Abstract

Tetracycline and its salts are prepared by culturing a new strain of Streptomyces aureofaciens designated S. aureofaciens NCIB-9114 myces. The new strain is prepared from a chlortetracycline producing strain of S. aureofaciens designated LSZ-536 by the following in conventional nutrient media for Streptosteps (a) UV-irradiation to produce mutant C-241, (b) X-ray irradiation of C-241 to produce mutant D-6, (c) UV-irradiation of D-6 in the presence of benzyl thiocyanate to produce mutant G-993, (d) X-ray irradiation of G-993 and chilling to produce mutant CD-815, (e) treatment of CD-815 with streptomycin to produce mutant CDS-425, (f) incubation of CDS-425 in Coenzyme A supplemented synthetic medium, followed by X-ray irradiation in the presence of KCN to produce mutant CDSD-314 (NCIB-9114). The tetracycline may be recovered in the conventional manner but with a step of treating an aqueous acid extract containing ferric salts e.g. chloride with a borohydride e.g. sodium borohydride and concentrating to precipitate the tetracycline. Reference has been directed by the Comptroller to Specification 747,779.ALSO:A tetracycline-producing strain of Streptomyces aureofaciens (NCIB-9114) is produced from a chlortetracycline-producing strain of S. aureofaciens designated LSZ-536 by the following steps: (a) UV-irradiation to produce mutant mutant C-241; (b) X-ray irradiation of C-241 to produce mutant D-6; (c) UV-irradiation of D-6 in the presence of benzyl thiocyanate to produce mutant G-993; (d) X-ray irradiation of G-993 and chilling to produce mutant CD-815; (e) treatment of CD-816 with streptomycin to produce mutant CDS-425; (f) incubation of CDS-425 in coenzyme A supplemented synthetic medium, followed by X-ray irradiation in the presence of KCN to produce mutant CDSD-314 (NCIB-9114). Reference has been directed by the Comptroller to Specification 747,779.
