Claims (1)
- 【特許請求の範囲】 1.コハク酸産生バクテリアを単離する方法であって、 a) ピルベートを異化作用する能力に欠けた、通性生物を単離し、 b) 好気的方法で、該生物のバイオマスを増大させ、 c) 嫌気性環境内で、該バイオマスをグルコースに富む培地に接種して、ピル ベート−異化作用可能な突然変異体の成長を可能とし、 d) 該突然変異体を単離する、 諸工程を含む、上記方法。 2.該通性生物が、ピルベートホルメートリアーゼおよびラクテートデヒドロゲ ナーゼ活性を欠く、請求の範囲第1項に記載の方法。 3.ピルベートを異化作用する能力に欠けた該通性生物が、NZN 111 である、請 求の範囲第1項に記載の方法。 4.該生物を、ルリアブロス上で好気的に培養する、請求の範囲第1項に記載の 方法。 5.該バイオマスを、1ml当たり約109〜1010細胞の範囲まで増大する、請求の範 囲第1項に記載の方法。 6.該グルコースに富む培地が、約1g/l〜30 g/lの範囲のグルコースを含む、請 求の範囲第1項に記載の方法。 7.醗酵生成物としてコハク酸、酢酸およびエタノールの混合物を生成し、ピル ベートホルメートリアーゼおよびラクテートデヒドロゲナーゼに対する遺伝子を 欠く親から誘導され、バクテリアのE.コリ群に属することを特徴とする、突然変 異体。 8.該親がNZN 111 である、請求の範囲第7項に記載の突然変異体。 9.請求の範囲第7項に記載のAFP 111 。 10.コハク酸が主な醗酵生成物である、請求の範囲第7項記載の突然変異体。 11.コハク酸を生産する生物の創製方法であって、 a) カルボン酸産生生物を、グルコースを欠く培地中に入れて、そのバイオマ スを増大させる工程と、 b) 該増大させたバイオマスを、嫌気性雰囲気下で、グルコースに富む醗酵培 地に接種する工程と、 c) コハク酸産生生物を該バイオマスから単離する工程と、 を含む上記方法。 12.該生物が浸透圧耐性である、請求の範囲第11項記載の方法。 13.該生物が、ピルベートをジカルボン酸に転化することのできる酵素を発現す るように、遺伝子操作されている、請求の範囲第11項記載の方法。 14.該酵素がリンゴ酸酵素である、請求の範囲第13項記載の方法。 15.該生物がラクトバチルスおよびE.コリバクテリアからなる群から選ばれる、 請求の範囲第14項記載の方法。 16.該生物がE.コリバクテリアであり、該バクテリアが、ピルベートホルメート リアーゼおよびラクテートデヒドロゲナーゼに対する遺伝子を欠く親から誘導さ れ、かつ該生成されるカルボン酸が主としてコハク酸である、請求の範囲第15項 記載の方法。 17.該コハク酸の生成を、水素に富む雰囲気内で増強する、請求の範囲第16項記 載の方法。 18.該生物がラクトバチルスバクテリアであり、かつ該生成されるカルボン酸が リンゴ酸である、請求の範囲第15項記載の方法。 19.該生物が、醗酵生成物としてコハク酸、酢酸およびエタノールの混合物を生 成し、ピルベートホルメートリアーゼおよびラクテートデヒドロゲナーゼに対す る遺伝子を欠く親から誘導され、かつバクテリアのE.コリ群に属する、請求の範 囲第11項記載の方法。 20.該グルコースに富む培地中の、該グルコースの濃度が約10〜30 g/lの範囲に ある、請求の範囲第11項記載の方法。
