(タンパク質との反応性試験)
CAPD液の加熱滅菌により生成するGDPsは細胞障害性を示すのみならず、腹膜組織構成タンパク質と結合し、AGE(Advanced Glycation Endproducts)を形成する。

CAPD療法においても腹膜組織へのAGEsの蓄積が確認されており、腹膜劣化を抑えるために、AGEsの形成力が弱い、即ち、反応性の高いGDPsが少ないCAPD液が必要とされている。

このような背景から、発明者らは、生体適合性の指標とした、3,4-DGEのタンパク質との反応性を調べた。
(実験方法)
モデルタンパクとしてウシ血清アルブミン(BSA)を使用した。BSA(10mg/mL)をPBS(pH7.4)に溶解させた。これに各種のGDPsを30mmol/Lの濃度で添加して37℃で24時間反応させた。BSAの変性(AGEs化)は、反応液の蛍光強度(励起波長360nm、蛍光波長430nm)で評価した。

(結果)
図13に示すように、3,4-DGEを加えたBSA溶液は、他のGDPsに比べて強い蛍光を示した。この結果から3,4-DGEはタンパク質との反応性に富み、強力なAGEsの前駆体となるGDPsである事が分かった。
