Abstract

1293541 Folic acid derivatives J A BLAIR and K J SANDERS 16 June 1970 [18 March 1969] 14027/69 Heading C2C The invention comprises the novel folic acid derivatives of the formula wherein A is OH, NH 2 , NHR, NR 2 or OR; R is CH 3 , CH 2 CH 3 , CH 2 CH 2 CH 3 , CH(CH 3 ) 2 or CH 2 CH 2 CH 2 CH 3 ; R<SP>1</SP> is H or CH 3 ; and X is the same or different, and is H, alkali metal or alkaline earth metal; provided that when R<SP>1</SP> is H, A is not OH or NH 2 , their preparation, the preparation of folic acid derivatives of the above formula in which A and X are as defined above, R is isobutyl, tertiary butyl, C 2-4 alkenyl or C 2-4 alkynyl; and R<SP>1</SP> is C 2-4 alkyl, C 2-4 alkenyl or C 2-4 alkynyl and the preparation of compounds of the above formula in which A is OH or NH 2 and R<SP>1</SP> is H, by reducing in aqueous solution compounds of the formula with a first portion of sodium borohydride in excess to give as intermediates 5, 6, 7, 8-tetrahydro intermediates, destroying the excess sodium borohydride, and immediately thereafter and without isolating said intermediates reductively condensing the intermediates with the appropriate aldehydes or ketones and sodium borohydride.
