IX73(オリンパス社)を用いた顕微鏡観察でウイルスによる細胞変性効果が認められず、かつ、培養上清のRT-qPCRでウイルスゲノムが検出できなかった場合にSARS-CoV-2を不活化したと判断した。RT-qPCRではCt(Threshold Cycle)値が30未満となった場合にウイルスが増殖したと判断した。
(protocol)
1. VeroC1008 was plated on a 96-well plate to 100% confluency.
2. Component (a) or component (a') (comparative component for component (a)) was diluted with autoclaved tap water to prepare evaluation solutions shown in Table 1. The pH of the evaluation solution in Table 1 was not adjusted and was taken as is.
3. 27 μL of evaluation solution and 3 μL of SARS-CoV-2 solution (equivalent to about 4×10 5 TCID 50 ) were mixed.
4. The reaction was allowed to proceed for 10 minutes at room temperature.
270 μL of 5.2% FBS medium was added to stop the reaction.
6.5. 20 μL of the solution was added to and mixed with 96 plates of Vero C1008 that had been pre-exchanged with 180 μL of 2% FBS medium.
7. Infected for 1 hour at 37°C/5% CO2 .
Washed twice with 200 μL of 8.2% FBS medium.
9. Incubated for 3 days at 37°C/5% CO2 .
10. SARS-CoV-2 inactivation was determined by viral cytopathic effect and RT-qPCR.
SARS-CoV-2 was inactivated when no cytopathic effect due to the virus was observed by microscopic observation using IX73 (Olympus) and the viral genome could not be detected by RT-qPCR of the culture supernatant. It was judged. When the Ct (Threshold Cycle) value was less than 30 in RT-qPCR, it was determined that the virus had grown.

(a) Use as an enveloped virus inactivator containing, as an active ingredient, an arylsulfonic acid or a salt thereof optionally substituted with 1 to 3 hydrocarbon groups having 1 to 5 carbon atoms.
