Abstract
A method for detecting or enriching a target nucleic acid in a nucleic acid sample comprising fragmenting the sample into fragments containing said target nucleic acid, rendering said fragments single stranded, contacting the fragments with a partially double-stranded probe comprising a selector oligonucleotide with two non-adjacent target specific regions and a non-target specific region which comprises at least two binding sites for a vector oligonucleotide, and at least one vector oligonucleotide not complementary to the target sequence and which comprises a nucleotide sequence complementary to the vector-binding site of said selector oligonucleotide, said vector oligonucleotide also comprising a region which is not complementary to the selector oligonucleotide which comprises a nucleotide sequence containing element(s) for detection/enrichment, allowing hybridisation of complementary portions of the probe oligonucleotides and the target to occur, ligating said vector oligonucleotide(s) and target to produce a probe-target fragment hybrid, and detecting or enriching for said hybrid.
