Abstract
Red polimérica que comprende un polímero de quitosano derivatizado con grupo dicarboxilo reticulado a un polímero de dextrano derivatizado con grupo aldehído.
Description
La Figura 14 es un gráfico que compara puntuaciones de costra del campo quirúrgico activo frente al control a lo largo del tiempo (Ejemplo 8).
DESCRIPCIÓN DETALLADA DE LA INVENCIÓN
5 En la presente memoria, el término «quitosano» significa un polisacárido lineal compuesto de D-glucosamina ligada por β-(1,4) y N-acetil-D-glucosamina distribuidas aleatoriamente. El quitosano se puede producir desacetilando la quitina. Tanto α-como β-quitosano son adecuados para su uso en la invención. El grado de desacetilación (% DA) influye en la solubilidad y en otras propiedades del quitosano. El quitosano disponible en el mercado tiene normalmente un grado de desacetilación de entre aproximadamente un 50 y un 100 %. En la Fórmula I, más abajo,
10 se muestra una unidad monomérica de un quitosano completamente desacetilado.
En la presente memoria, el término «polímero de quitosano derivatizado con grupo dicarboxilo» significa un polímerodequitosano quehasido derivatizado mediantelareaccióndeun anhídrido cíclico con elgrupo amina de alguno de los residuos de D-glucosamina del polímero de quitosano. Ejemplos de grupos dicarboxilo incluyen el N
15 succinilo, el N-maloilo y el N-ftaloilo. Se prefiere el N-succinilo.
El «polímero de quitosano derivatizado con grupo dicarboxilo» también se puede modificar parcialmente para derivatizarlo con otros grupos funcionales. Esta derivatización secundaria puede tener lugar, bien en posiciones amina que no estén derivatizadas con un grupo dicarboxilo, o bien en los grupos hidroxilo de los residuos de Dglucosamina. Por ejemplo, la reacción del anhídrido cíclico con un grupo OH del quitosano puede provocar que
20 algunosmonómeros contengan grupos éster, en lugar o además delsustituyente amida.
Si la derivatización secundaria está presente en la posición amina del polímero de quitosano derivatizado con grupo dicarboxilo, el polímero debe retenersuficientes grupos amina libres para poder formar reticulaciones con el polímero de dextrano derivatizado con grupo aldehído. Preferiblemente, el polímero de quitosano derivatizado con grupo dicarboxilo solo se derivatiza mediante la reacción del anhídrido cíclico con el grupo amina de algunos de los
25 residuos de D-glucosamina.
En la presente memoria, el término «polímero de N-succinil-quitosano» significa un quitosano que ha sido derivatizado añadiendo un grupo N-succinilo al grupo amina de alguno de los residuos de D-glucosamina del polímero de quitosano. En la Fórmula II, más abajo, se muestra una unidad monomérica de un polímero de Nsuccinil-quitosano.
El grado de succinilación puede variar. Por lo general, este se encuentra entre aproximadamente el 30 y el 70 %, pero el polímero de N-succinil-quitosano debe retener suficientes grupos amina libres para poder formar
