ポリマー(25% w/w の50/50ラクチド−コ−グリコリドコポリマー、固有粘度、0.59)を最小量のアセトンに溶解する。純粋なポリエチレングリコール400(PEG400)をそのポリマー溶液に添加する。その溶液を攪拌して均一な混合物を生成する。常時攪拌しながら60〜75℃に加熱することによりその混合物からアセトンを減圧留去する。空の製剤を60〜75℃で一晩減圧オーブン中に静置して確実に完全にアセトンを除去する。その結果得られた製剤は、粘液様のコンシステンシーを有するマトリクスであった。3種の濃度のオキシテトラサイクリン塩基(10、20、または30% w/w)をその空の製剤に添加し、十分に混合して確実にその製剤中に該薬物を均一に分配させる。その薬物を充填した製剤からの薬物の放出は、抗酸化剤としてリン酸ナトリウムを含有する等張リン酸緩衝液中、37℃で実施した。図6は、上述の組成で調製した製剤から放出されたオキシテトラサイクリンの累積量を示している。製剤中において薬物の割合が10〜30% w/wまで増加すると、360時間の終了時に放出された薬物累積量が増えた。この増加は、薬物充填が多くなるにつれて、製剤の表面上の放出可能な薬物が増えたために起こった。更に、10% w/wの薬物充填に比べて30% w/wの薬物充填では、製剤と溶出液との間の薬物濃度勾配がより高くなることが予想される。

3種の濃度(10、20、または25% w/w)のポリマー(50/50ラクチド−コ−グリコリドコポリマー、固有粘度、0.59)を最小量のアセトンに溶解する。純粋なPEG400(90、80、または75% w/w)をそのポリマー溶液に添加する。その溶液を攪拌して均一な混合物を生成する。常時攪拌しながら60〜75℃に加熱することによりその混合物からアセトンを減圧留去する。空の製剤を60〜75℃で一晩減圧オーブン中に静置して確実に完全にアセトンを除去する。その結果得られた製剤は、種々の粘度またはコンシステンシーを有するマトリクスであった。ポリマーが25%の製剤は、ポリマーが10%のものよりも顕著に粘度が高かった。オキシテトラサイクリン塩基(20% w/w)を各空の製剤に添加し、十分に混合して確実にその製剤中に該薬物を均一に分配させる。その薬物を充填した製剤からの薬物の放出は、抗酸化剤としてリン酸ナトリウムを含有する等張リン酸緩衝液中、37℃で実施した。図8は、上述の組成で調製した製剤から放出されたオキシテトラサイクリンの累積量を示している。製剤中のポリマーの割合が25%から10%まで減少すると、薬物の放出が劇的に増加することが図から明らかである。これは、ポリマーの濃度の25%から10%への減少と、それに相応した可塑剤の濃度の75%から90%への増加の結果、ガラス転移温度、粘度が減少して、製剤中のポリマー鎖の流動性が上昇したためである。それゆえ、ポリマーを10%含む製剤は、ポリマーを25%用いて調製されたものに比べてマトリクスからの薬物の拡散に対する抵抗が顕著に低くなった。
