Abstract

2,4,7- Triamino- 6-phenylpteridines of formula <FORM:0932256/IV(a)/1> wherein R is hydrogen, 1 to 3C alkyl, 1 to 3C alkoxy or halogen, are produced by reacting 5-nitroso-2,4,6-triaminopyrimidine with the appropriate phenylacetonitrile in an organic solvent in the presence of sodium or an anhydrous sodium alkoxide such as sodium methoxide, 2,4-diaminopteridines of formula <FORM:0932256/IV(a)/2> in which R1 and R2 are hydrogen, methyl or ethyl, are produced by reacting 2,4,5,6-tetraaminopyrimidine, or a salt thereof, with a 1,2-dicarbonyl compound such as glyoxal, biacetyl or a dione such as pentanedione or hexanedione. Examples illustrate the preparation of 2,4,7-triamino-6-phenylpteridine; 2,4,7-triamino-6-o-methylpteridine; 2,4,7-triamino-6-p - chlorophenylpteridine; 2,4-diamino-6-7-di-methylpteridine; 2,4-diaminopteridine; 2,4-diamino-6-methylpteridine and 2,4-diamino-7-ethyl-6-methylpteridine.ALSO:Diuretic and anti-hypertensive medicines for oral or parenteral use in the treatment of humans and animals comprise a carrier and either a compound having the formula <FORM:0932256/VI/1> in which R is hydrogen, 1 to 3C alkyl, 1 to 3C alkoxy or halogen; or a compound of formula <FORM:0932256/VI/2> in which R1 and R2 are hydrogen, methyl or ethyl. The preparations are used orally in the form of tablets, capsules, lozenges, powders, suspensions in a vegetable oil or water and in the case of tablets time delay materials such as glyceryl mono- or di-stearate alone or with a wax may be present. The compositions may be added to animal feeds. Optional additions are (a) vitamins and (b) diuretics such as chlorothiazide, hydrochlorothiazide and acetazolamide.
