(2)生体試料中の当該レクチンと結合する生体成分の存在の有無又は存在量を分析すること、

(4)治療の有効性を判定するための、治療前と治療後の当該生体成分の存在の有無又は存在量の差のデータを収集すること、

また、上記レクチンをスライドグラスなど透明基板上に固定化しておき、溶液中に懸濁したがん細胞含有被検試料を、そのまま、もしくは希釈して、又はあらかじめタンパク質画分のみに濃縮した後、「Cy3-NHS ester」などで標識化した後、固定化レクチンと反応させ、プレートリーダー、蛍光スキャナー、エバネッセント波励起蛍光検査系などにより、その結合を分析してもよい。

被験個体の生体試料で分析された上記レクチンに結合する生体成分の量が、健常者又は悪性度の低いがん患者と比較して異なるレベルであるとき、被験個体のがんの悪性度が高いと判定することができる。
また、被験個体の上記レクチンに結合する生体成分の量を分析する際に、健常者又は悪性度の低いがん患者の上記レクチンに結合する生体成分の量をコントロールとして分析し、比較してもよい。
さらに、あらかじめ健常者又は悪性度の低いがん患者の生体試料中の上記レクチンに結合する生体成分の量を分析しておき、その量のカットオフ値を定め、被験個体の生体試料中の上記レクチンに結合する生体成分の量がカットオフ値を超えた場合(レクチンががん細胞由来の生体成分に特異的に結合する場合)又は下回った場合(レクチンが正常細胞由来の生体成分に特異的に結合する場合)に、該被験個体のがんの悪性度が高いと判断することができる。この際、生体試料が細胞試料の場合、上記レクチンに結合する生体成分の量は、上記レクチンに結合する生体成分を発現している細胞の割合で表してもよい。

本実施例では、臨床膵癌ゼノグラフトマウスモデル腫瘍へのTJA IIの結合活性を免疫組織化学的に評価した。
臨床膵癌ゼノグラフトマウスモデルから摘出した腫瘍をそれぞれホルマリン固定し、パラフィン包埋後、組織切片を作製した。得られた各組織切片を西洋ワサビペルオキシダーゼ(HRP)標識TJA IIで染色して、ヘマトキシリン/エオシン染色した後、その組織像を顕微鏡観察した。
その結果、臨床膵癌ゼノグラフトマウスモデル腫瘍ではがん細胞がHRP標識TJA IIで強く染色されることが分かり、特に腺管構造部位が強く染色されること、及び周辺の正常膵臓細胞部位は全く染色されないことがわかった(図1)。

DynabeadsM280 Streptavidin (Invitrogen)にビオチン化ADAを結合させた。そして、膵がん及び健常者血清と4℃一晩インキュベートして、洗浄後、0.2% SDSを含むトリスバッファーで95℃5分加熱することで、ADAに結合した糖タンパク質を溶出した。得られた溶出液を電気泳動して、PVDF膜に転写後、HRP標識ADAでブロットした。その結果、健常者血清ではADAブロットで反応性は見られなかったが、膵がん血清では250 kDa付近で反応性が認められた(図3)。そこで、レクチンブロットで使用したものと同じ血清からADA固定化ビーズで沈降させた糖タンパク質溶液を、電気泳動して銀染色MSキット(富士フイルム和光純薬、299-58901)を用いて銀染色したゲルを作製し、160 kDa付近のバンドを切り出した。切り出した各ゲル片からトリプシン加水分解を経てペプチド混合物を得た。ペプチド混合物をLC-MS/MSに供し、MS/MSデータを取得した。MS/MSデータのアミノ酸配列データベース検索の結果をペプチド同定一覧として出力した。その結果、フォンウィレブランド(Von Willebrand)因子がADAによりがん患者において検出される候補タンパク質として取得された。

As a result, no reactivity was observed in the ADA blot in the healthy serum, but reactivity was observed in the vicinity of 250 kDa in the pancreatic cancer serum (Figure 3). Therefore, the glycoprotein solution precipitated with ADA-immobilized beads from the same serum used in the lectin blot was electrophoresed and a silver-stained gel was prepared using a silver staining MS kit (Fujifilm Wako Pure Chemical, 299-58901), and the band near 160 kDa was excised. A peptide mixture was obtained from each excised gel piece through trypsin hydrolysis. The peptide mixture was subjected to LC-MS/MS to acquire MS/MS data. The results of an amino acid sequence database search of the MS/MS data were output as a list of peptide identifications. As a result, Von Willebrand factor was acquired as a candidate protein detected by ADA in cancer patients.

(実施例2-2)rGC2により認識される糖タンパク質の同定
本実施例では、rGC2結合ビーズで膵がん患者および健常者の血清中の糖タンパク質をそれぞれ沈降させ、沈降した糖タンパク質を溶出して、得られた溶出液を電気泳動し、膵がんに固有の糖タンパク質を質量分析により同定した。
(実施例2-1)と同様の手法を実施した。その結果、健常者血清ではrGC2ブロットで反応性は見られなかったが、膵がん血清では250 kDa付近、125 kDa付近、60 kDa付近で反応性が認められた(図4)。そこで、レクチンブロットで使用したものと同じ血清からrGC2固定化ビーズで沈降させた糖タンパク質溶液を、電気泳動して銀染色MSキット(富士フイルム和光純薬、299-58901)を用いて銀染色したゲルを作製し、250 kDa付近、125 kDa付近、60 kDa付近のバンドをそれぞれ切り出した。切り出した各ゲル片から(実施例2-1)と同様の手法で同定を実施した。その結果、分子量の大きい方からフィブロネクチン、α-マクログロブリン、フィブリノーゲンβ鎖がrGC2によりがん患者において検出される候補タンパク質として取得された。

Therefore, a glycoprotein solution precipitated with rGC2-immobilized beads from the same serum used in the lectin blot was electrophoresed to prepare a silver-stained gel using a silver staining MS kit (FUJIFILM Wako Pure Chemical Industries, 299-58901), and bands near 250 kDa, 125 kDa, and 60 kDa were excised. Identification was carried out from each excised gel piece using the same method as in (Example 2-1). As a result, fibronectin, α 2 -macroglobulin, and fibrinogen β chain were obtained as candidate proteins detected by rGC2 in cancer patients in order of molecular weight.

実施例3:糖タンパク質同定結果の妥当性確認
(実施例3-1)ADA固定化ビーズに結合性を示した糖タンパク質に対する、抗フォンウィレブランド因子抗体を用いたウエスタンブロット
本実施例では、膵がん及び健常者血清からADA固定化ビーズに結合性を示した糖タンパク質を電気泳動して、フォンウィレブランド因子に対する抗体とHRP標識2次抗体を用いてウエスタンブロットした。
[サンプル調製]
実施例2で用いたものと同一の健常者および膵がん患者(早期膵がん、後期膵がん)各3ロットずつを、(実施例2-1)と同様の手法でADA固定化ビーズで血清中の糖タンパク質をそれぞれ沈降させた後、溶出し、糖タンパク質溶液を得た。
2―メルカプトエタノールを2× Laemmli Sample Buffer(BIORAD、1610737)で2%(v/v)に希釈し、サンプルバッファーを調製した。上記糖タンパク質溶液1μLをサンプルバッファーで10μLに希釈し、サンプル溶液を調製した。このサンプル溶液を95℃で10分間加熱した。
[電気泳動]
電気泳動槽に泳動用緩衝液と電気泳動用ゲルU-PAGEL H (3-10%)(ATTO、UH-R310)をセットし、各ウェルに前工程で調製したサンプル溶液を全量添加し、定電圧180Vの条件下で75分間泳動した。
[転写]
ガラスから外したゲルを転写バッファー((48mM トリス、39mMグリシン)/20%メタノール)に浸漬し、室温で10分間振とうさせた。同時に転写用ろ紙2枚(BIO-RAD、1703967)とメタノールで活性化したPVDF膜(ATTO、WSE-4051)を転写バッファーに浸漬し、室温で10分間振とうさせた。トランスブロットSDセルにろ紙、PVDF膜、ゲルをセットし、定電圧25Vの条件下で30分間稼働させ、PVDF膜に糖タンパク質を転写させた。
[ウエスタンブロット]
ブロックエース(ケー・エー・シー、UKB80)を1%(w/v)になるように、PBS/0.05% Tween20で希釈し、ブロッキング溶液を調製した。ブロッキング溶液中に転写後のPVDF膜を入れ、室温で1時間振とうさせて、ブロッキングした。
ヒツジ抗ヒトフォンウィレブランド因子抗体(R&D SYSTEMS、AF2764)をブロッキング溶液で0.5μg/mLに希釈し、1次抗体溶液を調製した。1時間振とうさせたPVDF膜に、1次抗体溶液を加えて、室温で1時間振とうさせた。
1時間振とうさせたPVDF膜をPBS/0.05% Tween20で3回洗浄後、ウサギ抗ヒツジIgG抗体(HRP標識)(abcam、ab6747)をブロッキング溶液で5000倍に希釈した2次抗体溶液を加え、室温で30分間振とうさせた。
再度、PVDF膜をPBS/0.05% Tween20で3回洗浄後、ECL Prime(cytiva、RPN2232)をアプライし、室温で5分間反応させ、CCDイメージャー(GE Healthcare、LAS4000)でウエスタンブロット像を撮影した。
その結果、図5に示されるように、フォンウィレブランド因子の分子量250 kDa付近にバンドが見られたことから候補タンパク質はフォンウィレブランド因子であり、同定結果は妥当であることが確認された。またADA固定化ビーズに結合性を示すフォンウィレブランド因子は、健常者血清と比べて膵がん患者血清で有意に強く検出されることが明らかとなった。

[Sample preparation]
Three lots each of healthy subjects and pancreatic cancer patients (early stage pancreatic cancer, late stage pancreatic cancer) identical to those used in Example 2 were used to precipitate glycoproteins in the serum using ADA-immobilized beads in a manner similar to that used in Example 2-1, and then the glycoproteins were eluted to obtain glycoprotein solutions.
2-Mercaptoethanol was diluted to 2% (v/v) with 2x Laemmli Sample Buffer (BIORAD, 1610737) to prepare a sample buffer. 1 μL of the glycoprotein solution was diluted to 10 μL with the sample buffer to prepare a sample solution. This sample solution was heated at 95° C. for 10 minutes.
[Electrophoresis]
A running buffer and an electrophoresis gel U-PAGEL H (3-10%) (ATTO, UH-R310) were placed in the electrophoresis tank, and the entire amount of the sample solution prepared in the previous step was added to each well, followed by electrophoresis at a constant voltage of 180 V for 75 minutes.
[Transcription]
The gel removed from the glass was immersed in transfer buffer ((48 mM Tris, 39 mM glycine)/20% methanol) and shaken at room temperature for 10 minutes. At the same time, two sheets of transfer filter paper (BIO-RAD, 1703967) and a PVDF membrane activated with methanol (ATTO, WSE-4051) were immersed in the transfer buffer and shaken at room temperature for 10 minutes. The filter paper, PVDF membrane, and gel were placed in a Transblot SD cell and operated at a constant voltage of 25 V for 30 minutes to transfer the glycoprotein to the PVDF membrane.
[Western blot]
A blocking solution was prepared by diluting Block Ace (KAC, UKB80) to 1% (w/v) with PBS/0.05% Tween 20. The PVDF membrane after transfer was placed in the blocking solution and shaken at room temperature for 1 hour for blocking.
A sheep anti-human von Willebrand factor antibody (R&D SYSTEMS, AF2764) was diluted with the blocking solution to 0.5 μg/mL to prepare a primary antibody solution. The primary antibody solution was added to the PVDF membrane that had been shaken for 1 hour, and the membrane was then shaken at room temperature for 1 hour.
The PVDF membrane was shaken for 1 hour and then washed three times with PBS/0.05% Tween 20. A secondary antibody solution containing rabbit anti-sheep IgG antibody (HRP-labeled) (abcam, ab6747) diluted 5,000-fold with blocking solution was then added, and the membrane was shaken at room temperature for 30 minutes.
The PVDF membrane was again washed three times with PBS/0.05% Tween 20, after which ECL Prime (cytiva, RPN2232) was applied and reacted at room temperature for 5 minutes, and the Western blot image was photographed with a CCD imager (GE Healthcare, LAS4000).
As a result, as shown in Figure 5, a band was observed near the molecular weight of von Willebrand factor, 250 kDa, confirming that the candidate protein was von Willebrand factor and that the identification results were valid. It was also revealed that von Willebrand factor, which exhibits binding affinity to ADA-immobilized beads, was detected significantly more strongly in the sera of pancreatic cancer patients than in the sera of healthy subjects.

(実施例3-2)rGC2固定化ビーズに結合性を示した糖タンパク質に対する、抗フィブロネクチン抗体を用いたウエスタンブロット

その結果、図6に示されるように、フィブロネクチンの分子量250 kDa付近にバンドが見られたことから候補タンパク質はフィブロネクチンであり、同定結果は妥当であることが確認された。またrGC2固定化ビーズに結合性を示すフィブロネクチンは、健常者血清と比べて膵がん患者血清で有意に強く検出されることが明らかとなった。

As a result, as shown in Figure 6, a band was observed near the molecular weight of 250 kDa of fibronectin, confirming that the candidate protein was fibronectin and the identification results were valid. It was also revealed that fibronectin, which exhibits binding to rGC2-immobilized beads, was detected significantly more strongly in the sera of pancreatic cancer patients than in the sera of healthy subjects.

実施例4:シアル酸結合レクチンに結合性を示すフォンウィレブランド因子の検出
(実施例4-1)MAL固定化ビーズに結合性を示した糖タンパク質に対する、抗フォンウィレブランド因子抗体を用いたウエスタンブロット

実験は(実施例3-1)と同様の手順で実施した。1次抗体としてヒツジ抗ヒトフォンウィレブランド因子抗体(R&D SYSTEMS、AF2764)、2次抗体としてウサギ抗ヒツジIgG抗体(HRP標識)(abcam、ab6747)を使用した。
その結果、図7に示されるように、フォンウィレブランド因子の分子量250 kDa付近にバンドが見られ、ADAと同様にMAL固定化ビーズに結合性を示すフォンウィレブランド因子は、健常者血清と比べて膵がん患者血清で有意に強く検出されることが明らかとなった。

その結果、図8に示されるように、フォンウィレブランド因子の分子量250 kDa付近にバンドが見られ、ADAと同様にSNA固定化ビーズに結合性を示すフォンウィレブランド因子は、健常者血清と比べて膵がん患者血清で有意に強く検出されることが明らかとなった。
(実施例4-1)の結果とも合わせ、シアル酸結合レクチンに結合性を示すフォンウィレブランド因子を検出することで健常者血清と膵がん患者血清に差異を見出すことが可能であることが示唆された。

Combined with the results of Example 4-1, it was suggested that it is possible to find differences between the sera of healthy subjects and those of pancreatic cancer patients by detecting von Willebrand factor, which exhibits binding affinity to sialic acid-binding lectin.

実施例5:レクチンELISAを用いた検体測定
測定に使用した血清検体のリストは以下の通りである。

本実施例では、rGC2を固相化したアビジンプレートとHRP標識抗フィブロネクチン抗体を用いてフィブロネクチン検出用レクチンELISAを作製し、膵がん及び健常者血清検体中のフィブロネクチンを測定した。
[レクチンELISA作製]
アビジンプレート(ブロッキングレスタイプ)(住友ベークライト製。品番:BS-X7603)に、ビオチン化したrGC2をPBSに溶解した液を添加して室温でインキュベートした。
0.1% Tween/PBSでプレート洗浄後、健常者血清と膵がん患者血清それぞれのロットをPBSで100倍希釈し、各ウェルに50μLずつ添加した。
室温でインキュベートし0.1% Tween/PBSでプレート洗浄後、HRP標識抗フィブロネクチン抗体を各ウェルに添加し、室温でインキュベートした。
0.1% Tween/PBSでプレート洗浄後、TMB Solution (富士フイルム和光純薬製。品番:208-17371)を各ウェルに50μL添加して室温でインキュベートした。その後、1N HClを各ウェルに50μL添加した。
[検体測定]
OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値はrGC2により認識される糖鎖構造を持つフィブロネクチンの量に対応する。
その結果、健常者では測定値が低く膵がん患者では測定値が高い傾向が見られた(図9)。すなわち、rGC2により認識される糖鎖構造を持つフィブロネクチンは健常者血清で少なく、膵がん患者血清で多いという傾向が明らかになった。
以上より、フィブロネクチン検出用のHRP標識抗フィブロネクチン抗体-rGC2固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。

After incubation at room temperature and washing the plate with 0.1% Tween/PBS, HRP-labeled anti-fibronectin antibody was added to each well and incubated at room temperature.
After washing the plate with 0.1% Tween/PBS, 50 μL of TMB Solution (manufactured by Fujifilm Wako Pure Chemical Industries, product number: 208-17371) was added to each well and incubated at room temperature, after which 50 μL of 1N HCl was added to each well.
[Sample Measurement]
The values obtained by subtracting the OD 620 nm (second wavelength) measurement value from the OD 450 nm (main wavelength) measurement value were compared for each sample. The measured value corresponds to the amount of fibronectin having a sugar chain structure recognized by rGC2.
As a result, the measured values tended to be low in healthy subjects and high in pancreatic cancer patients (Figure 9). In other words, it was revealed that fibronectin with the glycan structure recognized by rGC2 tended to be low in the serum of healthy subjects and high in the serum of pancreatic cancer patients.
From the above, it was demonstrated that it is possible to distinguish between healthy subjects and pancreatic cancer patients by measurement using HRP-labeled anti-fibronectin antibody-rGC2 solid-phase ELISA for detecting fibronectin.

(実施例5-2)フォンウィレブランド因子検出用のHRP標識抗フォンウィレブランド因子抗体-MAL固相化ELISAを用いた検体測定
本実施例では、MALを固相化したアビジンプレートとHRP標識抗フォンウィレブランド因子抗体を用いてフォンウィレブランド因子検出用レクチンELISAを作製し、膵がん及び健常者血清検体中のフォンウィレブランド因子を測定した。

OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値はMALにより認識される糖鎖構造を持つフォンウィレブランド因子の量に対応する。
その結果、健常者では測定値が低く膵がん患者では測定値が高い傾向が見られた(図10)。すなわち、MALにより認識される糖鎖構造を持つフォンウィレブランド因子は健常者血清で少なく、膵がん患者血清で多いという傾向が明らかになった。
以上より、フォンウィレブランド因子検出用のHRP標識抗フォンウィレブランド因子抗体-MAL固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。また、ステージIBの膵がん患者検体(357)のフォンウィレブランド因子が健常者検体のフォンウィレブランド因子よりも多いことから、ステージIの早期膵がんを見分けうることが示された。

In other words, it was revealed that von Willebrand factor having a glycan structure recognized by MAL tends to be low in the serum of healthy subjects and high in the serum of pancreatic cancer patients.
From the above, it was shown that it is possible to distinguish between healthy subjects and pancreatic cancer patients by measuring with HRP-labeled anti-von Willebrand factor antibody-MAL solid-phase ELISA for detecting von Willebrand factor. In addition, since the von Willebrand factor in stage IB pancreatic cancer patient samples (357) was greater than that in healthy subject samples, it was shown that it is possible to distinguish early stage I pancreatic cancer.

(実施例5-3)フォンウィレブランド因子検出用のHRP標識抗フォンウィレブランド因子抗体-SNA固相化ELISAを用いた検体測定

その結果、健常者では測定値が低く膵がん患者では測定値が高い傾向が見られた(図11)。すなわち、SNAにより認識される糖鎖構造を持つフォンウィレブランド因子は健常者血清で少なく、膵がん患者血清で多いという傾向が明らかになった。
以上より、フォンウィレブランド因子検出用のHRP標識抗フォンウィレブランド因子抗体-SNA固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。また、ステージIBの膵がん患者検体(357)のフォンウィレブランド因子が健常者検体のフォンウィレブランド因子よりも多いことから、ステージIの早期膵がんを見分けうることが示された。

From the above, it was shown that it is possible to distinguish between healthy subjects and pancreatic cancer patients by measuring with HRP-labeled anti-von Willebrand factor antibody-SNA solid-phase ELISA for detecting von Willebrand factor. In addition, since the von Willebrand factor in stage IB pancreatic cancer patient samples (357) was greater than that in healthy subject samples, it was shown that it is possible to distinguish early stage I pancreatic cancer.

(実施例5-1)と同様の手順で実施した。
[検体測定]
OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値はHSAにより認識される糖鎖構造を持つフォンウィレブランド因子の量に対応する。
その結果、健常者では測定値が低く膵がん患者では測定値が高い傾向が見られた(図12)。すなわち、HSAにより認識される糖鎖構造を持つフォンウィレブランド因子は健常者血清で少なく、膵がん患者血清で多いという傾向が明らかになった。
以上より、フォンウィレブランド因子検出用のHRP標識抗フォンウィレブランド因子抗体-HSA固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。

As a result, the measured values tended to be low in healthy subjects and high in pancreatic cancer patients (Figure 12). In other words, it was revealed that von Willebrand factor having a glycan structure recognized by HSA tends to be low in the serum of healthy subjects and high in the serum of pancreatic cancer patients.
From the above, it was demonstrated that it is possible to distinguish between healthy subjects and pancreatic cancer patients by measurement using HRP-labeled anti-von Willebrand factor antibody-HSA solid-phase ELISA for detecting von Willebrand factor.

(実施例5-5)フォンウィレブランド因子検出用のHRP標識SNA-抗フォンウィレブランド因子抗体固相化ELISAを用いた検体測定
本実施例では、抗フォンウィレブランド因子抗体を固相化したアビジンプレートとHRP標識SNAを用いてフォンウィレブランド因子検出用レクチンELISAを作製し、膵がん及び健常者血清検体中のフォンウィレブランド因子を測定した。
[レクチンELISA作製]
アビジンプレート(ブロッキングレスタイプ)(住友ベークライト製。品番:BS-X7603)に、ビオチン化した抗フォンウィレブランド因子抗体をPBSに溶解した液を添加して室温でインキュベートした。
0.1% Tween/PBSでプレート洗浄後、健常者血清と膵がん患者血清それぞれのロットをPBSで100倍希釈し、各ウェルに50μLずつ添加した。
室温でインキュベートし0.1% Tween/PBSでプレート洗浄後、HRP標識SNAを各ウェルに添加し、室温でインキュベートした。
0.1% Tween/PBSでプレート洗浄後、TMB Solution (富士フイルム和光純薬製。品番:208-17371)を各ウェルに50μL添加して室温でインキュベートした。その後、1N HClを各ウェルに50μL添加した。
[検体測定]
OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値は血清中フォンウィレブランド因子のうち、SNAにより認識される糖鎖構造を持つフォンウィレブランド因子の量に対応する。
その結果、健常者では測定値が低く膵がん患者では測定値が高い傾向が見られた(図13)。すなわち、SNAにより認識される糖鎖構造を持つフォンウィレブランド因子は健常者血清で少なく、膵がん患者血清で多いという傾向が明らかになった。
以上より、フォンウィレブランド因子検出用のHRP標識SNA-抗フォンウィレブランド因子抗体固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。また、ステージIBの膵がん患者検体(357)のフォンウィレブランド因子が健常者検体のフォンウィレブランド因子よりも多いことから、ステージIの早期膵がんを見分けうることが示された。

[Lectin ELISA Preparation]
A solution of biotinylated anti-von Willebrand factor antibody dissolved in PBS was added to an avidin plate (blocking-less type) (manufactured by Sumitomo Bakelite, product number: BS-X7603), and incubated at room temperature.
After washing the plate with 0.1% Tween/PBS, each lot of serum from healthy subjects and pancreatic cancer patients was diluted 100-fold with PBS, and 50 μL of each was added to each well.
After incubation at room temperature and washing the plate with 0.1% Tween/PBS, HRP-labeled SNA was added to each well and incubated at room temperature.
After washing the plate with 0.1% Tween/PBS, 50 μL of TMB Solution (manufactured by Fujifilm Wako Pure Chemical Industries, product number: 208-17371) was added to each well and incubated at room temperature, after which 50 μL of 1N HCl was added to each well.
[Sample Measurement]
The values obtained by subtracting the OD 620 nm (minor wavelength) measurement value from the OD 450 nm (main wavelength) measurement value were compared for each sample. The measurement value corresponds to the amount of von Willebrand factor having a sugar chain structure recognized by SNA among the von Willebrand factor in serum.
As a result, the measured values tended to be low in healthy subjects and high in pancreatic cancer patients (Figure 13). In other words, it was revealed that von Willebrand factor having a glycan structure recognized by SNA tends to be low in the serum of healthy subjects and high in the serum of pancreatic cancer patients.
From the above, it was shown that it is possible to distinguish between healthy subjects and pancreatic cancer patients by measuring using HRP-labeled SNA-anti-von Willebrand factor antibody solid-phase ELISA for detecting von Willebrand factor. In addition, since the von Willebrand factor in stage IB pancreatic cancer patient samples (357) was greater than that in healthy subject samples, it was shown that it is possible to distinguish early stage I pancreatic cancer.

(実施例5-6)α-マクログロブリン検出用のHRP標識抗α-マクログロブリン抗体-TJA II固相化ELISAを用いた検体測定
本実施例では、TJA IIを固相化したアビジンプレートとHRP標識抗α-マクログロブリン抗体を用いてα-マクログロブリン検出用レクチンELISAを作製し、膵がん及び健常者血清検体中のα-マクログロブリンを測定した。
[レクチンELISA作製]
(実施例5-1)と同様の手順で実施した。
[検体測定]
OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値はTJA IIにより認識される糖鎖構造を持つα-マクログロブリンの量に対応する。
その結果、健常者では測定値が高く膵がん患者では測定値が低い傾向が見られた(図14)。すなわち、TJA IIにより認識される糖鎖構造を持つα-マクログロブリンは健常者血清で多く、膵がん患者血清で少ないという傾向が明らかになった。
以上より、α-マクログロブリン検出用のHRP標識抗α-マクログロブリン抗体-TJA II固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。

The values obtained by subtracting the OD 620 nm (minor wavelength) measurement value from the OD 450 nm (main wavelength) measurement value were compared for each sample. The measured value corresponds to the amount of α 2 -macroglobulin having a sugar chain structure recognized by TJA II.
As a result, the measured values tended to be high in healthy subjects and low in pancreatic cancer patients (Figure 14). In other words, it was revealed that α2 -macroglobulin, which has a glycan structure recognized by TJA II, tends to be abundant in the sera of healthy subjects and to be abundant in the sera of pancreatic cancer patients.
From the above, it was demonstrated that it is possible to distinguish between healthy subjects and pancreatic cancer patients by performing measurement using HRP-labeled anti-α 2 -macroglobulin antibody-TJA II solid-phase ELISA for detecting α 2 -macroglobulin.

(実施例5-7)α-マクログロブリン検出用のHRP標識TJA II-抗α-マクログロブリン抗体固相化ELISAを用いた検体測定
本実施例では、抗α-マクログロブリン抗体を固相化したアビジンプレートとHRP標識TJA IIを用いてα-マクログロブリン検出用レクチンELISAを作製し、膵がん及び健常者血清検体中のα-マクログロブリンを測定した。
[レクチンELISA作製]
(実施例5-5)と同様の手順で実施した。
[検体測定]
OD450nm(主波長)の測定値からOD620nm(副波長)の測定値を差し引いた値を各サンプルで比較した。測定値は血清中α-マクログロブリンのうちTJA IIにより認識される糖鎖構造を持つα-マクログロブリンの量に対応する。
その結果、健常者では測定値が高く膵がん患者では測定値が低い傾向が見られた(図15)。すなわち、TJA IIにより認識される糖鎖構造を持つα-マクログロブリンは健常者血清で多く、膵がん患者血清で少ないという傾向が明らかになった。
以上より、α-マクログロブリン検出用のHRP標識TJA II-抗α-マクログロブリン抗体固相化ELISAを用いて測定することで、健常者と膵がん患者を見分けうることが示された。また、ステージIBの膵がん患者検体(357)のα-マクログロブリンが健常者検体のα-マクログロブリンよりも少ないことから、ステージIの早期膵がんを見分けうることが示された。

The values obtained by subtracting the OD 620 nm (minor wavelength) values from the OD 450 nm (main wavelength) values were compared for each sample. The measured values correspond to the amount of α2 -macroglobulin in serum that has a glycan structure recognized by TJA II.
As a result, the measured values tended to be high in healthy subjects and low in pancreatic cancer patients (Figure 15). In other words, it was revealed that α2 -macroglobulin, which has a glycan structure recognized by TJA II, tends to be abundant in the sera of healthy subjects and to be abundant in the sera of pancreatic cancer patients.
These results suggest that the use of HRP-labeled TJA II-anti- α2 -macroglobulin antibody solid-phase ELISA for detecting α2 -macroglobulin makes it possible to distinguish between healthy subjects and pancreatic cancer patients. In addition, the amount of α2 -macroglobulin in stage IB pancreatic cancer patient specimens (357) was less than that in healthy subject specimens, suggesting that early stage I pancreatic cancer can be distinguished.
