Abstract

Uso de una composición que comprende una población de células, población que comprende células que contienen un sustrato de toxina clostridial exógeno que puede ser localizado en la membrana plasmática de dichas células; en el que dichas células son capaces de permitir una intoxicación por una toxina clostridial posibilitando el mecanismo celular global mediante el cual una toxina clostridial escinde proteolíticamente un sustrato, y engloba la unión de una toxina clostridial con un receptor de baja o alta afinidad, la interiorización del complejo de toxina/receptor, la translocación de la cadena ligera de la toxina clostridial en el citoplasma y la modificación de la diana enzimática de un sustrato toxina clostridial; en el que las células capaces de permitir una intoxicación por una toxina clostridial son células que expresan uno o más receptores de toxina clostridial de baja o de alta afinidad endógenos para uno o más serotipos de toxina clostridial; expresan uno o más receptores de toxina clostridial de baja o de alta afinidad exógenos para uno o más serotipos de toxina clostridial; o expresan una combinación de receptores de toxina clostridial de baja o alta afinidad endógenos y receptores de toxina clostridial de baja o alta afinidad exógenos para uno o más serotipos de toxina clostridial; en el que dicho sustrato de toxina clostridial exógeno consiste en un miembro fluorescente, en el que dicho miembro fluorescente es un péptido que bien puede absorber inherentemente energía luminosa de una cierta longitud de onda y emitir energía luminosa de una longitud de onda diferente o que forma parte de un complejo que absorbe energía luminosa de una cierta longitud de onda y emite energía luminosa de una longitud de onda diferente; un dominio de dirección a la membrana que dirige el sustrato a la membrana celular; y una secuencia de reconocimiento de toxina clostridial que comprende un sitio de escisión que es un enlace escindible junto con elementos de reconocimiento para la proteolisis detectable en el enlace escindible por parte de una toxina clostridial, en la que el sitio de escisión se encuentra entre dicho miembro fluorescente y dicho dominio de dirección a la membrana; en el que más del 50% de dicha población de células comprende dichas células que contienen dicho sustrato de toxina clostridial exógeno para la determinación de la actividad de una toxina clostridial.
