上澄み液を採取
して抗体を得て、タンパクA FPLCアフィニティカラム
を用いて抗体を精製した。
ルオキシダーゼ免疫組織化学法の使用 ヒトおよびカニクイサルの正常組織の新鮮凍結サンプ
ルならびにヒト腫瘍のサンプルから冷温にて切片をつく
り、カバーグラスの上に乗せ、K1を一次抗体として用い
て、前述のペルオキシダーゼ免疫組織化学法(Willingh
am,FOCUS 12:62−67(1990))を実施した。ペルオキ
シダーゼ基質とジアミノベンジジンとの反応を利用し
て、各種組織中の反応性抗原の局在を検出した。この実
施例では、反応性抗原の局在がヒトおよびサル双方の組
織のうち、腹膜、胸膜、心膜を裏打ちしている中皮にあ
ることが証明された。量は減るが、反応性抗原はまた、
気管、扁桃腺、輸卵管の上皮にも見いだされた。ヒト腫
瘍サンプルでは、卵巣、食道および子宮頚部それぞれの
腫瘍がK1と反応し、程度は低くなるが乳腫瘍と結腸腫瘍
とも反応した。
てを洗い流し、モノクロナール抗体K1(1μg/ml)と共
に緩衝食塩水中で4℃によりインキュベートした。洗っ
た後、細胞をローダミンと複合しているヤギのアンチマ
ウス免疫グロブリンG抗体中でインキュベートした後、
フォルムアルデヒドで固定し、前述した蛍光顕微鏡によ
り観察を行って(Willingham,FOCUS 12:62−67(199
0))結合している抗体を検出した。K1抗体に強い反応
を示した細胞のうちには、例えばKG、HeLa S3、OVCAR
−3、AGS、Kato IIIおよび1847細胞があった。

−3を、K1抗体またはOC125抗体(10μg/ml)のローダ
ミン標識の直接複合体と一緒に4℃でインキュベートし
た。このインキュベートを行う前に、この細胞を同一の
無標識の競合体である抗体と共に前インキュベートし
た。インキュベート後、細胞を洗い、フォルムアルデヒ
ドで固定して蛍光顕微鏡で観察を行った。K1−ローダミ
ンは、競合剤としての抗体の不存在下または競合剤とし
てのOC125の存在下では、強い標識を示したが、K1が競
合剤として使用された場合、標識を全く示さなかった。
反対にOC125の場合には、競合剤の不存在下、または余
剰のK1が競合抗体として存在するときに、強い標識を示
し、OC125を競合剤として使用すると標識を全く示さな
かった。この結果は、OC125とK1のエピトープの間に
は、交差反応が無いことを示している。
Claims (21)
- 【請求項1】細胞表面の抗原に対して特異性を持つモノ
クローナル抗体を産生し、寄託番号ATCC HB 10570の
ハイブリドーマ細胞系のハイブリドーマであって、該抗
原が霊長類の正常組織、培養されたヒトの悪性細胞系、
及びヒトの腫瘍に発現することを特徴とし、該抗原が培
養基又は血漿に流れ込まない上記ハイブリドーマ。 - 【請求項2】上記培養ヒト悪性細胞系がOVCAR−2、OVC
AR−3、OVCAR−4、1847、HTB77、HeLa S3、KB、AGS
及びHTB103からなる群より選ばれることを特徴とする請
求項1に記載のハイブリドーマ。 - 【請求項3】上記ヒト腫瘍が卵巣癌、食道癌又は子宮頚
部癌に由来する請求項1に記載のハイブリドーマ。 - 【請求項4】上記霊長類の正常組織が中皮である請求項
1に記載のハイブリドーマ。 - 【請求項5】細胞表面の抗原に特異性を持ち、かつ寄託
番号ATCC HB 10570のハイブリドーマ細胞系のハイブ
リドーマによって産生されると共に免疫グロブリンGク
ラスであるモノクローナル抗体であって、該抗原が霊長
類の正常組織、培養されたヒトの悪性細胞系、及びヒト
の腫瘍に発現することを特徴とし、該抗原が培養基又は
血漿に流れ込まない上記モノクローナル抗体。 - 【請求項6】上記培養ヒト悪性細胞系がOVCAR−2、OVC
AR−3、OVCAR−4、1847、HTB77、HeLa S3、KB、AGS
及びHTB103からなる群より選ばれることを特徴とする請
求項5に記載のモノクローナル抗体。 - 【請求項7】上記ヒト腫瘍が卵巣癌、食道癌又は子宮頚
部癌に由来する請求項5に記載のモノクローナル抗体。 - 【請求項8】上記霊長類の正常組織が中皮である請求項
5に記載のモノクローナル抗体。 - 【請求項9】上記抗体がK1である請求項5に記載のモノ
クローナル抗体。 - 【請求項10】請求項5に記載の癌の診断用モノクロー
ナル抗体。 - 【請求項11】毒素、放射性同位元素又は化学療法剤と
複合している請求項5に記載のモノクローナル抗体を含
む複合体。 - 【請求項12】癌の治療のための請求項11に記載の複合
体。 - 【請求項13】上記癌が卵巣癌である請求項12に記載の
複合体。 - 【請求項14】上記抗体が細胞の殺傷を仲介するように
修飾されている請求項12に記載の複合体。 - 【請求項15】請求項11に記載の複合体と医薬的に許容
できる担体を含有する癌の治療用の医薬組成物。 - 【請求項16】癌の成長を阻止するのに充分な濃度の請
求項5に記載のモノクローナル抗体と医薬的に許容でき
る担体とを含有する癌の治療用の医薬組成物。 - 【請求項17】a)腫瘍化している疑いのある細胞を含
む組織又は体液サンプルを患者から採取し、 b)該サンプルを細胞表面抗原と結合できる抗体と反応
させ、 c)該患者のサンプルからの細胞の表面において該抗体
と抗原の反応による抗原抗体結合体の存在を検出する ことを含む、 寄託番号がATCC HB 10570のハイブリドーマから分泌
される免疫グロブリンGクラスであるモノクローナル抗
体K1を用いて該細胞表面抗原を発現する患者の腫瘍細胞
を検出する方法。 - 【請求項18】腫瘍細胞が卵巣細胞に由来する請求項17
に記載の方法。 - 【請求項19】該患者からの組織又は体液サンプルにお
いて細胞表面抗原CA125を検出することをさらに含む請
求項18に記載の方法。 - 【請求項20】該抗原抗体結合体の存在が細胞調製物に
おいて検出される請求項17の方法。 - 【請求項21】該抗原抗体結合体の存在が組織サンプル
において検出される請求項17の方法。
