Abstract
1,208,129. Phenyl - imidazole compounds J. R. GEIGY A.G. 8 Dec., 1967 [9 Dec., 1966] No. 55922/67. Heading C2C. Novel compounds of the Formula I where R 1 is phenyl substituted in 2, 3 or 4 position by halogen (preferably fluorine, chlorine or bromine) or methoxy or ethoxy, one of R 2 and R 3 represents CH 3 or C 2 H 5 and the other H, CH 3 or C 2 H 5 , or pharmaceutically acceptable acid addition salts thereof are obtained by desulphurizing or reducing a compound of Formula II where X is -OH or -SH, or a tautomer thereof, and, optionally, converting it into an acid addition salt with an inorganic or organic acid. Where X is -SH in Formula II, the elimination of X is effected by desulphurization of the 1 - aryl - 2 - mercapto - imidazole or corresponding 2 - thiono derivative employing Raney Ni, HNO 3 or H 2 O 2 . Where X is -OH, the elimination of X is effected by reducing the 1 - aryl - 2 - hydroxyimidazoles or corresponding imidazol - 2 - ones with LiAlH 4 . 2,2 - Ethylenedioxypropyl isothiocyanate is obtained by reacting 1 - amino - 2,2 - ethylenedioxypropane with thiophosgene. 5 - Methyl - 1 - (2 - methoxyphenyl) - 2 - mercapto - imidazole is obtained by reacting 2,2 - ethylenedioxypropyl isothiocyanate with o-anisidine. 3 - Methoxyphenylisothiocyanate is obtained by reacting m-anisidine with thiophosgene. 5 - Methyl - 1 - (3 - methoxyphenyl) - 2 - mercaptoimidazole is obtained by reacting 3 - methoxyphenylisocyanate with 1 - amino - 2,2 - ethylenedioxypropane. Pharmaceutical compositions, suitable for treatment of hyperaldosteronism and associated disorders, useful as diuretics and cardiovascular agents, comprise an amount of 0À5- 90% of a compound of Formula I or a pharmaceutically acceptable acid addition salt thereof, and a pharmaceutical carrier in forms suitable for oral or parenteral administration.
