- 【特許請求の範囲】 1.次の各要素からなる隔離されたポリヌクレオチド断片: (a)少なくとも1つのブタのレトロウイルス(PoEV)発現体を符号化する、 (b)前記発現体の生理化学的に活性なおよび/または免疫原的な誘導体を符 号化する、または、 (c)上記(a)、(b)で定義されたポリヌクレオチドシーケンスに相補的な ポリヌクレオチド断片。 2.次の各要素からなる請求項1に記載の隔離されたポリヌクレオチド断片: (a)ポリメラーゼ(POL)ポリペプチドを符号化する、 (b)上記(a)のポリペプチドを生理化学的に活性なおよび/または免疫原的 な誘導体を符号化する、または、 (c)上記(a)、(b)で定義されたポリヌクレオチドシーケンスに相補的な ポリヌクレオチド断片。 3.次の各要素からなる請求項1に記載の隔離されたポリヌクレオチド断片: (a)ビリオンコアポリペプチド(GAG)および/またはエンベロープポリペ プチド(ENV)を符号化する、 (b)上記(a)のポリペプチドを生理化学的に活性なおよび/または免疫原 的な誘導体を符号化する、または、 (c)上記(a)、(b)で定義されたポリヌクレオチドシーケンスに相補的な ポリヌクレオチド断片。 4.次の各要素からなる請求項1に記載の隔離されたポリヌクレオチド断片: (a)ビリオンコアポリペプチド(GAG)、ポリメラーゼ(POL)およびエンベロ ープポリペプチド(ENV)を符号化する、 (b)上記(a)のポリペプチドを生理化学的に活性なおよび/または免疫原 的な誘導体を符号化する、または、 (c)上記(a)、(b)で定義されたポリヌクレオチドシーケンスに相補的な ポリヌクレオチド断片。 5.ポリヌクレオチド断片が、デオキシリボース核酸(DNA)断片である請求 項1〜4のいずれかに記載の隔離されたポリヌクレオチド断片。 6.次の各要素からなる前記請求項のいずれかに記載の隔離されたポリヌクレオ チド断片: (a)図3または4に示されるアミノ酸シーケンスを有する少なくとも1つの ポリペプチドを符号化する、 (b)上記(a)に定義の少なくとも1つのポリペプチドの生理化学的に活性 なおよび/または免疫原的な誘導体であるポリペプチドを符号化する、または、 (c)上記(a)、(b)で定義されたポリヌクレオチドシーケンスに相補的な ポリヌクレオチド断片。 7.次の各要素からなる前記請求項のいずれかに記載の隔離されたポリヌクレオ チド断片: (a)図1、2、3または4に示されるORFの少なくとも1つ、またはこれに 対応のRNAシーケンスを有する、 (b)上記(a)に記載のシーケンスと同一の実質的なヌクレオチドシーケン スを有するシーケンスを有する、または、 (c)上記(a)、(b)に記載されたシーケンスに相補的なシーケンスを有す る。 8.請求項1〜7のいずれかに記載のポリヌクレオチド断片を有する組換え核酸 分子。 9.組換え核酸分子が、ポリヌクレオチド断片の発現を制御するためにポリヌク レオチド断片に機能的に結合する制御シーケンスを有する請求項8に記載の組換 え核酸分子。 10.請求項8または9に記載の組換え核酸分子を有するベクター。 11.ウイルスまたはプラスミドである請求項10に記載のベクター。 12.請求項1〜11のいずれかに記載のポリヌクレオチド断片、組換え核酸分 子、またはベクターにより形質変成された原核生物型または真核生物型宿主細胞 。 13.POL PoEV生理化学的および/または免疫原的活性を示す組換えPoEVポリペ プチドまたはその誘導体。 14.GAGおよび/またはENV PoEV生理化学的および/または免疫原的活性 を示す組換えPoEVポリペプチドまたはその誘導体。 15.GAG、POLおよびENV PoEV生理化学的および/または免疫原的活性を示す組 換えPoEVポリペプチドまたはその誘導体。 16.図3または4に示されるシーケンスを有する請求項13〜15に記載のい ずれかの組換えPoEVポリペプチドまたはその機能的に活性な誘導体。 17.請求項13〜16のいずれかに記載の組換えPoEVポリペプチド、または不 活性なPoEVウイルスおよび医学的に許容されるキャリアを有するワクチン。 18.請求項13〜16のいずれかに記載のポリペプチドまたは断片に結合可能 な抗体または断片。 19.PoEV特性であるポリヌクレオチドプライマー。 20.PoEVポリヌクレオチドシーケンスに対し具体的にハイブリッド形成できる ポリヌクレオチドプローブ。 21.図1、2、3または4に示される分子のストランドと、またはその相補的 なストランド、対応のRNA分子または該分子の一部を有する、と同一の実質的な ヌクレオチドシーケンスを有する請求項19または20に記載のプローブまたは プライマー。 22.請求項19〜21のいずれかに記載のポリヌクレオチドプライマーまたは プローブを有するPoEV検出キット。 23.サンプル中のPoEVの検出におけるPoEV特性ポリヌクレオチドの使用。 24.サンプル中のPoEVの検出のPCRにおけるPoEV特性ポリヌクレオチドの使用 。 25.ヒトに感染する感染性PoEVを出さないように変成したブタ。 26.請求項25に記載のブタから得られる細胞、組織または器官。 27.ワクチンの調製における請求項13〜16に記載の組換えPoEVポリペプチ ドの使用。 28.サンプルにおけるPoEV核酸の存在を検出することのできる検出キットの調 製における請求項19〜21に記載のポリヌクレオチドプライマーまたはプロー ブの使用。 29.治療または診断における請求項1〜7、13〜16、26のいずれかに記 載のポリヌクレオチド、ポロペプチド、細胞、組織、または器官の使用。 30.治療または診断に使用する薬剤の調製における請求項1〜7、13〜16 、26のいずれかに記載のポリヌクレオチド、ポロペプチド、細胞、組織、また は器官。 31.ここまでに実質的に記載された発明。
