Abstract

A concentrated foot-and-mouth disease vaccine is prepared by adding to an aqueous suspension or living attenuated foot-and-mouth disease virus particles (1) a water-soluble polysaccharide or an ionic derivative thereof, (2) a water-soluble cellulose derivative or a water-soluble polyalkylene or polyoxyalkylene alcohol or glycol, the alkylene having 2 to 4 carbon atoms, and adjusting the ionic concentration e.g. by the addition of sodium chloride, to form an aqueous two-phase system comprising a phase in which the virus preferentially collects and which is smaller in volume than the original virus suspension, and separating the phase preferentially containing the virus. Polymers (1) and (2) may be (a) dextran and methylcellulose, (b) sodium dextran sulphate and methylcellulose, (c) dextran and polyethylene glycol, (d) sodium dextran sulphate and polyethylene glycol, or (e) sodium dextran sulphate and polyvinyl alcohol. The ionic concentration is adjusted to between 0.2 m and 0.5 m chloride ion. A polyvalent vaccine is obtained by mixing a vaccine obtained as above with other attenuated foot-and-mouth disease viruses.
