ヒト皮膚角層を細粉化したものを水に浸し、アミノ酸や尿素などの水溶性成分を完全に除去したものを凍結乾燥して水分を完全に除去した角層をケラチン試料とした。試験はケラチン試料10mgを各サンプル溶液1g中に浸し、5℃、4時間放置したのち、表面を洗浄して、凍結乾燥したものを水分量0wt%試料とし、それぞれ各水分量に調節した。結合水量の測定は、Journal of Investigative Dermatology 87, 574-576,
1986に準じてDSC測定を用いて行った。この結果を表1に示す。コントロールとして用いた精製水の結果を100%として各サンプルのパーセンテージを算出した。
なお、本試験では便宜のためにヒト皮膚角層をケラチン試料として結合水量の評価をしたが、爪および毛髪・皮膚を構成しているケラチンタンパク質を対象とした試験でも、同様の結果が得られるものと推定できる。

なお、本試験では便宜のために市販のケラチン試薬を用いて溶解力の評価をしたが、爪および毛髪・皮膚を構成しているケラチンタンパク質を対象とした試験でも、同様の結果が得られるものと推定できる。
