Claims (23)
- 配列番号3に少なくとも80%のアミノ酸配列同一性を有するβ−グルコシダーゼ1(BGL1)変異体において、
(a)前処理済みトウモロコシ茎葉(PCS)加水分解活性、
(b)セロビアーゼ活性、
(c)タンパク質発現、
(d)アンモニアで前処理したトウモロコシ穂軸(CC)の存在下でのセロビアーゼ活
性又はCC加水分解活性のいずれかにより測定されるβ−グルコシダーゼ活性、
(e)熱安定性、
(f)リン酸膨潤セルロース(PASC)加水分解活性、及び
(g)グルコース存在下での加水分解活性、
からなる群から選択される少なくとも2つの活性が配列番号3のアミノ酸配列を有する野生型β−グルコシダーゼ1(BGL1)よりも改良されており、
上記BGL1変異体が配列番号3のI567、Q226、E170、N263、S683、及びN238から選択される位置に対応する位置における置換を含むBGL1変異体。 - 請求項1に記載のBGL1変異体において、
(1)置換位置がQ226である場合、前記置換はQ226Y又はC226Fであり、
(2)置換位置がE170である場合、前記置換はE170Fであり、
(3)置換位置がN263である場合、前記置換はN263C又はN263Tであり、
(4)置換位置がS683である場合、前記置換はS683Wであり、
(5)置換位置がN238である場合、前記置換はN238Fである、BGL1変異体。 - 請求項2に記載のBGL1変異体において、該変異体が(1)の置換を有する場合、更に配列番号3のE170、T242、S312、N369、G372、V603、P661、及び/又はT666に対応する位置における1つ以上の置換を更に含むBGL1変異体。
- 請求項3に記載のBGL1変異体において、前記置換がE170F、T242S,S312Y、N369Y、G372A、V603G、P661F、及び/又はT666CであるBGL1変異体。
- 請求項2に記載のBGL1変異体において、該変異体が(2)の置換を有する場合、更に配列番号3のQ226、T242,S312、N369、G372、V603、P661、及び/又はT666に対応する位置における1つ以上の置換を更に含むBGL1変異体。
- 請求項5に記載のBGL1変異体において、前記置換がQ226Y、T242S,S312Y、N369Y、G372A、V603G、P661F、及び/又はT666CであるBGL1変異体。
- 請求項2に記載のBGL1変異体において、該変異体が(3)の置換を有する場合、更に配列番号3のK345、N369、G372、K428、P661、及び/又はS683に対応する位置における1つ以上の置換を更に含むBGL1変異体。
- 請求項7に記載のBGL1変異体において、前記置換がK345E、N369E、N369T、G372A、K428N、P661E、及び/又はS683WであるBGL1変異体。
- 請求項2に記載のBGL1変異体において、該変異体が(4)の置換を有する場合、更に配列番号3のN263、K345、N369、G372、K428、及び/又はP661、に対応する位置における1つ以上の置換を更に含むBGL1変異体。
- 請求項9に記載のBGL1変異体において、前記置換がN263C、K345E、N369E、N369T、G372A、K428N、P661E、P661L、及び/又はS683WであるBGL1変異体。
- 請求項2に記載のBGL1変異体において、該変異体が(5)の置換を有する場合、更に配列番号3のR179、N238、N264、N265、N369、D370、及び/又はK656に対応する位置における1つ以上の置換を更に含むBGL1変異体。
- 請求項11に記載のBGL1変異体において、前記置換がR179V、N238F、N264M、N265M、N369I、D370W、及び/又はK656RであるBGL1変異体。
- 請求項1に記載のBGL1変異体において、
(i)置換がI567Qである場合、改良された活性が(a)および(b)である、
(ii)置換がI567Vである場合、改良された活性が(b)、(c)および(e)である、
