<P>PROBLEM TO BE SOLVED: To provide a detergent composition which is low irritant and can decompose and remove a cell and a biofilm as a bacterial metabolite by using an enzyme and an anionic surfactant and a method for evaluating bacteria-removing power and deterging power of a detergent composition on stains in the case that clothes, kitchens and bathtubs are left stained. <P>SOLUTION: The detergent composition comprises the enzyme and the anionic surfactant, and has a biofilm decomposition activity of at least 40 FU (cloth biofilm decomposition activity) or at least 40 HU (hard surface biofilm decomposition activity). The detergent composition decomposes and removes the biofilm that causes malodors from clothes, kitchens, bathtubs or the like left stained. 本発明は、衣料用及び硬質表面用に適した洗浄剤組成物及び除菌・洗浄力評価方法に関する。

【従来の技術】
様々な生活の場面で、人に不具合や不都合を与える菌類を除菌する方法としては、殺菌剤を使用する方法や漂白剤を使用する方法が知られている(例えば、特許文献1、2参照)。また、菌類を除去する又は、バイオフィルムを分解するために酵素を利用したものとして、微生物の細胞壁や細胞膜に作用する酵素、抗菌性タンパク質を配合したコンタクトレンズの洗浄及び消毒用組成物(例えば、特許文献3参照)、バイオフィルムの酵素的処理方法(例えば、特許文献4)が知られている。

さらに、出願人は特願2001−245988号において、界面活性剤を酵素と組み合わせることにより菌活性阻害及び汚れ分解作用により洗濯後における繊維類の乾燥時の悪臭発生を抑制することを既に報告している。一方、毎日洗濯ができず、汚れた衣類を放置した場合には、衣類に付着した汚れと菌の相乗作用により、悪臭が発生することや、同様に台所や浴室の汚れを放置することにより悪臭が発生することが知られている。この場合、生活環境中に存在する菌は菌自身と菌が代謝生産する代謝物と一体となり粘性を持ち、洗浄除去しにくいバイオフィルムを形成している場合が多くみられ、ここでは菌はバイオフィルム内に保護される形で生息しており、バイオフィルムを形成している菌は除菌・殺菌剤等からの作用を受けにくくなることから、このようにしてできた汚れ(バイオフィルム)は落ちにくいものであった。そこでバイオフィルムを除去し、菌を除菌しやすくできるようなものが望まれていた。

また、上述したような、汚れた衣類や汚れた台所、浴槽を放置した場合における汚れに対する、洗浄剤組成物の除菌・洗浄力評価方法について、より実際の効果に近い評価ができるものが望まれていた。

【発明が解決しようとする課題】
本発明は上記事情に鑑みなされたもので、酵素及びアニオン界面活性剤を用いることにより刺激が低く効率的に菌体及び菌代謝物であるバイオフィルムを分解・除去できる洗浄剤組成物、及び汚れた衣類や汚れた台所、浴槽を放置した場合における汚れに対する、洗浄剤組成物の除菌・洗浄力評価方法について、より実際の効果に近い評価方法を提供することを目的とする。

【課題を解決するための手段及び発明の実施の形態】
本発明者は、上記目的を達成するため鋭意検討した結果、特定の酵素及びアニオン界面活性剤を用いて、バイオフィルムを処理することにより、菌体及び代謝物であるバイオフィルムを効率的に分解・除去できること、布帛又は硬質表面を有する物体を汚れ成分及び培地に接触させて菌類を培養することにより、バイオフィルムの付着した布帛又は硬質表面を有する物体を調製し、前記バイオフィルムの付着した布帛又は硬質表面を有する物体を洗浄剤組成物で洗浄し、洗浄後のバイオフィルム分解量を測定することにより、汚れた衣類、台所及び浴槽等の汚れを放置した場合の、洗浄剤組成物の除菌・洗浄力評価方法について、より実際の効果に近い評価ができることを見出し、本発明をなすに至ったものである。

従って、本発明は、酵素及びアニオン界面活性剤を含有し、バイオフィルム分解活性が40FU(布バイオフィルム分解活性)以上又は40HU(硬質表面バイオフィルム分解活性)以上であることを特徴とする洗浄剤組成物、及び布帛又は硬質表面を有する物体を汚れ成分培地に接触させて菌類を培養することにより、バイオフィルムの付着した布帛又は硬質表面を有する物体を調製し、前記バイオフィルムの付着した布帛又は硬質表面を有する物体を洗浄剤組成物で洗浄し、洗浄後のバイオフィルム分解量を測定することを特徴とする洗浄剤組成物の除菌・洗浄力評価方法を提供する。

以下、本発明につき更に詳しく説明する。
本発明の構成は以下のとおりである。
(1)酵素及びアニオン界面活性剤を含有し、バイオフィルム分解活性が衣料用では、40FU(布バイオフィルム分解活性)以上、又は硬質表面用では40HU(硬質表面バイオフィルム分解活性)以上であることを特徴とする洗浄剤組成物。
(2)酵素がオキシドレダクターゼ(酸化還元酵素:Oxidoreductases、EC(Enzyme Class(EC)1群に属する酵素、基質Aを酸化し基質Bを還元する酵素))、トランスフェラーゼ(転移酵素:Transferases、EC2群に属する酵素、基質A−XからXを他の基質Bに移してB−Xを作る酵素))、ヒドロラーゼ(加水分解酵素:Hydrolases、EC3群に属する酵素、基質A−BをHOで、A−OHとBHに分解する酵素)、リアーゼ(Lyases、EC4群に属する酵素、基質A−BをAとBに分解する酵素、加水分解酵素とは異なり、他分子の関与なし))、イソメラーゼ(異性化酵素:Isomerases、EC5群に属する酵素、分子の分子内原子配置を変化させる酵素)、及びリガーゼ(Ligases、EC6群に属する酵素、分解反応と共役して二つの分子の結合反応を触媒する酵素)からなる群から選ばれる1種以上の酵素であることを特徴とする(1)に記載の洗浄剤組成物。
(3)酵素が、上記(2)に記載の酵素2種類以上を含有することを特徴とする上記(1)に記載の洗浄剤組成物。
(4)酵素が、ヒドロラーゼ(加水分解酵素)及びオキシドレダクターゼ(酸化還元酵素)から選ばれる2種以上を含有する上記(3)に記載の洗浄剤組成物。
(5)酵素が、上記(2)に記載のヒドロラーゼ(加水分解酵素)のうちプロテアーゼ、セルラーゼ、アミラーゼ、リパーゼ、及びオキシドレダクターゼ(酸化還元酵素)のうちラッカーゼ、ペルオキシダーゼから選ばれる2種以上を含有する上記(3)に記載の洗浄剤組成物。
(6)アニオン界面活性剤を3〜30質量%、特に15〜30質量%含有することを特徴とする上記(1)〜(5)のいずれかに記載の洗浄剤組成物。
(7)アニオン界面活性剤として、ベンゾシクロアルカン誘導体の含有量が3質量%未満の直鎖アルキルベンゼンスルホン酸塩3〜30質量%を含有することを特徴とする上記(6)に記載の洗浄剤組成物。
(8)アニオン界面活性剤が、上記直鎖アルキルベンゼンスルホン酸塩の他に、アルファオレフィンスルホン酸塩、アルキル硫酸エステル塩、ポリオキシアルキレンアルキルエーテル硫酸塩、アルファスルホ脂肪酸メチルエステル塩、高級脂肪酸塩、及び二級アルカンスルホン酸塩のアルカリ金属塩からなる群から選ばれる1種以上のアニオン界面活性剤を含有することを特徴とする上記(6)に記載の洗浄剤組成物。
(9)アニオン界面活性剤が、上記直鎖アルキルベンゼンスルホン酸塩の他に、アルファオレフィンスルホン酸塩、アルキル硫酸エステル塩、ポリオキシアルキレンアルキルエーテル硫酸塩、アルファスルホ脂肪酸メチルエステル塩、高級脂肪酸塩、及び二級アルカンスルホン酸塩のアルカリ金属塩からなる群から選ばれる2種以上のアニオン界面活性剤を含有することを特徴とする上記(8)に記載の洗浄剤組成物。
(10)アニオン界面活性剤の対イオンが、ナトリウム、カリウム、モノエタノールアミン、ジエタノールアミン、又はトリエタノールアミンであることを特徴とする上記(1)〜(9)のいずれかに記載の洗浄剤組成物。
(11)さらに、ノニオン界面活性剤を1〜40質量%、特に1〜20質量%含有することを特徴とする上記(1)〜(10)のいずれかに記載の洗浄剤組成物。
(12)ノニオン界面活性剤が、ポリオキシエチレンアルキルエーテル、アルコールEO/PO付加物、オキシアルキレンの付加モル数が3以上の脂肪酸ポリオキシアルキレンアルキルエーテル(MEE)、オキシアルキレンの付加モル数が3以上の脂肪酸ポリオキシアルキレンエーテル(FAE)及び脂肪酸アルカノールアミドからなる群から選ばれる1種又は2種以上のノニオン界面活性剤であることを特徴とする上記(11)に記載の洗浄剤組成物。
(13)さらに、アミノカルボン酸誘導体型カルシウム捕捉剤、アルミノケイ酸塩(A型ゼオライト、P型ゼオライト)、ポリカルボン酸/塩、及びリン酸塩からなる群から選ばれる1種又は2種以上のカルシウム捕捉剤を含有することを特徴とする上記(1)〜(12)のいずれかに記載の洗浄剤組成物。
(14)アミノカルボン酸誘導体型カルシウム捕捉剤が、アミノメチルグリシンジ酢酸塩、β−アラニンジ酢酸塩、イミノジコハク酸塩、ヒドロキシイミノジコハク酸塩(HIDS)、セリンジ酢酸塩、イソセリンジ酢酸塩、アスパラギン酸ジ酢酸塩(ASDA)、エチレンジアミン四酢酸塩(EDTA)、ニトリロトリ酢酸塩(NTA)、ヒドロキシエチレンジアミン5酢酸塩(HEDTA)、及びジエチレントリアミン5酢酸塩(DTPA)からなる群から選ばれる1種又は2種以上であることを特徴とする上記(13)に記載の洗浄剤組成物。
(15)ポリカルボン酸及び/又はその塩が、ポリアクリル酸、マレイン酸とアクリル酸の共重合体、及び多糖類系ポリカルボキシレートの高分子電解質からなる群から選ばれる1種又は2種以上であることを特徴とする上記(13)又は(14)に記載の洗浄剤組成物。
(16)リン酸塩が、トリポリリン酸塩、オルソリン酸塩、ピロリン酸塩、メタリン酸塩、ヘキサメタリン酸塩、及びフイチン酸からなる群から選ばれる1種又は2種以上であることを特徴とする上記(13)〜(15)のいずれかに記載の洗浄剤組成物。
(17)アニオン界面活性剤、ノニオン界面活性剤及び上記(5)に記載の酵素を含有する洗浄剤組成物。
(18)アニオン界面活性剤、ノニオン界面活性剤、カルシウム捕捉剤及び上記(5)に記載の酵素を含有する洗浄剤組成物。

本発明の組成物に用いられる酵素としては、オキシドレダクターゼ(酸化還元酵素)、トランスフェラーゼ(転移酵素)、ヒドロラーゼ(加水分解酵素)、リアーゼ、イソメラーゼ(異性化酵素)、及びリガーゼ等が挙げられる。

ヒドロラーゼ(加水分解酵素)としては、糖質分解酵素、タンパク分解酵素、脂質分解酵素等が挙げられ、糖質分解酵素としては、アミラーゼ、セルラーゼ、デキストラナーゼ、グルコシダーゼ、ガラクトシダーゼ、マンノシダーゼ、アガラーゼ、ラクトーゼ、ムタナーゼ、リゾチーム、キチナーゼ、キトサナーゼ等が挙げられる。市販品としては、アミラーゼの「ターマミル」、「デュラミル」(いずれもノボザイムズ製)、「α−アミラーゼ」(生化学工業製)、「プライマーゼ LC」、「プライマーゼ HT」(いずれもエイチビィアイ製)、セルラーゼの「セルザイム」、「ケアザイム」(いずれもノボザイムズ製)、デキストラナーゼの「デキストラナーゼ L「アマノ」」(天野エンザイム製)が好適である。

タンパク分解酵素としては、ペプシン、トリプシン、キモトリプシン、コラゲナーゼ、ゼラチナーゼ、ケラチナーゼ、エラスターゼ、スブチリシン、パパイン、プロメリン、カルボキシペプチダーゼA及びB、アミノペプチダーゼ、サーモリシン、アクロモペプチダーゼ等が挙げられる。市販品としては、「サビナーゼ12T」、「サビナーゼ16L」、「エスペラーゼ4T」、「エバラーゼ8T」、「デュラザイム6T」、「アルカラーゼ2T」(いずれもノボザイムズ製)、ズブチリシンカールズベルグ(Subtilisin Carlsberg、Type VIII:Bacterial、P5380)(シグマ製)、「アルカリプロテアーゼ」(生化学工業製)が好ましい。さらに、「サビナーゼ12T」、「サビナーゼ16L」、「エスペラーゼ4T」、「エバラーゼ8T」、「ズブチリシン カールズベルグ(Subtilisin Carlsberg)」、「アルカリプロテアーゼ」が好ましい。

脂質分解酵素としては、リパーゼ類、植物リパーゼ類、クチナーゼ、ホスホリパーゼ類が挙げられる。なかでも、微生物起源のシュードモナス属、アスペルギルス属、フミコラ属、及びアクロモベクター属から誘導されるリパーゼが好ましく、特に、「リポラーゼ100T」、「リポラーゼ100L」、「リポラーゼウルトラ50T」(ノボザイムズ製)、「アルカリリパーゼ」(生化学工業製)が好ましい。その他の加水分解酵素としては、アルカリホスファターゼ等が挙げられる。

リアーゼとしては、アルドラーゼ、セリンデヒドラターゼ等が挙げられ、イソメラーゼ(異性化酵素)としては、UDP−グルコース−4−エピメラーゼ、グルコースリン酸イソメラーゼ等が挙げられる。リガーゼとしては、リジン−tRNAシンテターゼ、グルタミンシンテターゼ、DNAリガーゼ等が挙げられる。これら酵素は1種を単独で又は2種以上を適宜組み合わせて使用することができる。

酵素の配合量は、特に制限されるものではなく、その種類によって適宜選択できるが、通常、組成物中に、酵素タンパク質として0.0001〜2質量%、好ましくは0.001〜0.2質量%、より好ましくは0.005〜0.1質量%となるように配合すると好適である。また、上記市販の酵素造粒物としては、通常、組成物中に0.001〜20質量%、0.01〜10質量%、より好ましくは0.05〜5質量%となるように配合すると、好適である。上記酵素の配合量が少なすぎると、本発明の目的とするバイオフィルムの分解・除去、悪臭抑制効果が十分に得られ難くなる場合があり、多すぎると、それ以上の配合による効果の増強が得られず、不経済となる場合がある。

本発明のアニオン界面活性剤は、特に制限されないが、例えば、アルキルベンゼンスルホン酸塩、アルファオレフィンスルホン酸塩、アルキル硫酸塩、アルケニル硫酸塩、アルキルエーテル硫酸塩、アルケニルエーテル硫酸塩、アルキルエーテルカルボン酸塩、アルケニルエーテルカルボン酸塩、α−スルホ脂肪酸塩、α−スルホ脂肪酸エステル塩、石鹸等が挙げられる。これらのアニオン界面活性剤は、ナトリウム塩、カリウム塩等のアルカリ金属塩が好ましい。アニオン界面活性剤としては、直鎖のアルキルベンゼンスルホン酸塩が好ましい。

直鎖アルキルベンゼンスルホン酸塩の製造に用いられる直鎖アルキルベンゼンは、通常軽灯油を分留して得られた直鎖パラフィンを出発物質として用い、このものを塩素化パラフィン法もしくはオレフィン法と呼ばれる手法でハロゲンを導入したのちベンゼンを付加することにより合成することができる。この直鎖アルキルベンゼンは混合物で得られ、アルキル基の炭素数は8〜22のものが好ましく、10〜14のものが更に好ましい。また、ベンゼン置換位置は直鎖パラフィンの2位から7位まで分布する。直鎖アルキルベンゼンスルホン酸塩は、上記直鎖アルキルベンゼンに例えばSOガスを吹き込んでスルホン化し、更に中和することにより得られるものであるが、入手し得る商業ベースの直鎖アルキルベンゼンスルホン酸塩は、アルキル基の炭素数・ベンゼン置換位置とも分布を持ち、かつベンゾシクロアルカン誘導体(スルホン化物も含む)等の副生物の不純分を含むものである。

直鎖アルキルベンゼンスルホン酸塩としては、バイオフィルムに対し浸透性が良く、酵素との併用によりその除去効果をさらに向上させる点で、ベンゾシクロアルカン誘導体の含有量が3質量%未満である直鎖アルキルベンゼンスルホン酸塩であることが好ましい。この直鎖アルキルベンゼンスルホン酸塩を得る方法としては、直鎖アルキルベンゼンを精製してベンゾシクロアルカン誘導体の含有量を低減させたものをスルホン化し、更に必要により中和してベンゾシクロアルカン誘導体の含有量を3質量%未満とする方法で得ることができる。ベンゾシクロアルカン誘導体としては、インダン、テトラリンもしくはその誘導体等のベンゾシクロアルカン誘導体が含まれる。

菌及び培地としては、特に限定されるものではなく、布帛及び硬質表面上でバイオフィルムを形成する菌類及び培地であればよい。これらの菌としては、エシェリキア、シュードモナス、アシネトバクター、クレブシエラ、コリネバクテリウム、ミクロコッカス、バチルス、スタフィロコッカス属に属し、物体に付着し、粘性のある代謝物を産生する能力のあるものが挙げられる。好ましくは、エシェリキア コリ、シュードモナス アエルギノーサ、シュードモナス ランデンシス、シュードモナス ペルツシノゲーナ、シュードモナス フルオレッセンス、シュードモナス プチダ、スタフィロコッカス アウレルス、スタフィロコッカス エピデルミディスが好ましい。より好ましくは、スタフィロコッカス エピデルミディスが好ましい。その中でもスタフィロコッカス エピデルミディスATCC12228が好ましい。また、培地としては、普通ブイヨン培地、ハートインフュージョンブイヨン培地、トリプトソイ(SCD)ブイヨン培地、SCDLPブイヨン培地、乳糖ブイヨンJP培地(以上栄研社製)、ブレインハートインフュージョン(BHI)培地、ニュートリエントブイヨン培地(以上DIFCO社製)が挙げられるが、なかでもBHI培地が好ましい。

本発明の洗浄剤組成物によれば、汚れを放置したままの衣料、台所及び浴槽等の悪臭の原因となるバイオフィルムを分解・除去することができる。また、本発明の除菌・洗浄力評価方法によれば、汚れが付着したまま放置した衣類の汚れや、汚れが付着したまま放置した台所や浴槽等の汚れに対する、除菌・洗浄力評価がより実際に近い形でできる。

以下、実施例及び比較例を示し、本発明を具体的に説明するが、本発明は下記の実施例に制限されるものではない。なお、下記の例において%は質量%を示す。

バイオフィルム分解活性の測定方法
1.布上及び硬質表面上のバイオフィルムの作製
1−1.前培養
20mLのブレインハートインフュージョン(BHI)培地(DIFCO社製)が入った100mL容の三角フラスコに、1白金耳分の表皮ブドウ球菌を添加し、37℃110rpmで18時間振盪培養器を用い(振盪恒温水槽、ファイン製)培養した。
1−2.バイオフィルム作製
表皮ブドウ球菌の前培養した液を、静岡県三方が原より採取した赤土を5質量%又は玉川圧縮C級カーボンを0.01質量%含むBHI培地に終濃度2質量%となるように添加した。予め高圧蒸気滅菌した平織り綿布(品番12000、柄色番DFシタザラシ、カネボウ繊維製、大きさ5cm×5cm)又は、スライドガラス(品番S1111、旭テクノガラス社製)をプラスチックシャーレ(商品名バイオビックI−90G、バイオビック製)に敷き、菌の入った培地をプラスチックシャーレ1枚当り正確に20mLとなるようにそれぞれ注いだ。
その後、プラスチックシャーレを37℃の恒温槽(ヤマト科学製)にて4日静置した。布及びスライドガラス上に細菌が増殖し、ゼラチン状のバイオフィルムが形成され、バイオフィルム汚垢布(FU評価用)、又は、バイオフィルム汚垢ガラス(HU評価用)を作製した。

2.バイオフィルム分解活性の測定
2−1.汚垢布上のバイオフィルム分解活性測定法
1−2で調製した汚垢布を2Lのビーカーにて3゜DH硬水(塩化カルシウムでCaOとして30mg/L、以下水とは3゜DH硬水を指す)1Lに入れ、スターラー(多連式マグネチックスターラー、アドバンテック製)の回転速度150rpmでスターラーバー(攪拌子 長さ50mm、直径8mm)を用いて1分間回転させて濯いだ。次いで、500mL容のマヨネーズ瓶(マヨネーズ450、日本山村硝子製)に予め試験洗浄液を666ppmとなるように水を用いて調製した液を250mL添加し、そこに汚垢布を1枚添加し蓋をした。

マヨネーズ瓶を回転装置(マツシタ工業社製 品名タンブラー)を用いてマヨネーズ瓶の重心付近を中心とし垂直円軌道に、回転速度60回/分で10分間回転させ、汚垢布を洗浄した。洗浄時の温度は、25℃で行なった。

1Lのビーカーを用いて、700mLの水でスターラーを用い250rpmの回転速度で3分間回転洗浄したのち、乾燥機(東京理化器械社製 型式NDO−SD型)で100℃5時間乾燥させ、その後シリカゲルの入ったデシケーターに入れ室温にて5時間保管したものの質量を測定し、洗浄後の汚垢ガラスの乾燥質量とした。

FU(布バイオフィルム分解活性)で調製した汚垢布を水で濯いだ後、500mL容のマヨネーズ瓶に、表1に記載の洗浄剤組成物を予め666ppmに水を用いて調製した洗浄液を250mL添加し、布を挿入した後に蓋をした。その後、回転装置(マツシタ工業社製)を用いて回転速度60±3回/分で10分間回転させた。
布を取りだし、ビニール袋に移し、SCDLP液体培地(DIFCO社製)50mLを加え、ストマッカー(ELMEX社製 形式SH−IIM)により1分間抽出操作をした。その後、抽出液を順次希釈しBHI培地に塗抹し37℃で48時間培養し、生菌数をカウントした。洗浄剤組成物と同様に標準洗剤(Tween80、666ppm)でも処理し、標準洗剤と洗浄剤組成物との生菌数の対数値の差を布に対する除菌力とし、下記評価基準で評価した。

HU(硬質表面バイオフィルム分解活性)で調製したバイオフィルム汚垢ガラスを水で濯ぎ、汚垢ガラスに表2に記載の洗浄剤組成物(液状)を0.1g滴下し、出来るだけ均一に塗布し、10分間静置した。プラスチックシャーレにBHI培地を10mL添加し、その容器に10分間静置した板を挿入した。その後、ガラスビーズ5g及びBHI培地を5mL添加し、手で毎分150回転を最大として1分間攪拌した。
得られた菌懸濁液を順次希釈し、BHI寒天培地に塗抹し37℃で48時間培養し、生菌数を測定した。洗浄剤組成物と同様に標準洗剤(Tween80原液)で洗ったものも生菌数を測定した。標準洗剤と洗浄剤組成物との生菌数の対数値の差を布に対する除菌力とし、下記評価基準で評価した。
