Abstract
La présente invention concerne une séquence peptidique caractérisée en ce qu'elle comprend ou est constituée par un fragment d'au moins environ 10 acides aminés issus de la séquence SEQ ID NO : 1 suivante : LRNITNSpQAPDGRRKSEAGSG SEQ ID NO : 1 dans laquelle le résidu sérine en position 7 est phosphorylé, ledit fragment susmentionné contenant ledit résidu sérine phosphorylé. La présente invention concerne également un anticorps polyclonal ou monoclonal susceptible de reconnaître une séquence peptidique telle que définie ci-dessus.
Description
NOUVELLES SEQUENCES PHOSPHORYLEES DE LA PHOSPHATASE CDC25B, ANTICORPS DIRIGÉS CONTRE CES SÉQUENCES AINSI QUE LEUR UTILISATION
La présente invention a pour objet de nouvelles séquences phosphorylées de la phosphatase CDC25B ainsi que des anticorps polyclonaux ou monoclonaux dirigés contre ces séquences. La présente invention a également pour objet l'utilisation de ces nouvelles séquences phosphorylées notamment pour la mise en œuvre d'une méthode de diagnostic in vitro de cancers chez l'homme ou l'animal. Les mécanismes qui contrôlent la division des cellules mettent en jeu de nombreux acteurs dont les activités sont régulées par des réactions de phosphorylation et de déphosphorylation, impliquant des kinases et des phosphatases. Des dérégulations de ces mécanismes ont été identifiées dans de nombreux cancers. Leur identification et leur caractérisation ouvrent aujourd'hui de nouvelles perspectives pour le diagnostic et le traitement de la maladie cancéreuse. CDC25B est une phosphatase régulatrice du cycle cellulaire essentielle pour le contrôle de l'entrée en mitose. Elle appartient à une famille qui compte trois membres codés par des gènes différents (CDC25A, B et C) chez les mammifères. La protéine CDC25B est exprimée et active en fin. de phase G2 du cycle cellulaire (Baldin et al.,
1997 ; Gabrielli et al., 1996). Sa localisation intracellulaire est régulée par des séquences NES et NLS (Davezac et al., 2000) et par son interaction avec les protéines 14-3-3 (Mils et al, 2000 ; Forrest et al, 2001). Il a été suggéré que CDC25B puisse agir comme un "starter" des événements mitotiques précoces (Nilsson et al., 2000). Elle pourrait jouer un rôle dans l'activation initiale d'une population de CDC2/cycline B au niveau du centrosome avant sa translocation nucléaire (Kumagai et al., 1992 ; Hoffmann et al., 1993). CD25B active les complexes CDK/cycline pour permettre les remaniements architecturaux et biochimiques qui sont nécessaires au processus de division cellulaire. Son activité est régalée par les variations de son expression, par son association à des partenaires régulateurs et par des événements de phosphorylation. Lors de son cycle cellulaire, la cellule reçoit de nombreux signaux notamment concernant la lésion de son ADN. Ainsi, en réponse à la présence de lésions de son ADN, la cellule stoppe son cycle cellulaire en phase Gl ou en phase G2, afin de permettre aux éléments nécessaires à la réparation de l'ADN d'entrer en action ; une
fois l'ADN réparé, la cellule prolifère de nouveau. L'inactivation de la fonction des trois phosphatases CDC25A, CDC25B et CDC25C est probablement responsable de l'arrêt en phase G2, appelé également "point de contrôle G2/M". La phosphorylation de ces phosphatases en réponse à la présence de lésions de l'ADN conduit à leur séquestration cytoplasmique et/ou à leur dégradation. Des sites phosphorylés sur
CDC25A et CDC25C en cas de lésions de l'ADN ont déjà été identifiés (Mailand et al., 2002 ; Donzetti et al., 2002 ; Peng et al., 1977). A ce jour, un seul site de phosphorylation sur CDC25B (serine 323) a été identifié à la suite de lésions de l'ADN (Bulavin et al., 2002). La présente invention découle de la mise en évidence par les Inventeurs d'un nouveau site de phosphorylation in vitro et in vivo de la phosphatase CDC25B dans le cas de lésions de l'ADN cellulaire, induites notamment par un agent genotoxique tel que la bléomycine, et de l'identification d'une nouvelle séquence phosphorylée de CDC25B sur le résidu serine en position 169. Un des buts de l'invention consiste à fournir un nouvel anticorps dirigé contre l'épitope phosphorylé de la phosphatase CDC25B phosphorylée, ledit anticorps pouvant être utilisé dans le cadre d'un procédé de détection de composés présentant un effet genotoxique sur des cellules, ou dans le cadre d'un procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie anticancéreuse. La présente invention concerne une séquence peptidique caractérisée en ce qu'elle comprend ou est constituée par un fragment d'au moins environ 10 acides aminés issus de la séquence SEQ ID NO : 1 suivante : LRNITNSpQAPDGRRKSEAGSG SEQ ID NO : 1 dans laquelle le résidu serine en position 7 est phosphorylé, ledit fragment susmentionné contenant ledit résidu serine phosphorylé. L'expression "résidu phosphorylé" désigne un acide aminé porteur d'un groupement phosphate.
Selon un mode de réalisation avantageux, la séquence peptidique de l'invention est caractérisée en ce qu'elle comprend ou est constituée par la séquence SEQ ID NO : 2 suivante : ITNSpQAPDGRRKSEA SEQ ID NO : 2 dans laquelle le résidu serine en position 4 est phosphorylé. La séquence SEQ ID NO : 2 correspond à un fragment de la séquence SEQ ID NO : 1 susmentionnée. Plus exactement, elle correspond au fragment de SEQ ID NO : 1 délimité de l'acide aminé en position 4 à l'acide aminé en position 18. La présente invention concerne également une séquence peptidique telle que définie ci-dessus, caractérisée en ce qu'elle comprend ou est constituée par la séquence
SEQ ID NO : 3, représentant un variant d'épissage CD25B3 de la protéine d'origine humaine de la phosphatase CDC25B, dont le résidu serine en position 169 est phosphorylé. La présente invention concerne également un anticorps polyclonal ou monoclonal susceptible de reconnaître une séquence peptidique telle que définie ci-dessus. Un anticorps polyclonal avantageux de l'invention est caractérisé en ce qu'il est susceptible de reconnaître la séquence SEQ ID NO : 2 telle que définie ci-dessus. Un tel anticorps dirigé contre l'épitope phosphorylé de séquence SEQ ID NO : 2 est généré en immunisant des lapins avec ledit épitope. Plus précisément, ledit épitope est couplé de façon covalente avec une protéine porteuse telle que l'hémocyanine, le BSA ou l'ovalbumine. Les lapins sont alors immunisés pendant 3 mois (4 injections au total) et la saignée finale permet la récupération d'environ 50 ml de sérum. Le sérum est ensuite doublement purifié par affinité sur une colonne de peptide phosphorylé puis sur une colonne de peptide non phosphorylé. La présente invention concerne également un procédé de préparation d'un anticorps monoclonal tel que défini ci-dessus, dirigé contre la séquence peptidique SEQ ID NO : 2 telle que définie ci-dessus, caractérisé en ce qu'il comprend les étapes suivantes : - la fusion entre des myélomes d'un animal immunisé par injection de la séquence peptidique telle que définie ci-dessus et des splénocytes d'un animal afin d'obtenir des hybridomes, - la mise en culture des hybridomes ainsi obtenus,
- la récupération et purification par clonage d'un hybridome, choisi parmi ceux obtenus à l'étape précédente et sécrétant un anticorps dirigé contre la séquence peptidique telle que définie ci-dessus. L'animal utilisé pour l'étape d'immunisation est notamment une souris. Les myélomes utilisés pour la fusion proviennent notamment d'une souris. Les splénocytes utilisés pour la fusion proviennent d'un animal de la même espèce que celle dont proviennent les myélomes, à savoir notamment une souris. On choisit les hybridomes qui sécrètent les anticorps contre la séquence peptidique SEQ ID NO : 2 sur la base de la production d'anticorps capables de reconnaître dans un test ELISA le peptide phosphorylé utilisé pour l'immunisation mais pas le peptide non phosphorylé. La présente invention concerne également un procédé de préparation d'un anticorps monoclonal tel que défini ci-dessus, notamment dirigé contre la séquence peptidique SEQ ID NO : 2 telle que définie ci-dessus, caractérisé en ce qu'il comprend une étape de sélection dans une banque d'expression d'un ADNc codant pour toute ou partie d'un anticorps, ledit anticorps étant sélectionné sur la base de sa capacité à reconnaître dans un test ELISA, par transfert de protéines ou par toute autre méthode appropriée le peptide phosphorylé tel que défini ci-dessus, utilisé pour l'immunisation, mais pas le peptide non phosphorylé. La présente invention concerne également une composition pharmaceutique caractérisée en ce qu'elle contient à titre de substance active une séquence peptidique telle que définie ci-dessus ou un anticorps tel que défini ci-dessus, en association avec un vecteur pharmaceutiquement acceptable. Une composition pharmaceutique avantageuse selon l'invention est caractérisée en ce qu'elle contient à titre de substance active la séquence peptidique représentée par la séquence SEQ ID NO : 2. La présente invention concerne également l'utilisation de la séquence peptidique telle que définie ci-dessus ou d'un anticorps tel que défini ci-dessus, pour la préparation d'un médicament destiné au traitement de maladies yperprolifératives telles que les cancers. La séquence peptidique phosphorylée selon l'invention ou une molécule de synthèse mimant sa structure est utilisée pour bloquer par compétition l'activité de l'enzyme qui en est responsable et par conséquent empêcher la phosphorylation de CDC25B sur ce site. L'anticorps tel que défini ci-dessus dirigé contre la protéine de
l'invention ou une molécule de synthèse liant le site phosphorylé peut être utilisé pour masquer le site phosphorylé et perturber sa fonction dans l'inhibition de l'activité de CDC25B. La présente invention concerne également l'utilisation d'un anticorps tel que défini ci-dessus, pour la mise en œuvre d'un procédé de détection de composés présentant un effet genotoxique sur des cellules. Selon un mode de réalisation avantageux, la présente invention concerne l'utilisation d'un anticorps polyclonal tel que défini ci-dessus, dirigé contre l'épitope phosphorylé de séquence SEQ ID NO : 2, pour la mise en œuvre d'un procédé de détection de composés présentant un effet genotoxique sur des cellules. La présente invention concerne l'utilisation d'un anticorps tel que défini ci- dessus, pour la mise en œuvre d'un procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie cancéreuse. Selon un mode de réalisation avantageux, la présente invention concerne l'utilisation d'un anticorps polyclonal tel que défini ci-dessus, dirigé contre l'épitope phosphorylé de séquence SEQ ID NO : 2, pour la mise en œuvre d'un procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie anticancéreuse. La présente invention concerne un procédé de détection de composés présentant un effet genotoxique sur des cellules, caractérisé en ce qu'il comprend : - la mise en présence dudit composé avec un anticorps tel que défini ci-dessus, et - la détection de la liaison dudit composé avec ledit anticorps par une méthode appropriée, notamment par transfert de protéines (Western blot), par ELISA ou par immunofluorescence indirecte. Pour ce faire, des cellules en culture sont exposées à un agent ou à un mélange supposé présenter une activité genotoxique. Les cellules sont alors par exemple fixées et perméabilisées, puis incubées en présence de l'anticorps contre la forme phosphorylée de CDC25B. La détection est par exemple effectuée avec un anticorps secondaire couplé à un agent fluorochrome détectable par immunofluorescence indirecte. De façon similaire, les cellules exposées peuvent être lysées. L'extrait cellulaire est alors soumis par exemple à une électrophorèse monodimensionnelle puis à un transfert des protéines sur membrane pour la réalisation d'une expérience de western
blot utilisant l'anticorps contre la forme phosphorylée de CDC25B puis un anticorps secondaire couplé à un système de révélation. La présente invention concerne également un procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie cancéreuse, caractérisé en ce qu'il comprend : - la mise en présence dudit composé avec des cellules, et - la sélection des composés inhibiteurs par détection de l'absence de liaison de l'anticorps tel que défini ci-dessus, dirigé contre la forme phosphorylée de CDC25B, par une méthode appropriée, notamment en utilisant les techniques ELISA, de transfert de protéines (Western Blot) ou d'immunofluorescence indirecte, ladite absence de liaison correspondant à l'absence de la phosphorylation de la serine en position 169 de la séquence protéique de l'invention SEQ ID NO : 3. Pour ce faire, les cellules sont mises en culture en présence de collections de molécules (criblage à haut débit) et d'un agent connu pour activer la phosphorylation de
CDC25B sur la serine en position 169 (bléomycine). Les cellules peuvent par exemple être lysées puis les extraits soumis à une détection par ELISA de la forme phosphorylée de CDC25B. L'immunofluorescence peut également être utilisée pour détecter la négativité de reconnaissance par l'anticorps, ce qui correspond à une absence de phosphorylation, donc d'inhibition de la réponse.
DESCRIPTION DES FIGURES Les figures 1A et 1B représentent les résultats d'expériences de détection de la forme phosphorylée de CDC25B sur le résidu serine en position 169 par immunofluorescence. La figure 1A correspond aux cellules non traitées et la figure 1B aux cellules traitées par la bléomycine pendant 30 minutes. Les figures 2A et 2B représentent les résultats d'expériences de détection de la forme phosphorylée de CDC25B sur le résidu serine en position 169 par western blot. La figure 2A correspond aux cellules non traitées et la figure 2B aux cellules traitées par la bléomycine pendant 30 minutes.
METHODES ET RESULTATS
La phosphatase CDC25B est phosphorylée sur la serine 169 L'analyse par spectrométrie de masse réalisée sur CDC25B a permis de détecter la phosphorylation du résidu serine en position 169. La protéine CDC25B a été purifiée puis digérée par la trypsine. L'analyse par spectrométrie de masse MS/MS a révélé la présence d'un peptide monophosphorylé. La fragmentation de ce peptide a permis l'identification du groupement phosphate sur la serine 169.
Production d'anticorps contre la protéine CDC25B phosphorylée sur l'acide aminé serine 169 Le peptide de séquence ITNS(p)QAPDGRRKSEA (SEQ ID NO : 2), où (p) désigne la serine phosphorylée, a été utilisé pour l'immunisation de lapins. Après sacrifice des animaux, le sérum a été purifié par chromatographie en deux étapes : la première sur une colonne de peptide phosphorylé pour retenir les anticorps spécifiques, puis la deuxième sur une colonne du même peptide non phosphorylé de séquence ITNSQAPDGRRKSEA, de manière à purifier dans l'éluat les anticorps spécifiques de la forme phosphorylée. La reconnaissance du peptide phosphorylé par les anticorps a été validée dans un test ELISA. Par la suite, ces anticorps sont désignés sous le nom de anti-S169P.
Les anticorps anti-S169P reconnaissent la protéine CDC25B dans des cellules traitées par des agents génotoxiques. Des cellules HeLa ont été traitées par des rayons UN ou par des agents génotoxiques comme l'étoposide ou la bléomycine. A) Elles ont été fixées et utilisées pour effectuer une analyse par immunofluorescence avec ces anticorps. Les cellules ont également été colorées avec le 4'-6 diamino-2-phénylindole (DAPI) pour localiser le noyau. Les images présentées aux figures 1A et 1B (expériences avec la bléomycine) sont représentatives d'observations sur un grand nombre de cellules. Ceci indique que la protéine CDC25B phosphorylée sur la serine 169 (SEQ ID ΝO : 2) est accumulée de façon très abondante dans les cellules traitées. Ce marquage est aboli lorsqu'une compétition est effectuée avec le
peptide phosphorylé ayant servi à l'immunisation mais pas avec le peptide non phosphorylé (ITNSQAPDGRRKSEA) ni avec un peptide phosphorylé sans rapport. B) Des extraits cellulaires ont été préparés et analysés par transfert de protéines (western blot) avec les anticorps contre la serine 169 phosphorylée. Comme le montre les figures 2A et 2B, la protéine phosphorylée SEQ ID NO : 2 est détectée de façon très abondante après traitement par la bléomycine (figure 2B) alors qu'elle est absente ou très peu abondante sans traitement (figure 2A). Ces observations valident l'utilisation de ces anticorps dans des approches expérimentales basées sur les techniques d'immunofluorescence et de western blot.
DOMAINES D'APPLICATION DE CES RÉACTIFS A - Marqueur d'efficacité de candidats médicament : La réponse normale d'une cellule, lorsqu'elle détecte des lésions de son ADN induites par les traitements comme la radiothérapie est de stopper sa prolifération pour les réparer, puis de proliférer de nouveau. L'objectif de nouvelles stratégies thérapeutiques du traitement des cancers, actuellement en cours de validation, vise à empêcher les cellules cancéreuses de détecter et/ou de répondre "correctement" à la présence de .lésions (c'est la notion de point de contrôle ou checkpoint). Tout se passe en quelque sorte comme si on cherchait à empêcher la cellule de prendre en compte ses altérations pour qu'elle continue à proliférer et la faire entrer dans une situation catastrophique la forçant ensuite à mourir. Il est donc nécessaire de visualiser la capacité cellulaire à détecter les lésions et à les prendre en compte pour arrêter la prolifération. La phosphorylation de la serine 169 de CDC25B est un des marqueurs de cette réponse aux dommages de l'ADN. La détection de la phosphorylation de CDC25B en réponse à des lésions de l'ADN permet de tester in vitro sur des protéines purifiées, sur des cellules en culture ou sur des tumeurs expérimentales, l'efficacité de molécules candidats médicaments inhibiteurs de la transmission de l'information "lésion de l'ADN" à la machinerie du cycle cellulaire et du contrôle de la prolifération.
Les cellules tumorales en culture (ou un animal modèle porteur de tumeurs expérimentales) sont exposées à un agent genotoxique utilisé en chimiothérapie ou à une radiothérapie, et simultanément exposées à un candidat médicament testé pour sa capacité à bloquer la détection correcte des dommages sur l'ADN. Les cellules ou les
coupes de tissus sont fixées et perméabilisées, puis incubées en présence de l'anticorps contre la forme phosphorylée de CDC25B. La détection est réalisée avec un anticorps secondaire couplé à un agent fluorochrome détectable par immunofluorescence indirecte. L'absence de détection de la forme phosphorylée de CDC25B témoigne de l'efficacité du candidat médicament.
B — Mise en place de crible à haut débit : Dans la perspective d'identifier de nouvelles molécules répondant à l'objectif thérapeutique présenté ci-dessus, des tests de criblages basés sur l'inhibition d'activité enzymatique in vitro ont été développés. Par ailleurs, la présente invention permet de mettre en œuvre des tests fonctionnels, basés sur des systèmes cellulaires ou acellulaires et dans laquelle l'efficacité de molécules interférant avec le point de contrôle peut être appréciée en utilisant les anticorps contre la serine 169 phosphorylée.
C - Aide à la décision thérapeutique : Différentes approches thérapeutiques actuellement disponibles ou en cours de validation ne sont pertinentes que si la cellule tumorale possède (ou ne possède pas) une voie totalement opérationnelle de signalisation capable de prendre en compte la présence de lésions de l'ADN et de bloquer la prolifération. En effet, la prise en compte de la capacité des cellules tumorales à répondre correctement à l'exposition à des agents physiques ou chimiques lésant l'ADN peut avoir une influence majeure dans le choix de la décision thérapeutique. La détection de la phosphorylation de CDC25B sur S 169 a un intérêt dans cette perspective. Les cellules tumorales sont prélevées (lors d'une biopsie ou de l'exérèse chirurgicale) et soumises in vitro à un traitement avec un agent genotoxique ou à une radiothérapie. La détection de la phosphorylation de CDC25B est ensuite réalisée par immunofluorescence indirecte après fixation et en utilisant les anticorps spécifiques. L'absence de réponse positive (phosphorylation) est un facteur prédictif d'efficacité de la chimiothérapie ou de la radiothérapie.
D - Détection de l'effet de génotoxiques : La recherche en génotoxicologie a pour objectif de détecter et de quantifier les lésions de l'ADN induites par des agents génotoxiques. Cette recherche concerne la recherche pharmacologique, les biotechnologies, la chimie, la cosmétique, l'environnement, etc.. Peu de tests in vivo sont disponibles à ce jour pour détecter la présence de lésions de l'ADN et aucune méthode simple n'a encore été proposée pour détecter l'activation de la cascade de signalisation qui témoigne de la présence de ces dommages et de leur impact sur la prolifération. L'anticorps de l'invention dirigé contre la serine en position 169 phosphorylée de
CDC25B peut être utilisé pour la détection de l'effet de génotoxiques sur des systèmes cellulaire eucaryotes.

Claims
1. Séquence peptidique caractérisée en ce qu'elle comprend ou est constituée par un fragment d'au moins environ 10 acides aminés issus de la séquence SEQ ID NO : 1 suivante : LRNITNSpQAPDGRRKSEAGSG SEQ ID NO : 1 dans laquelle le résidu serine en position 7 est phosphorylé, ledit fragment susmentionné contenant ledit résidu serine phosphorylé.
2. Séquence peptidique selon la revendication 1, caractérisée en ce qu'elle comprend ou est constituée par la séquence SEQ ID NO : 2 suivante : ITNSpQAPDGRRKSEA SEQ ID NO : 2 dans laquelle le résidu serine en position 4 est phosphorylé.
3. Séquence peptidique selon la revendication 1 ou 2, caractérisée en ce qu'elle comprend ou est constituée par la séquence SEQ ID NO : 3, représentant un variant d'epissage CD25B3 de la protéine d'origine humaine de la phosphatase CDC25B, dont le résidu serine en position 169 est phosphorylé.
4. Anticorps polyclonal ou monoclonal susceptible de reconnaître une séquence peptidique selon l'une quelconque des revendications 1 à 3.
5. Anticorps polyclonal susceptible de reconnaître la séquence SEQ ID NO : 2 telle que définie dans la revendication 2.

Procédé de préparation d'un anticorps monoclonal selon la revendication 4, dirigé contre la séquence peptidique SEQ ID NO : 2 telle que définie dans la revendication 2, caractérisé en ce qu'il comprend les étapes suivantes : - la fusion entre des myélomes d'un animal immunisé par injection de la séquence peptidique selon la revendication 2 et des splénocytes d'un animal afin d'obtenir des hybridomes, - la mise en culture des hybridomes ainsi obtenus, - la récupération et purification par clonage d'un hybridome, choisi parmi ceux obtenus à l'étape précédente et sécrétant un anticorps dirigé contre la séquence peptidique selon la revendication 2.
7. Procédé de préparation d'un anticorps monoclonal selon la revendication 4, notamment dirigé contre la séquence peptidique SEQ ID NO : 2 telle que définie dans la revendication 2, caractérisé en ce qu'il comprend une étape de sélection dans une banque d'expression d'un ADNc codant pour toute ou partie d'un anticorps, ledit anticorps étant sélectionné sur la base de sa capacité à reconnaître dans un test ELISA, par transfert de protéines ou par toute autre méthode appropriée le peptide phosphorylé selon l'une quelconque des revendications 1 à 3, utilisé pour l'immunisation, mais pas le peptide non phosphorylé.
8. Composition pharmaceutique caractérisée en ce qu'elle contient à titre de substance active une séquence peptidique selon l'une quelconque des revendications 1 à 3, un anticorps selon la revendication 4 ou 5, en association avec un vecteur pharmaceutiquement acceptable.
9. Utilisation de la séquence peptidique selon l'une des revendications 1 à 3, ou d'un anticorps selon la revendication 4 ou 5, pour la préparation d'un médicament destiné au traitement de maladies hyperprolifératives telles que les cancers.
10. Utilisation d'un anticorps selon la revendication 4 ou 5, pour la mise en œuvre d'un procédé de détection in vitro de composés présentant un effet genotoxique sur des cellules.
11. Utilisation d'un anticorps selon la revendication 4 ou 5, pour la mise en œuvre d'un procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie anticancéreuse.
12. Procédé de détection in vitro de composés présentant un effet genotoxique sur des cellules, caractérisé en ce qu'il comprend : - la mise en présence dudit composé avec un anticorps selon la revendication 4 ou 5, et - la détection de la liaison dudit composé avec ledit anticorps par une méthode appropriée.
13. Procédé de criblage in vitro de composés inhibiteurs de la réponse cellulaire aux lésions de son ADN, lesdits composés inhibiteurs pouvant être notamment utilisés dans le cadre d'une thérapie anticancéreuse, caractérisé en ce qu'il comprend : - la mise en présence dudit composé avec des cellules, et - la sélection des composés inhibiteurs par détection de l'absence de liaison de l'anticorps selon la revendication 4 ou 5, ledit anticorps étant susceptible de reconnaître une séquence peptidique selon l'une quelconque des revendications 1 à 3, par une méthode appropriée, notamment en utilisant les techniques ELISA, de transfert de protéines (Western Blot) ou d'immunofluorescence indirecte.
