Abstract

Die Erfindung betrifft eine Nanostruktur aus einer Vielzahl von Nanokristallen auf mindestens einer Oberfläche oder einem Oberflächenbereich eines Titankörpers und ein Verfahren zur Erzeugung dieser Nanostruktur durch hydrothermische Oxidation. Dabei haben die Nanokristalle zumindest bereichsweise eine tetragonalpyramidale Grundform. Die Flächendichte der Nanokristalle liegt pro μm2 zwischen 40 und 400, wobei die Flächendichte mit zunehmender Kristallhöhe abnimmt. Der mittlere Abstand von 50 bis 160 nm benachbarter Nanokristalle ergibt sich bei einer Nano- kristallhöhe von 23 bis 100 nm. Mit der vorliegenden Erfindung wird eine titanbasierte bakteri- zid und hydrophil wirkende Nanostruktur für Implantatoberflächen geschaffen und zugleich ein entsprechendes Herstellungsverfahren zur Verfügung gestellt, mit dem die Größe und die Verteilung der eine einheilungserleichternde Nanostruktur bildenden Nanokristalle bestimmbar sind.

Description

Nanostruktur und Verfahren zu ihrer Erzeugung
Die Erfindung betrifft eine Nanostruktur aus einer Vielzahl von Nanokristallen auf mindestens einer Oberfläche oder einem Ober flächenbereich eines Titankörpers und ein Verfahren zur Erzeu gung dieser Nanostruktur durch hydrothermische Oxidation.
Eine wichtige Voraussetzung für eine erfolgreiche und langfris tige Implantatintegration im menschlichen Körper ist eine stabile Verankerung des Implantats im jeweiligen Knochen. Das Implantat muss osseointegriert sein, also im direkten funktio nellen Knochenkontakt stehen. Als Implantatwerkstoff werden u.a. Titan und Titanlegierungen verwendet. Diese haben sich aufgrund ihrer Biokompatibilität, ihrer chemischen Inertheit und ihrer hervorragenden Korrosionsbeständigkeit als geeignet erwiesen. Während der Implantation und der unmittelbar danach beginnenden Einheitsphase ist der Schutz vor bakterieller Besiedelung von
BESTATIGUNGSKOPIE Keimen besonders wichtig. Um ein Entstehen unerwünschter Bio filme zu verhindern, gibt es u.a. die Möglichkeit einer medika mentösen Beschichtung oder des Verwendens von antibakteriellen Oberflächen.
In dem Sientific Reports-Artikel „Antibacterial titanium nano- patterned arrays inspired by dragonfly wings" vom 18.11.2015 der Autoren Chris M. Bhadra, Vi Khanh Truong, Vy T. H. Pha , Mohammad Al Kobaisi, Gediminas Seniutinas, James Y. Wang, Saulius Juodka- zis, Russell J. Crawford und Elena P. Ivanova wird die Herstellung einer antibakteriell wirkenden Nanostruktur beschrieben. Dabei wird eine mechanisch aufgerauhte Titanprobenoberfläche eine Stunde lang bei einem Druck von 1,7 bis 2,03*105 PA und einer Temperatur von 121 °C in einer Kaliumhydroxidlösung hydrothermisch oxidiert. Der Oxidation folgt eine dreistündige Heißluftwärmebehandlung bei 400 °C.
Mit diesem Verfahren wird eine fragile und spröde Nanostruktur hergestellt. Bei einer Belastung der Oberfläche, z.B. beim Im plantieren eines prothetischen Zahnersatzes besteht die Möglich keit, dass unzählige Titanoxidnanopartikel aus der Nanostruktur herausbrechen. Deren toxische Wirkung verursacht oft eine Ent zündung des Implantatbetts und einen damit einhergehenden Kno chenverlust im entsprechenden Kieferbereich.
Der vorliegenden Erfindung liegt die Problemstellung zugrunde, eine titanbasierte, bakterizid und hydrophil wirkende Nanostruk tur für Implantatoberflächen zu entwickeln und zugleich ein ent sprechendes Herstellungsverfahren zur Verfügung zu stellen, mit dem die Größe und die Verteilung der eine einheilungserleich- ternde Nanostruktur bildenden Nanokristalle bestimmbar sind. Diese Problemstellung wird mit den Merkmalen des Hauptan
spruchs 1 gelöst. Dabei haben die Nanokristalle zumindest be reichsweise eine tetragonal-pyramidale Grundform. Die Flächen dichte der Nanokristalle liegt pro pm zwischen 40 und 400, wobei die Flächendichte mit zunehmender Kristallhöhe abnimmt. Der mittlere Abstand von 50 bis 160 nm benachbarter Nanokristalle ergibt sich bei einer Nanokristallhöhe von 23 bis 100 nm.
Der selbstständige Patentanspruch 8 beschreibt ein Verfahren zur Erzeugung einer Nanostruktur auf mindestens einer Oberfläche o- der einem Oberflächenbereich eines Titankörpers durch hydrother- mische Oxidation. Dazu wird die Ausgangsfläche zur Nanostruktur- erzeugung einer 160 bis 374,12 °C heißen Dampfdruckatmosphäre ausgesetzt, wobei das dampfbildende Medium demineralisiertes Wasser ist. Hierbei sind die Nanokristallflächendichte und die Nanokristallgröße sowie die Farbe der Oxidschicht der Nanostruk tur jeweils eine Funktion der Dampfdruckeinwirkzeit, wobei - bei dem angegebenen Dampfdrucktemperaturbereich - für eine nutzbare Flächendichte und Größe der Nanokristalle die Einwirkzeit 1 bis 100 h beträgt.
Die mit diesem Verfahren erzeugte Oberflächentopographie weist kompakte und stabile Nanokristalle auf, die bei sachgemäßem Um gang während der Implantation nicht zum Abbrechen oder Absplit tern neigen. Die Topographie begünstigt zum einen aufgrund ihrer Rauheit das Anlagern von eukaryotischen Zellen wie insbesondere z.B. Fibroblasten und/oder Endothelzellen. Auf diese Weise kann beispielsweise nach einer Zahnprothesenimplantation ein Anwach sen des Zahnfleisches beschleunigt werden. Zum anderen behindert die erzeugte Oberflächentopographie ein vermehrtes Ansiedeln von gramnegativen Bakterien. Zu diesen gehört u.a. das Stäbchenbak terium Pseudomonas aeruginosa , ein typischer Krankenhauskeim, der inzwischen gegen mehrere Antibiotika resistent ist.
Diese Bakterien interagieren mit der Nanostruktur . Laut der mo mentan gültigen Modellvorstellung kommt es aufgrund von Oberflä cheneffekten zur Adsorption der Zellmembran zwischen die Nano- strukturen. Die Adsorption basiert u.a. auf der hydrophilen Ei- genschaft der vorgestellten Nanostruktur, wodurch diese eine überdurchschnittlich große Oberflächenbenetzbarkeit erlangt.
Bei der Adsorption verletzen die scharfkantigen Nanokristalle der Nanostruktur die Zellmembran des jeweiligen Bakteriums. Die Ruptur der Zellmembran führt zwangsläufig zum Absterben des Bak teriums, da das Zellplasma aus der aufgerissenen Zellmembran ausfließt .
Versuche mit den beschriebenen Nanostrukturen haben gezeigt, dass sich auf einem Titanprobenkörper ansiedelnde gramnegativen Escherichia coli-Bakterien nach einer zweistündigen Inkubation - durch die hier vorliegende Kontakttötung - unschädlich machen lassen . Die im Ausführungsbeispiel dargestellten Nanostrukturen lassen sich auf nahezu jeder 3D-Freiformfläche erzeugen. Der tragende Untergrund des Grundmaterials kann hierbei eine Makro- und/oder Mikrostruktur haben. Alle beschriebenen Nanostrukturen sind keine Beschichtungen eines Grundmaterials. Weitere Einzelheiten der Erfindung ergeben sich aus den Unteran sprüchen und der nachfolgenden Beschreibung mehrerer schematisch dargestellter Ausführungsformen .

Die Figuren 1 und 2 zeigen - in Form einer technischen Zeichnung bezüglich der Geometrie, nicht jedoch der Größenverhältnisse - vergrößert einen äußeren Bereich eines Implantatkörpers (10), dessen Oberfläche 1 pm2 entspricht. An dieser bioinspirierten Oberfläche versucht sich ein im Wesentlichen zylindrisches Bak terium (60) anzusiedeln. Die Oberfläche ist bedeckt mit einer bakteriziden und hydrophilen Nanostruktur (20) , die auf einem Grundkörper (11) aus Reintitan (Grade 4) oder einer Titanlegie rung Ti6Al4V ELI (Grade 23) erzeugt ist.
Die Nanostruktur (20) wird mithilfe der hydrothermischen Oxida tion erzeugt. Letztere stellt ein Verfahren dar, bei dem unter Vorhandensein einer wässrigen Lösung unter hohem Druck und hoher Temperatur eine Oxidation des Implantatkörpers (10) durchgeführt wird. Die Oxidation findet in einem dampfdicht abgeschlossenen Druckbehälter statt. Der Druckbehälter wird zusammen mit dem eingelegten Implantatkörper (10) , einer bestimmten Menge an wässriger Lösung wie z.B. Reinstwasser bzw. demineralisiertem Wasser und einer für die Oxidation erforderlichen SauerstoffZu gabe auf z.B. 220 °C erhitzt. Ggf. kann der Sauerstoffanteil im Druckkessel zusätzlich erhöht oder verringert werden. Der sich im Druckbehälter einstellende Dampfdruck geht mit einer Verschiebung des thermodynamischen Gleichgewichts der jeweiligen Kristallisationsreaktion einher. Auf der dem Dampf zugänglichen Oberfläche des Implantatkörpers (10) stellt sich ein Kristall wachstum ein. Es entsteht eine Nanotopographie aus stabilen Kristallen, die vorzugsweise nahezu senkrecht aus dem Grundwerk stoff herauswachsen, vgl. Figur 3.
Nach den Figuren 1 und 2 hat der Grundkörper (11) eine z.B.
ebene Grundfläche (12), aus der die hydrothermische Oxidation die Nanokristalle (40) herauswachsen lässt. Über dieser Grund fläche (12) hat sich dabei eine Wachstumsschicht (21) aufgebaut, aus der die Nanokristalle (40) herausragen. Zwischen den großen, als Pyramiden abgebildeten Nanokristallen (40) ist - hier idea- lisiert - eine ebene Bodenhüllfläche (23) dargestellt. Der Be reich unterhalb der Bodenhüllfläche (23) ist die Boden
schicht ( 22 ) .
Die Figuren 1 und 2 zeigen nach DIN 4760 reine geometrische Oberflächen.
Die Größe der realen Nanokristalle (40) ist eine Funktion der Verweilzeit des Titankörpers (10) im Druckbehälter. Mit zuneh mender Verweilzeit wächst die Nanostruktur in der Regel unter Vergrößerung der Nanokristalle (40) bei gleichzeitiger Abnahme ihrer Verteilungsdichte. Im Folgenden ist eine Tabelle darge stellt, die die geometrischen Abmessungen nach drei Verweilzei ten wiedergibt. Die Verweilzeiten betragen 6, 24 und 96 h.
Die Verteilung- oder Flächendichte wird in der Tabelle mit „Dichte" bezeichnet. Der „Abstand" bezeichnet die mittlere Ent fernung zweier benachbarter Nanokristalle (40), vgl. auch Fi gur 3. Die „Höhe" gibt die Gesamthöhe eines Nanokristalls (40) oberhalb der Bodenhüllfläche (23) an. Die „Basisbreite" ist die Breite der Basisfläche (41) eines Nanokristalls (40) . Der „Win- kel" gibt den Pyramidenwinkel der Nanokristalle (40) wieder. Der „Durchmesser" ist ein Maß für die Breite der Spitze eines realen Nanokristalls (40) . Sie wird beim Nanokristall (40) in einer Höhe über der Bodenhüllfläche (23) gemessen, die 85 bis 95 % der Nanokristallgesamthöhe entspricht. Der „Durchmesser" bezieht sich auf einen zur Grundfläche (12) parallelen Umkreis, den min destens zwei Kanten eines realen Nanokristalls (40) kontaktie ren. Die Prozentsatzuntergrenze gilt hierbei besonders für Nano kristalle (40) mit großem Pyramidenwinkel und kleiner Gesamt höhe .
Die Geometrieverhältnisse der Darstellungen der Figuren 1 und 2 gelten für einen Titankörper (10) aus der Titanlegierung Ti6Al4V, dessen Kristallwachstum 96 h bei einer Druckbehälter temperatur von 220 °C angedauert hat. Dies gilt auch für die Profildarstellung nach Figur 3.
In den Figuren 1 und 2 ist die Wandstärke der Wachstums
schicht (21) erkennbar. Sie ist der kürzeste Abstand zwischen der Grundfläche (12) und der Bodenhüllfläche (23) . Sie misst 0,33 ± 30 % dieses Anteils der durchschnittlichen Höhe (46) der die Bodenhüllfläche (23) überragenden Nanokristalle (40). In der Bodenhüllfläche (23) liegen die Basisflächen (41) der Nanokris talle (40) .
Nach Figur 1 liegt auf der Nanostruktur (20) - auf der Diagona len des gezeigten Ausschnitts - ein gramnegatives Bakte
rium (60). Als Beispiel wird das Bakterium Pseudomonas aerugi nosa vereinfacht dargestellt, dessen äußere Gestalt im Allgemei nen aus einer geraden zylinderförmigen oder einer schwachge krümmten torussegmentförmigen Zellwandung mit halbkugelförmigen Enden besteht. Der Durchmesser (65) des Bakteriums (60) beträgt 0,5 bis 1,0 pm, während seine Länge 1,5 bis 5 , 0 pm misst. Das Bakterium (60) hat eine Zellwand mit einer Wandstärke von ca. 2 nm. Die Zellwandstärke beträgt somit weniger als 1,2 % der mitt leren Höhe (46) der beispielhaft dargestellten Nanokris
talle (40) . In den Figuren 1 und 2 sind die Bakteriengeißeln nicht abgebildet.
Das sich am Titankörper (10) anlegende Bakterium (60) kontak tiert die Nanostruktur (20), nach Figur 1, nur im Bereich der Nanokristallspitzen (43).
Die Figur 2 zeigt das Bakterium (60) unmittelbar nach dem ersten die Zellmembran (61) schädigenden Kontakt mit der Nanostruk tur (20) . Die Zellmembran (61) hat im mittleren Bereich des Bak- teriums (60) einen ersten Riss (62), über den das Zell plasma (63) - z.B. unter einem Einknicken des Bakteriums (60) in seinem mittleren Bereich - ausläuft und sich zwischen den Nano- kristallen (40) auf der Bodenhüllfläche (23) verteilt.
Schon nach kurzer Zeit hat sich die Zellmembran (61) weitgehend entleert, so dass sie als leere Hülle auf der Nanostruktur (20) aufliegt, ohne nennenswert über die Spitzen (43) der Nanokris- talle (40) hinauszuragen.
Nur beispielhaft ist in Figur 2 ein Kontaktwinkel (71) einge zeichnet. Der Kontaktwinkel (71) bzw. Rand- oder Benetzungswin kel ist ein Maß für die Benetzbarkeit einer FeststoffOberfläche mit einer Flüssigkeit. Die Wechselwirkung zwischen der Implan- tatoberfläche und der benetzenden Flüssigkeit ist bei Nanostruk- turen (20) mit den Geometriewerten der oben wiedergegebenen Ta belle groß, so dass die auf die Nanostruktur (20) treffende Flüssigkeit, einschließlich der Bakterien (60) und deren Zell plasma (63), zwischen die Nanokristalle (40) gesaugt wird. Der Kontaktwinkel (71) beträgt im Ausführungsbeispiel weniger als 20 Winkelgrade.
Die Figur 3 zeigt einen zur ebenen Grundfläche (12) senkrechten realen Profilschnitt (35) . Er ist nach DIN 4760 eine Oberfläche, die messtechnisch mit einem Rasterkraftmikroskop erfasst wurde. Durch die mechanische Abtastung der realen Oberfläche (39), vgl. Figuren 4 und 5, mit einer nanoskopisch kleinen Nadel werden na hezu alle scharfkantigen Übergänge abgerundet wiedergegeben . Im Diagramm sind auf der Abszisse die Höhenwerte der Nanostruktur aufgetragen, während auf der Ordinate - auf der Länge eines Mik rometers - die entsprechenden Querausdehnungen abgetragen sind. Neben den Nanokristallgrößen wie Abstand (55) , Spitzendurchmes ser (47), Basisbreite (45), Pyramidenwinkel (51) und Höhe (46) ist zusätzlich eine Nanokristallmittellinie (52) mit dem ent sprechenden Neigungswinkel (53) dargestellt. Der eingezeichnete Neigungswinkel (53) beträgt hier z.B. 15 Winkelgerade. Alle an deren Geometriewerte sind der obigen Tabelle - mit Ausnahme des Abstands (55) - der dritten Inhaltszeile als Mittelwerte zu ent nehmen .
Die Figuren 4 und 5 zeigen zwei Draufsichten auf Realoberflächen der Abmessung 2,2 * 1,5 um, siehe Maßstab rechts, unten. Bei Fi gur 4 beträgt die Verweilzeit im Druckbehälter 24 h. Die dazuge hörigen Geometriewerte sind in der zweiten Inhaltszeile der obi gen Tabelle wiedergegeben . Die Figur 5 zeigt in der Draufsicht eine Nanostruktur, die sich 96 h bei einer Temperatur von 220 °C im Druckbehälter befand. Ihre Geometriewerte sind der dritten Inhaltszeile der obigen Ta belle zu entnehmen. Die Draufsichten belegen das Vorhandensein der in der Regel scharfkantigen Nanokristalle, die überwiegend senkrecht zur Grundfläche (12) gewachsen sind. Auch ist eine gleichmäßige Ver teilung der aus der Bodenhüllfläche hervorragenden Nanokristalle erkennbar, sodass die einzelnen Oberflächentopographien an den jeweiligen Verwendungszweck gezielt anpassbar sind. Bei einem Einhalten der jeweiligen vorgebbaren Prozessparameter sind die Nanostrukturen wiederholgenau reproduzierbar.

Claims

Patentansprüche :
1. Nanostruktur (20) aus einer Vielzahl von Nanokristallen auf mindestens einer Oberfläche (39) oder einem Oberflächenbereich eines Titankörpers (10),
dadurch gekennzeichnet,
- dass die Nanokristalle (40) zumindest bereichsweise eine te- tragonal-pyramidale Grundform haben,
- dass die Flächendichte der Nanokristalle (40) pro pm2 zwischen
40 und 400 liegt, wobei die Flächendichte mit zunehmender Kristallgröße abnimmt und
- dass der mittlere Abstand benachbarter Nanokristalle (40) bei einer Höhe von 23 bis 100 nm bei 50 bis 160 nm liegt.
2. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 1, dadurch gekennzeichnet, dass mindestens 60 % der Nano kristalle (40) eine viereckige, quadratische, rechteckige, rau- tenförmige, parallelogrammförmige, trapezförmige oder drachen viereckförmige Basisfläche (41) haben.
3. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An- spruch 2, dadurch gekennzeichnet, dass die Nanokristalle (40) einen Pyramidenwinkel (51) von 16 bis 46 Winkelgraden aufweisen.
4. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 2, dadurch gekennzeichnet, dass die Nanokristalle (40) eine Basisbreite (45) von 50 bis 150 nm haben.
5. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 2, dadurch gekennzeichnet, dass der Spitzenbereich der Nanokristalle (40) in einer Höhe, die 85 bis 95 % der Nanokris- tallgesamthöhe (46) entspricht, einen zur Grundfläche (12) pa rallelen Umkreis mit einem Durchmesser von 10 bis 50 nm hat, an dem mindestens zwei Nanokristallkanten (42) anliegen.
6. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 5, dadurch gekennzeichnet, dass der größere Prozentsatz der Nanokristallgesamthöhe (46) für Nanokristalle (40) mit klei nem Pyramidenwinkel (51) gilt.
7. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 2, dadurch gekennzeichnet, dass der Neigungswinkel (53) der Nanokristalle (40), der zwischen der Normalenrichtung (32) der Grundfläche (12) und der Mittellinie (52) des einzelnen Na- nokristalls (40) liegt, kleiner als 30 Winkelgrade ist.
8. Nanostruktur aus einer Vielzahl von Nanokristallen gemäß An spruch 1, dadurch gekennzeichnet, dass sich die Nanokristallflä- chendichte auf die Nanokristalle (40) bezieht, deren Gesamt höhe (46) größer ist als das 0,3-fache der maximalen Rauheits profilhöhe (48) .
9. Verfahren zur Erzeugung einer Nanostruktur (20) aus tetrago- nal-pyramidalen Nanokristallen auf mindestens einer Oberflä che (39) oder einem Oberflächenbereich eines Titankörpers (10) durch hydrothermische Oxidation, dadurch gekennzeichnet,
- dass die Ausgangsfläche (12) zur Nanostrukturerzeugung einer 160 bis 374,12 °C heißen Dampfdruckatmosphäre ausgesetzt ist,
- dass das dampfbildende Medium demineralisiertes Wasser ist und
- dass die Nanokristallflächendichte und die Nanokristallgröße sowie die Farbe der Oxidschicht der Nanostruktur (20) jeweils eine Funktion der Dampfdruckeinwirkzeit sind, wobei bei dem angegebenen Dampfdrucktemperaturbereich - für eine nutzbare Flächendichte und Größe der Nanokristalle (40) - die Einwirk zeit 1 bis 100 h beträgt.
10. Verfahren zur Herstellung einer Nanostruktur gemäß An spruch 9, dadurch gekennzeichnet, dass das Reinstwasser einen wasserstoffabsorbierenden Zusatz enthält.
