1323521 Gel plate for enzyme determination BAXTER LABORATORIES INC 22 Jan 1971 3024/71 Heading B1X An agar gel plate for use in a micro method for the quantitative determination of low levels of DPN-or TPN-linked enzyme activity comprises a shallow plate carrying an agar gel medium buffered to a pH of 6 to 9 and containing 0.2 to 4% agar, DPN or TPN, a substrate for the enzyme and a chromogenic or fluorogenic substance. The plate may be packed in metal foil or plastics film. For the determination, the sample under test is diffused from a 1 to 10 mm within the gel medium and the diameter of the coloured or fluorescent radial diffusion zone produced is measured and compared with that obtained with control samples. The DPN or TPN &c may be incorporated into the gel medium when the plate is prepared or alternatively a solution of these components is laid on the plate after the plate has been incubated with the test sample in the wells, and the plate incubated again. Examples relate to determination of LDH using DPN, lithium lactate, p-nitrotetrazolium blue, phenazine methosulphate and cyanide ion; other enzyme-substrate systems are mentioned.
