Abstract
1351836 Determination of thyroxine YISSUM RESEARCH DEVELOPMENT CO OF THE HEBREW UNIVERSITY OF JERUSALEM 28 June 1971 [29 June 1970] Heading G1B Apparatus for determination of thyroxine in serum comprises a tubular body having closure means at both ends and containing a column of dextran gel cross-linked with epichlorhydrin and having a water regain of 1 to 5 gm per gm dry gel, the dextran gel being in equilibrium with an aqueous medium having a pH of at least 11. The apparatus may comprise a syringe barrel closed by polyethylene caps, the gel being retained between porous sintered polyethylene discs. In carrying out the determination, predetermined amounts of the serum and of radioactive thyroxine solution are allowed to flow into the column, the column is washed with an aqueous alkaline solution pH 8 to 10 to remove protein, a predetermined quantity of a binding agent for the thyroxine, for example human or bovine serum, human alpha globulin or bovine gamma globulin is added.to the column to partially remove the thyroxine from the column, the ratio of radioactive thyroxine retained by the column to that originally added is determined, and the thyroxine content of the serum is calculated by comparing the ratio determined and ratios obtained with known concentrations of thyroxine in serum.
