Abstract

1,254,268. Erythromycin B derivatives. ABBOTT LABORATORIES. 16 March, 1970 [14 March, 1969], No. 12628/70. Addition to 1,198,226. Heading C2A. Novel erythromycin B derivatives of formula wherein R 1 is amino, azido or dimethylamino and R 2 is hydroxy, or R 1 and R 2 taken together represent an epoxy group, or acid addition salts thereof, are prepared by reacting 3<SP>1</SP>-(dedimethylamino)-#<SP>31 41</SP> -erythromycin B with an epoxidizing agent, e.g. m-chloroperbenzoic acid, to give 3<SP>1</SP>-(dedimethylamino)-3<SP>1</SP>,4<SP>1</SP>-epoxyerythromycin B, and if required opening the epoxide ring with an azide to give 3<SP>1</SP>-(dedimethylamino)- 3<SP>1</SP>-azido-4<SP>1</SP>-hydroxyerythromycin B, which may be reduced, e.g. with catalytic hydrogen, to the corresponding 3<SP>1</SP>-amino derivative which is treated with formaldehyde under reducing conditions to give 4<SP>1</SP>-hydroxyerythromycin B.
