本発明は、細胞培養装置に係り、特に、継代培養を行う細胞培養装置に関する。

【従来の技術】
細胞の継代培養を行う場合、培養器内の培地の交換や、継代培養のための新しい培地への細胞の再播種などといった煩雑な作業が手作業により行われている。また、これらの継代培養に伴う作業は、コンタミネーションなどの発生を抑制するため、熟練した作業者が行う必要がある。

【発明が解決しようとする課題】
このように、継代培養に伴う作業は、煩雑であるにもかかわらず手作業で行われているのが現状であり、また、熟練した作業者を必要とするものであるため、継代培養は、作業性が悪く、容易に実施し難いものである。

特に、培養細胞による有用成分の生産や、再生医療用の組織を構築するための幹細胞の培養などといった継代培養を行う必要がある工業的な細胞培養では、継代培養に伴う作業の作業性の悪さから、量産化が難しく、継代培養の作業性の向上が課題となっている。

本発明の課題は、継代培養に伴う作業の作業性を向上することにある。

【課題を解決するための手段】
本発明の細胞培養装置は、内部環境が調整可能で細胞が播種された培地の入った少なくとも1つの培養器を収容して細胞の培養を行う培養手段と、培養器内の培地の交換を行う培地交換手段と、培養器内の細胞の状態を計測する細胞状態計測手段と、培養手段で培養された培養器内の細胞を細胞が播種されていない別の培地に播種する細胞播種手段と、培養器を搬送する搬送手段と、培養手段、培地交換手段、細胞状態計測手段、細胞播種手段、及び搬送手段を収容した気密に形成された筐体とを備え、この筐体は、少なくとも1つの開閉部を有し、搬送手段は、少なくとも開閉部と細胞播種手段との間で培養器を搬送する構成とすることにより上記課題を解決する。

このような構成とすることにより、細胞の培養、培養器内の培地の交換、培養器内の細胞の状態の計測、培養器内の細胞の状態がコンフルエントになった培養器内の細胞の新しい培地への再播種といった継代培養に伴う作業を1つの装置で行うことができる。さらに、これらの継代培養に伴う作業に際してコンタミネーションの発生を抑制する必要があるが、これらの継代培養に伴う作業を全て気密に形成された筐体内で外部環境と隔離した状態で行うことができ、コンタミネーションの発生を抑制できる。したがって、継代培養に伴う煩雑な作業を熟練した作業者による手作業で行う必要がなくなり、継代培養の作業性を向上できる。

また、培養手段は、培養器が1つずつ収容され、隔壁で隔てられた複数のスロットを有する構成とすれば、隣り合うスロットに収容された培養器の接触を防ぎ、培養器間の微生物の感染によるコンタミネーションやクロスコンタミネーションなどを防ぐことができるので好ましい。

さらに、筐体は、この筐体内の培養手段を構成する培養器を収容する空間と、この培養器を収容する空間の以外の空間とを隔てる内部隔壁を有し、この内部隔壁は、少なくとも1つの内側開閉部を有している構成とする。このように構成すれば、この筐体内の培養手段を構成する複数の培養器を収容する空間を、筐体内の他の空間、つまり培地の交換や播種などを行う空間と隔離することができるため、培養器を収容する空間の内部環境の調整が容易になるので好ましい。

また、細胞播種手段は、細胞状態計測手段での計測結果に応じて、この細胞の状態を計測した培養器内の細胞を細胞が播種されていない別の培地に播種する構成とすれば、装置が細胞状態計測手段での計測結果に応じて自動的に継代作業を行うため、継代培養に伴う作業の作業性をより向上できる。

また、細胞状態計測手段は、培養器を撮影するカメラを有し、このカメラで撮影した画像を解析して細胞の状態を計測する構成とすれば、作業者が顕微鏡などによる観察によって細胞の状態を計測する必要を無くすことができるため、継代培養に伴う作業の作業性をさらに向上できるので好ましい。

さらに、筐体に搬入する前または後のいずれかで培養器に付された識別情報を読取る識別情報読取り手段を備えた構成とする。このような構成とすれば、装置内に搬入された初代培養を行う培養器に収容された細胞の種類や由来に関する情報や、装置内での培養器内の培養の状態や培養器の位置に関する情報といった培養器に関する情報の管理が容易になるので好ましい。

さらに、筐体内に細胞を播種した培地を収容した培養器が在るとき、警報及び筐体の開閉部を閉じた状態に保持することの少なくとも一方により、別の培養器の筐体内への搬入を阻止する培養器搬入阻止手段を有する構成とする。このような構成とすれば、クロスコンタミネーションを阻止するため、1つの細胞培養装置を1つの細胞の培養に用いる場合、別の細胞が収容された培養器が誤って装置内に搬入されるのを防ぐことができるので好ましい。

また、筐体に搬入する前に培養器に付された識別情報を入力または読取ることにより認識した識別情報から、この識別情報を付した培養器内の細胞が筐体内に在る培養器内の細胞と由来が異なることを検出したとき、警報及び筐体の開閉部を閉じた状態に保持することの少なくとも一方により、異なる由来の細胞を収容した培養器の筐体内への搬入を阻止する培養器搬入阻止手段とを有する構成とする。このような構成とすれば、クロスコンタミネーションを阻止するため、1つの細胞培養装置を1つの由来の細胞の培養に用いる場合、異なる由来の細胞が収容された培養器が誤って装置内に搬入されるのを防ぐことができるので好ましい。

さらに、筐体内の気圧を筐体周囲の気圧よりも陽圧に調整する気圧調整手段を有する構成とすれば、筐体の開閉部を例えばエアロック式といった筐体外部の雰囲気が筐体内部に流入し難い構造にする必要がなく、筐体の開閉部の構造を簡素化できるので好ましい。

【発明の実施の形態】
以下、本発明を適用してなる細胞培養装置の一実施形態について図1乃至図5を参照して説明する。図1は、本発明を適用してなる細胞培養装置の概略構成と動作を示すブロック図である。図2は、本実施形態の細胞培養装置のより具体的な構成を示す断面図である。図3は、本実施形態の細胞培養装置の制御器と各作動部分との接続状態の概略構成を示すブロック図である。図4は、本発明を適用してなる細胞培養装置の外観を示す斜視図であり、(a)は、単体で細胞培養装置を設置した状態を、(b)は、複数台の細胞培養装置を連結して設置した状態を示す図である。図5は、本発明を適用してなる細胞培養装置の概略の動作を示すフロー図である。

本実施形態の細胞培養装置は、図1に示すように、培養器1に付されたこの培養器1に付された識別情報つまりIDを読取るID読取り手段3、細胞の状態を計測する細胞計測手段5、搬送手段6の一部をなし、培養器1の搬送経路を分岐させて培養器1の搬送先を選択する分流手段7、培養器1内の培地の交換を行う培地交換手段9、培養器1内の細胞を別の培養器1内の培地に播種して分配する細胞播種手段11、ID読取り手段3で読み取った識別情報に関連させた関連識別情報つまり関連IDを作成して細胞播種手段11で細胞が播種された培養器1に付与する関連ID付与手段13、そしてインキュベータである培養手段15などを備えている。

さらに、本実施形態の細胞培養装置は、これらのID読取り手段3、細胞計測手段5、搬送手段6、分流手段7、培地交換手段9、細胞播種手段11、関連ID付与手段13、そして培養手段15などを気密に形成された筐体17内に収容したものである。また、本実施形態の細胞培養装置には、細胞培養装置の動作を制御するための制御器21が、筐体17の外部に設けられている。

培養手段15は、筐体17内の他の空間と内部隔壁23によって隔てられている。内部隔壁23には、内側開閉部となる内側搬入口25aと内側搬出口25bとが設けられている。したがって、搬送手段6は、培地交換手段9から内側搬入口25aへ、または関連ID付与手段13から内側搬入口25aへ培養器1を搬送し、また、内側搬出口25bから細胞計測手段5へ培養器1を搬送する。さらに、培養手段15は、培養手段15内部の二酸化炭素濃度を検出する二酸化炭素濃度センサ27、そして、一端に二酸化炭素ボンベ29が連結され、他端が培養手段15の培養器1を収容する培養空間に連通する二酸化炭素供給管路31などを有している。二酸化炭素供給管路31には電磁バルブ33が設けられており、電磁バルブ33と二酸化炭素濃度センサ27とは、配線35を介して制御器21に電気的に接続されている。

培養手段15は、制御器21が二酸化炭素濃度センサ27で検出した培養手段15を構成する空間、つまり培養手段15の培養器1を収容する培養空間内の二酸化炭素濃度に応じて二酸化炭素供給管路31の電磁バルブ33により流量を制御することにより、培養手段15の培養器1を収容する培養空間内の二酸化炭素濃度を一定の範囲に保持することができる。さらに、培養手段15は、図示していないがヒータなどの加熱器や温度センサなどを備えており、培養手段15の培養器1を収容する培養空間内の温度を一定の範囲に保持することができる。このように、培養手段15は、炭酸ガスインキュベータとなっており、培養手段15の培養器1を収容する培養空間内の二酸化炭素濃度や温度、つまり内部環境を調整可能である。なお、培養手段15は、図1では示していないが、培養手段15の培養空間内に培養器1を収容するスロットや、このスロットと内側搬入口25aや内側搬出口25bとの間で培養器1を搬送する培養手段内搬送手段などを有している。

一方、筐体17内の培養手段15の培養空間以外の空間にも、培養手段15と同様に電磁バルブ33が設けられた二酸化炭素供給管路31が連結されており、また、筐体17内の培養手段15の培養空間以外の空間内における二酸化炭素濃度を検出する二酸化炭素濃度センサ27が設けられている。このため、筐体17内の培養手段15の培養空間以外の空間内における二酸化炭素濃度を、培養手段15の培養空間内と同じ状態に調整でき、培養手段15の培養空間に培養器1を搬入または搬出する際に、培養手段15を画成する内部隔壁23に設けられた内側搬入口25aや内側搬出口25bを開けても培養手段15の培養空間の内部環境つまり培養環境を変化し難くできる。

このような本実施形態の細胞培養装置のより具体的な構成について以下に説明する。本実施形態の細胞培養装置では、図2に示すように、筐体17の側壁の1面に図1で示した細胞搬入口19aと細胞搬出口19bとを兼ねる1つの開閉部19を設け、筐体17内の開閉部19を設けた側壁側の空間を培地交換及び細胞播種部37とし、この培地交換及び細胞播種部37となる空間にID読取り手段3、細胞計測手段5、搬送手段6、分流手段7、培地交換手段9、細胞播種手段11、関連ID付与手段13などを構成する各機器類を収容している。一方、筐体17内の開閉部19を設けた側と対向する側壁側の空間を培養部39とし、この培養部39となる空間が、培養器1を収容して、この培養器1内の培地に播種された細胞の培養を行うための培養空間となる。培地交換及び細胞播種部37と培養部39との間は、内部隔壁23で仕切られている。内部隔壁23には、図1で示した内側搬入口25aと内側搬出口25bとを兼ねる1つの内側開閉部25が設けられている。

筐体17に設けられた開閉部19は、閉じたときに筐体17内を密閉状態とし、筐体17の開閉部19の開口41を覆うシャッター43、シャッター43の上縁部分に固定されたワイヤ45、筐体17の壁面の開口41の上方部分に固定されたシャッターモータ47、そして、シャッターモータ47の回転軸に固定されてワイヤ45を巻き取るプーリー49などで構成されている。そして、開閉部19のシャッターモータ47は、図3に示すように、制御器21のI/Oポート21aに配線35を介して接続されている。内部隔壁23に設けられた内側開閉部25は、図2に示すように、内部隔壁23に形成された開口51を覆うシャッター53、シャッター53の上縁部分に固定されたワイヤ55、内部隔壁23の開口51の上方部分に固定されたシャッターモータ57、そして、シャッターモータ57の回転軸に固定されてワイヤ55を巻き取るプーリー59などで構成されている。

このような構成の細胞培養装置の動作と本発明の特徴部について説明する。ここでは、クロスコンタミネーションを防ぐため、1台の細胞培養装置で1つの培養器から細胞を継代して細胞を増やす場合の一例を説明する。操作者が、制御器21の入力用機器類などで開閉部19のシャッター43を開けることを指令するスイッチなどをオンすると、図5に示すように、制御器21は、細胞が筐体17内に既に搬入されているか、つまり既に細胞の培養が行われているかを判断する(ステップS1)。ステップS1において細胞が筐体17内に既に搬入されていることを判断すると、警報機能をオンし、警報ランプ113を点灯すると共に警報用スピーカ115から警報音を発し、さらに、開閉部19のシャッター43が開かないようにロックする(ステップS3)。一方、ステップS1において細胞が筐体17内に搬入されていないことを判断すると、紫外線ランプ82を消灯し(ステップS5)、その後、開閉部19のシャッター43が開く(ステップS7)。
