Abstract

The invention relates to the broad spectrum antibiotics R-451 B(X) and R-451 D(X) which are obtained by recrystallizing from an organic solvent the antibiotics R-451 B and R-451 D produced by cultivating micromonospora carbonacea according to Specification 1,021,402. Antibiotic R-451 B(X) has a m.pt. 154-157 DEG C., an elementary analysis C=51.02%, H=6.68%, N=1.23%, O=34.72%, Cl=3.97%, optical rotation [a ]25D= -25.5 (1% pyridine), U.V. max. absorption at 288 mm with E1% in <PICT:1074235/C2/1> <PICT:1074235/C2/2> methanol = 12 and an infra-red absorption spectrum as in Fig. 1. Antibiotic R-451 D(X) has a m.pt. 160-161 DEG C., an elementary analysis C=51.79%, H=6.35%, N=1.40%, O=36.35%, Cl=3.98%; an optical rotation [a ]25D= -25.3 DEG (1% in methanol); ultra-violet absorption maximum at 289 mm with E1% in methanol=22, and infra-red spectrum as shown in Fig. 2. R-451 D(X) methyl ether is prepared by treating R-451 D(X) with diazomethane in a solvent. R-451 D(X) acetate is prepared by treating R-451 D(X) with acetic anhydride in pyridine. The antibiotics form salts with bases, ethers (as above) and O-acyl dervatives (as above). Pharmaceutical compositions comprise one or more antibiotics identified as R-451 B(X), R-451 D(X) and R-451 D(X) acetate and a carrier. They are active against gram-positive pathogenic microorganisms in man and animals, e.g. mastitis in cattle and pleuropneumonia diseases in poultry.
